Fragmentos da junção uterovaginal de 16 exemplares de fêmeas adultas de peru foram examinados, com o objetivo de determinar as modificações morfométricas e histológicas dos túbulos armazenadores de espermatozoides (SST), durante os períodos de atividade e repouso reprodutivo. Os estudos foram realizados a partir de dissecações do oviduto esquerdo, utilizando-se técnicas histológicas, histoquímica e histométricas de rotina. Essas estruturas originam-se de invaginações do epitélio das pregas da região uterovaginal e apresentam-se tubulosas, simples e não ramificadas, com epitélio formado por células prismáticas estreitas, claras e escuras, com núcleo vesiculoso basal e microvilos apicais. As secreções dessas células apresentaram reações negativas para PAS e Alcian Blue, indicando ausência de mucossubstâncias. Os parâmetros morfométricos utilizados mostraram que há diferenças significativas entre os períodos de atividade reprodutiva e de repouso. Na fase de atividade reprodutiva, o comprimento tubular, o volume tubular, o volume do lume, o número de SST na junção uterovaginal, o diâmetro externo e interno, a altura celular e o número de células por túbulos estão aumentados, e observam-se espermatozoides agrupados na luz do lume e células claras e escuras nos túbulos com vesículas supranucleares no citoplasma das células tubulares. Na fase de repouso, os parâmetros morfológicos estão reduzidos e não são encontrados espermatozoides na luz do lume. Nesta fase, não são evidenciadas as células escuras nem as vesículas supranucleares no citoplasma celular.

The aim of the present study was to investigate the thoracic part of the esophagus in the bronze turkey, using Masson’s trichrome stain and Alcian blue-PAS staining. Thirty-six clinically healthy bronze turkeys (eighteen males and eighteen females) were studied. The groups of the birds were at age 1, 7, 14, 28, 35 and 49 days. Each group consisted of three male and three female birds. The histological features of the organ were similar in the studied groups. It consisted of tunica mucosa, tunica submucosa, tunica muscularis, and tunica serosa. There were no specifics in the organ regarding the sex and the age of the birds. The esophageal glands in all of the studied groups, demonstrated intensive PAS and AB reaction, because of the mucous, produced by the epithelial glandular cells. Lamina propria exhibited a weak PAS reaction, visible in all ages, either in males, either in females.

     The study includes six adult healthy hen turkey. The cloaca has three compartments separated by two circular folds and next to rectum is the coprodeum which the largest and measured (2.5±0.2cm) in length and (2.1±0.1cm) in diameter. The middle and smallest is the urodeum that measured about (1.01±0.2cm) in length and (1.2±1cm.) in diameter, that receives the ureter and left oviduct. Posteriorly, the proctodeum that measured about (1.4±0.1cm) in length and (1.6±1cm.) in diameter. The coprodeum mucosa has short and flat villi. The urodeum mucosa has tall branching pyramidal mucosal villi. The mucosa of proctodeum has lost the villi pattern. The three compartments were lined with mucous secreting columnar cells, most of these epithelial cells were positive for Alcian blue (2.5pH) stain and few were positive for PAS stain. The epithelial tubular glands showed positive reaction for Alcian blue satin only. The lamina propria of these compartments was cellular loose connective tissue displayed numerous of lymphatic nodules. The muscular mucosa is lost in proctodeum only. Tunica muscular is composed of two layers of smooth muscles fibers in coprodeum and urodeum and turned into skeletal type in proctodeum. The mean height of mucosal villi has significantly increased in urodeum.

Developing blood vessels in the rat central nervous system exhibit several unusual luminal features. Hannah (1975) used high voltage electron microscopy to demonstrate numerous ridges of endothelium, some near junctional complexes. The ridges produced troughs (which may appear as depressions) in the endothelial surface. In some areas ridges extended over the troughs, removing them from direct contact with the luminal surface. At no time were the troughs observed to penetrate the basal laminae. Fingerlike projections also extended into the lumina.To determine whether any chemical specializations accompanied the unusual morphological features of the luminal surface, we added 0.1% Alcian blue (Behnke and Zelander, 1970) to the 3% glutaraldehyde perfusate (cacodylate buffer, pH 7.4). After Alcian blue had reacted with the luminal glycocalyces, the dye was dissociated with MgCl2 via critical electrolyte concentration method of Scott and Dorling (1965). When these methods are applied together, it is possible to differentiate mucopolysaccharides (glycosaminoglycans or GAG) with the electron microscope.

Aims: To report on the clinico-pathological features of a series of 14 intra-oral mucoepidermoid carcinomas showing exclusive intra-cystic growth. Materials and methods: All mucoepidermoid carcinomas diagnosed in the period 1990–2012 were retrieved; the original histological preparations were reviewed to confirm the diagnosis and from selected cases, showing exclusive intra-cystic neoplastic components, additional sections were cut at three subsequent 200 m intervals and stained with Hematoxylin–Eosin, PAS, Mucicarmine and Alcian Blue, to possibly identify tumor invasion of the adjacent tissues, which could have been overlooked in the original histological preparations. Additionally, pertinent findings collected from the clinical charts and follow-up data were analyzed. Results: We identified 14 intraoral mucoepidermoid carcinomas treated by conservative surgery and with a minimum follow up of five years. The neoplasms were located in the hard palate (nine cases), the soft palate (two), the cheek (two) and the retromolar trigone (one). In all instances, histological examination revealed the presence of a single cystic space, containing clusters of columnar, intermediate, epidermoid, clear and mucous-producing cells, the latter exhibiting distinct intra-cytoplasmic mucin production, as confirmed by PAS, Mucicarmine and Alcian Blue stains. The cysts were entirely circumscribed by fibrous connective tissue, and no solid areas or infiltrating tumor cell clusters were detected. Conservative surgical resection was performed in all cases, and no recurrences or nodal metastases were observed during follow up. Conclusions: Mucoepidermoid carcinomas showing prominent (>20%) intra-cystic proliferation currently are considered low-grade tumors. In addition, we also unveil the possibility that mucoepidermoid carcinomas, at least in their early growth phase, may display an exclusive intra-cystic component and might be considered as in situ carcinomas, unable to infiltrate adjacent tissues and metastasize.

Genome characterization and analysis is an imperative step in identifying and selectively breeding for improved traits of agriculturally important species. Expressed sequence tags (ESTs) represent a transcribed portion of the genome and are an effective way to identify genes within a species. Downstream applications of EST projects include DNA microarray construction and interspecies comparisons. In this study, 694 ESTs were sequenced and analyzed from a library derived from a 24-day-old turkey embryo. The 437 unique sequences identified were divided into 76 assembled contigs and 361 singletons. The majority of significant comparative matches occurred between the turkey sequences and sequences reported from the chicken. Whole genome sequence from the chicken was used to identify potential exon–intron boundaries for selected turkey clones and intron-amplifying primers were developed for sequence analysis and single nucleotide polymorphism (SNP) discovery. Identified SNPs were genotyped for linkage analysis on two turkey reference populations. This study significantly increases the number of EST sequences available for the turkey.Key words: turkey, cDNA, expressed sequence tag, single nucleotide polymorphism.

Objective: To investigate a better method of inducing hUC-MSCs into chondrocytes in different culture system in vitro. Method: hUC-MSCs were isolated and cultured by tissue block culture, and the cells surface antigens were identified by flow cytometry, hUC-MSCs were cultured with chondrogenic media and stained with Alcian Blue. The production of matrix was estimated from the determination of hydroxyproline content and Alcian Blue method. Expressions of glycosaminoglycan (GAG), type II collagen and Sox-9 were assayed by real-time fluorescence quantitative PCR. Results: The cultured hUC-MSCs phenotype was CD105+/CD29+/CD44+/ CD31-/CD34-/ CD40-/CD45-/HLA-DR-. hUC-MSCs weakly expressed chondrocyte marker, which strongly expressed GAG and type II collagen after chondrogenic induction, and the cells were incubated in pellet culture with higher expression. Real-time PCR results demonstrated that chondrogenic induction cells were expressed GAG, type II collagen and Sox-9, and the cells were incubated in pellet culture with higher expression. Conclusion: hUC-MSCs incubated in pellet culture is more conducive to differentiate into chondrocytes than those cultured in monolayer culture system.
