This short review deals with the properties and significance of the determination of selenium, which is in trace amounts an essential element for animals and humans, but toxic at high concentrations. It may cause oxidative stress in cells, which leads to the chronic disease called selenosis. Several analytical techniques have been developed for its detection, but electroanalytical methods are advantageous due to simple sample preparation, speed of analysis and high sensitivity of measurements, especially in the case of stripping voltammetry very low detection limits even in picomoles per liter can be reached. A variety of working electrodes based on mercury, carbon, silver, platinum and gold materials were applied to the analysis of selenium in various samples. Only selenium in oxidation state + IV is electroactive therefore the most of voltammetric determinations are devoted to it. However, it is possible to detect also other forms of selenium by indirect electrochemistry approach.

Niemann-Pick type C2 (NP-C2) disease is a rare hereditary disease caused by mutations in the NPC2 gene. NPC2 is a small, soluble protein consisting of 151 amino acids, primarily expressed in late endosomes and lysosomes (LE/LY). Together with NPC1, a transmembrane protein found in these organelles, NPC2 accomplishes the exclusion of cholesterol; thus, both proteins are essential to maintain cellular cholesterol homeostasis. Consequently, mutations in the NPC2 or NPC1 gene result in pathophysiological accumulation of cholesterol and sphingolipids in LE/LY. The vast majority of Niemann-Pick type C disease patients, 95%, suffer from a mutation of NPC1, and only 5% display a mutation of NPC2. The biochemical phenotype of NP-C1 and NP-C2 appears to be indistinguishable, and both diseases share several commonalities in the clinical manifestation. Studies of the pathological mechanisms underlying NP-C2 are mostly based on NP-C2 animal models and NP-C2 patient-derived fibroblasts. Recently, we established induced pluripotent stem cells (iPSCs), derived from a donor carrying the NPC2 mutations c.58G>T/c.140G>T. Here, we present a profile of pathophysiological in vitro features, shared by NP-C1 and NP-C2, of neural differentiated cells obtained from the patient specific iPSCs. Profiling comprised a determination of the NPC2 protein level, detection of cholesterol accumulation by filipin staining, analysis of oxidative stress, and determination of autophagy. As expected, the NPC2-deficient cells displayed a significantly reduced amount of NPC2 protein, and, accordingly, we observed a significantly increased amount of cholesterol. Most notably, NPC2-deficient cells displayed only a slight increase of reactive oxygen species (ROS), suggesting that they do not suffer from oxidative stress and express catalase at a high level. As a site note, comparable NPC1-deficient cells suffer from a lack of catalase and display an increased level of ROS. In summary, this cell line provides a valuable tool to gain deeper understanding, not only of the pathogenic mechanism of NP-C2, but also of NP-C1.

AbstractOxidative stress, associated with an imbalance between the oxidants (reactive oxygen species) and the antioxidants in the body, contributes to the development of many diseases. The body’s fight against reactive oxygen species is supported by antioxidants. Nowadays, there are too many analytical methods, but there is no one universal technique for assessing antioxidant properties. Moreover, the applied different ways of expressing the results lead to their incompatibility and unreasonable interpretation. The paper is a literature review concerning the most frequent ways of antioxidant activities expression and for an easy and universal method of the obtained results discussion. This paper is an attempt to point out their disadvantages and advantages. The manuscript can support the searching interpretation of the obtained results which will be a good tool for the development of a number of fields, especially medicine what can help in the future detection and treatment of many serious diseases. Berberis lyceum Royle has such pronounced medicinal values that it is used as cure of many diseases and has exhibited great therapeutic effects among the local communities throughout the world. The present research was carried out to evaluate the quantitative ethnobotanical status and phytochemical analysis of B. lyceum. Regular field trips were arranged to the study area (Shangla District) in August 2017 to October 2019 and interviews with 100 residents (age range: 30 to 50 years) were conducted. The approach adopted for ethnobotanical data was semi-scientific as the inhabitants were not aware about the modern names of some diseases and therefore physician prescriptions were also consulted. Ethnobotanical data were examined using relative frequency of citation and % use value. The % use value of B. lyceum indicates that the people of District Shangla mostly used it for curing of different diseases. In spite of tremendous uses the plant still survived in this area mainly due to the non-accessibility for humans of the mountain tops; otherwise, increase in anthropogenic activities even in these hilly areas poses a threat of the eradication of this plant. To correlate the folkloric uses with its phytochemical composition, HPLC (high performance liquid chromatography) analysis was performed and a total of six phenolic compounds (quercetin, chlorogenic acid, berberine, rutin, mandellic acid, and hydroxy benzoic acid) were identified in its root. As most of the health complications are correlated to oxidative stress therefore in vitro antioxidant activity were also performed using DPPH (2, 2- diphenyl-1-picrylhydrazyl) and ABTS (2, 2′-azinobis-3-ethyl benzo thiazoline -6- sulfonic acid) assays. The observed antioxidant potential may most probably be due to berberine and chlorogenic acid that were present in highest concentration in the analyzed extract. The effectiveness of the selected plant as remedy for a number of diseases (that were pointed out by the local community) may be due to its phytochemical composition especially berberine and chlorogenic acid as oxidative stress is the root cause of many diseases. The plant extract exhibited high antioxidant potential (DPPH IC50 = 165µg/mL; ABTSIC50 = 110µg/mL) in relation to the detected concentration of berberine and chlorogenic acid. It can be inferred from experimental results that the ethnopharmacological efficacy of this endangered species may be due to its phytochemical composition and antioxidant activities. This case study helps to revive the importance of B. lyceum in local communities and emphasizes the need for its conservation.
