FACTORS INFLUENCING THE UPTAKE OF [α-14C]AMINOISOBUTYRIC ACID BY RAT TESTES

SUMMARY The action of various agents on the distribution ratio of [α-14C]aminoisobutyric acid ([14C]AIB) in the testes of adult and growing rats was studied. A decrease in the distribution ratio of [14C]AIB was observed in the testes during maturation. Follicle-stimulating hormone (FSH) administered in vivo to 22-day-old rats or to mature hypophysectomized animals increased the distribution ratio of [14C]AIB in testicular cells. Dibutyryl adenosine 3′: 5′-cyclic phosphate augmented the [14C]AIB uptake into testes of intact or hypophysectomized mature animals. This nucleotide was ineffective in rats under 32 days of age. Testes from mature and immature rats preincubated in vitro with cycloheximide for 1 h showed a decrease in the distribution ratio of [14C]AIB. Cycloheximide also blocked the action of FSH on [14C]AIB uptake in the testes of 22-day-old rats.

Biochemical properties of isolated Leydig cells, Sertoli cells and spermatocytes from rat testes have been investigated after in-vivo or in-vitro exposure of these cells to abdominal temperature (37 °C). The rate of production of testosterone and pregnenolone by isolated Leydig cells from cryptorchid and normal testes from mature rats was not different. Production of pregnenolone by mitochondria prepared from cryptorchid testes was 6·7 times higher than production by mitochondria from normal testes. Sertoli cells prepared from immature rats and incubated in vitro at 32 or 37 °C showed, on day 1 of the culture period, an initial twofold increase in the secretion of androgen-binding protein which was absent after 6 days in culture. In contrast, incorporation of [3H]leucine into secreted proteins was stimulated twofold on day 1 as well as by day 6 of culture. Secretion of oestradiol was increased 30-fold by day 6 when compared with the level found on day 1 when cells had been cultured at 37 °C and the increased secretion of oestradiol was maintained for approximately 2 days when the temperature of incubation was decreased to 32 °C Spermatocytes isolated from seminiferous tubules incubated for 20 h at 37 °C were active in the synthesis of RNA. No degeneration of these cells was observed in testes of 25-day-old rats 5 days after experimental cryptorchidism, whereas under similar conditions massive degeneration of spermatocytes was shown in the testes of mature rats. These results suggest that the effects of temperature on the different testicular cells greatly depend on the experimental conditions used to study the effect of temperature.

Abstract We have investigated the role of Ca2+ in the control of FSH-induced estradiol secretion by Sertoli cells isolated from 8-10 days old rats. Exogenous Ca2+ (4-8 mmol/1) inhibited FSH-stimulated E2 secretion such that, with 8 mmol/l Ca2+ and FSH (8 IU/l) E2 secretion decreased from 2091±322 to 1480±84 pmol/l (p<0.002), whilst chelation of Ca2+ in the culture medium with EGTA (3 mmol/l) increased E2 secretion from 360±45 to 1242±133 pmol/l) in the absence of FSH. Further, EGTA (3 mmol/l) markedly potentiated FSH (8 IU/l), forskolin (1 μmol/l) and dibutyryl cAMP (1 mmol/l)-stimulated E2 secretion. Addition of the Ca2+ ionophores, ionomycin (2-5 μmol/l) and A23187 (2 μmol/l), inhibited FSH (8 IU/l)-stimulated E2 secretion by >80%. The effect of ionomycin was totally reversible, whereas that of A23187 was irreversible. Ionomycin (5 μmol/l) had no effect on EGTA-induced E2 secretion in the absence of FSH, but reduced EGTA-provoked E2 secretion by 59% in the presence of FSH (8 IU/l). Similarly, forskolin- and dibutyryl cAMP-provoked E2 production was inhibited 46-50% by ionomycin (5 μmol/l). We conclude that FSH-induced E2 secretion from immature rat Sertoli cells is modulated by intra- and extracellular Ca2+.

We have been studying the differing characteristics of oligodendrocyte-type-2 astrocyte (O-2A) progenitors isolated from optic nerves of perinatal and adult rats. These two cell types display striking differences in their in vitro phenotypes. In addition, the O-2Aperinatal progenitor population appears to have a limited life-span in vivo, while O-2Aadult progenitors appear to be maintained throughout life. O-2Aperinatal progenitors seem to have largely disappeared from the optic nerve by 1 mo after birth, and are not detectable in cultures derived from optic nerves of adult rats. In contrast, O-2Aadult progenitors can first be isolated from optic nerves of 7-d-old rats and are still present in optic nerves of 1-yr-old rats. These observations raise two questions: (a) From what source do O-2Aadult progenitors originate; and (b) how is the O-2Aadult progenitor population maintained in the nerve throughout life? We now provide in vitro evidence indicating that O-2Aadult progenitors are derived directly from a subpopulation of O-2Aperinatal progenitors. We also provide evidence indicating that O-2Aadult progenitors are capable of prolonged self renewal in vitro. In addition, our data suggests that the in vitro generation of oligodendrocytes from O-2Aadult progenitors occurs primarily through asymmetric division and differentiation, in contrast with the self-extinguishing pattern of symmetric division and differentiation displayed by O-2Aperinatal progenitors in vitro. We suggest that O-2Aadult progenitors express at least some properties of stem cells and thus may be able to support the generation of both differentiated progeny cells as well as their own continued replenishment throughout adult life.

To study the effects of bicarbonate and magnesium on bone, mild acidosis and/or hypermagnesemia were produced in growing rats by feeding ammonium chloride and/or magnesium sulfate. Bone composition, quantitative histomorphometry, and mineral x-ray diffraction (XRD) characteristics were measured after 6 wk of treatment. The results demonstrated that both acidosis (decreased HCO3) and hypermagnesemia inhibited periosteal bone formation, and, when combined, results were summative; and the previously observed in vitro role of HCO3- and Mg2+ as inhibitors of crystal growth were confirmed in vivo. XRD measurements demonstrated that decreased plasma HCO3 resulted in larger crystals and increased Mg resulted in smaller crystals. However, the combined XRD effects of acidosis and hypermagnesemia resembled acidosis alone. It is postulated that the final composition and crystal structure of bone are strongly influenced by HCO3- and Mg2+, and the effects are mediated by the combined influence on both osteoblastic bone formation and the growth of hydroxyapatite.

Актуальность. Моделирование in vitro травматического повреждения мозга помогает выяснить патологические механизмы, ответственные за гибель клеток или их последующую дисфункцию в деталях, труднодостижимых in vivo. Цель. Определить изменения внутриклеточной концентрации свободного Са2+ ([Ca2+]i) и митохондриального потенциала (m) в первичной нейроглиальной культуре непосредственно в момент нанесения механической травмы. Методы и материалы. Методом флуоресцентной микроскопии отслеживали изменения [Ca2+]i) и m в первичной нейроглиальной культуре из коры головного мозга 1-2-дневных крыс. Возраст культуры в момент измерений 11-14 дней. Результаты. Обнаружено, что нейротравма вызывает скачок [Ca2+]i и совпадающее с ним по времени резкое падение m. Эти изменения затрагивали клетки, расположенные не далее 100мкм от границы травмы. Блокирование ионотропных глутаматных рецепторов NMDA-типа с помощью МК-801 снижало в 8,5 раз долю нейронов, имевших высокий подъем [Ca2+]i. Выводы. Поступления Са2+ в клетки при механическом повреждении первичной нейроглиальной культуры происходит преимущественно по NMDA-каналам и отчасти, вероятно, по АТФ-активируемым каналам. Background. In vitro modeling of traumatic brain injury helps clarifying pathological mechanisms responsible for cell death or their subsequent dysfunction in detail, which is difficult to accomplish in vivo. Aim. To determine changes in intracellular free Ca2+ concentration ([Ca2+]i) and mitochondrial potential (m) in a primary neuroglial culture during infliction of a mechanical injury (scratch). Methods and materials. Changes in [Ca2+]i and m in the primary neuroglial culture from the cerebral cortex of 1-2 day old rats were monitored using a fluorescence microscopy technique. Measurements were performed in 11-14-day old cultures. Results. Neurotrauma resulted in a sharp increase in [Ca2+]i and a synchronous profound drop of m. These changes affected cells located not farther than 100 µm from the boundary of the injury. Inhibition of NMDA-type ionotropic glutamate receptors with MK-801 reduced by approximately 8.5 times the proportion of neurons, which indicated a high [Ca2+]i rise. Conclusion. Са2+ influx into cells during mechanical injury of the primary neuroglial culture occurs predominantly through NMDA-channels and perhaps partially through ATP-activated channels.

Abstract. Groups of immature mice were injected sc with radiocarbon-labelled alpha-aminoisobutyric acid (AIB) after being given a single sc injection of hFSH or of 0.9% saline. As an index of the transport of AIB, the specific activity of isotope was measured in homogenates of testis and of liver. FSH treatment caused statistically significant increases in the specific activity of isotope in the testes and in the ratio of testicular to liver specific activity. The effect was greatest in 9-day-old mice injected with FSH 16 h before removal of the testes. Uptake of labelled AIB was not stimulated after administration of hCG or testosterone. Doses of cycloheximide sufficient to reduce the rate of protein synthesis by over 99% did not impair testicular uptake of labelled AIB or the influence of FSH on AIB uptake. These results suggest that FSH stimulates amino acid transport into cells of the immature testis and that this action is independent of the stimulatory effect of FSH on testicular protein synthesis.

To study the relation between dietary-induced increase of intestinal lactase activity and lactose absorption, 11-wk-old rats were fed either a high-starch (70 cal%), low-fat (7 cal%) diet or a low-starch (5 cal%), high-fat (73 cal%) diet for 7 days. Food intake and body weight changes were similar in the two dietary groups. In the first experiment, lactose absorption was studied in vivo after oral administration of 600 mg lactose (10% solution in water with added [3H]PEG) to rats fasted for 16 h. Groups of rats were killed at time 0 and at 1-h intervals for the next 3 h. Lactase activity and lactose absorption were significantly higher (P less than 0.01) in the high-starch group than in the low-starch group. In the subsequent experiment, 9-wk-old rats were fed the two isocaloric diets for 3 days. By use of the everted sac technique, we have demonstrated a significantly higher absorption of monosaccharides from lactose in the high-starch diet group; also, glucose transport was higher in the high-starch diet-fed animals. When Tris, an inhibitor of lactase, was added into the mucosal fluid, absorption of lactose was abolished and no effect was seen on glucose absorption (in vivo and in vitro). In both experiments, significant linear regression was established between lactase activity and lactose absorption. Our results thus show that the increase in lactase activity, induced by feeding a high-starch diet to adult rats, is accompanied by an increased capacity to hydrolyze lactose and absorb the constituent monosaccharides.
