The objective of this study was to determine suitable cultivars to be used as pollinators for 0900 Ziraat. 0900 Ziraat was used as a female cultivar; Bigarreu Gaucher, Bing, Noble, Starks Gold, Stella, Van, and Vista were used for pollination in the experiments. Starks Gold had the highest values in terms of anther number, average number of pollens per anther, number of pollen per flower and the morphological homogeneity. The pollen viability rates showed significant differences according to stain tests. <i>In vitro</i> pollen germination in 0.5% agar + 15% sucrose + 5 ppm boric acid medium increased with increasing incubation period, and the highest germination was obtained after 48 hours for all cultivars. In orchard trials parallel to pollen tube growth experiments in the laboratory, 0900 Ziraat &times; Starks Gold combination gave the best fruit set results.

In vitro and in vivo studies on pollen germination of Paeonia tenuifolia L. (Paeoniaceae) revealed that pollen grains are shed at two-celled stage. Normal and abnormal pollens were observed. Pollen viability was recorded between 55 and 75%. In vitro studies revealed 85% germination and usually the germination was monosphonic. Some pollen tubes with swollen tube tip and undulations were found. Styles and stigma were found to contain many pollen tubes 24 hrs after pollination. The effect of boric acid (0-450 ppm) and sucrose (0-40%) on pollen germination and pollen tube growth in Eucalyptus globulus, E. morrisbyi, E. ovata and E. tirnigera was examined in vitro. Over the con- centrations tested, sucrose had by far the largest effect upon both pollen germination and tube lengths. The optimum sucrose concentration for pollen germination (30%) and pollen tube growth (20%) differed markedly with very little (<lo%) germination occurring in the absence of sucrose. The interaction of sucrose and boric acid was significant. However, in general both pollen germination and pollen tube growth were increased by the addition of up to 100 ppm boric acid, but above this level the response plateauxed. The four species differed significantly in their pattern of response to both boric acid and sucrose and the predicted optima derived from analysis of response surfaces differed between species. The predicted sucrose concentration for optimal germination and growth of E. urnigera pollen was consistently less than the other species and in terms of the optimal level of boric acid for pollen tube growth species can be ranked in the order E. globulus > E. ovata > E. morrisbyi = E. urnigera. Pollen germination and tube growth of all four species on a medium comprising 20% sucrose and 200 ppm boric acid would not differ significantly from the observed maximum response of each species and this could suffice as a generalised medium. However, if only percentage germination is to be assessed 30% sucrose would be preferable. It is argued that subtle interspecific differences in optimal in vitro con- ditions for pollen germination and pollen tube growth are likely to reflect differences in pollen physiology which in vivo may have important implications for the success of hybridisation where pollen competition occurs.

The effect of boron (B) on in vivo and in vitro development of almond [Prunus dulcis (Mill.) D.A. Webb (syn. P. amygdalus Batsch)] pollen and pollen tubes and the resultant effect on fruit set was studied in mature trees. The cultivars Mono (pistil donor) and Butte (pollinizer) in an orchard with low soil B in Fresno, California were sprayed with B at 0, 0.8, 1.7, or 2.5 kg·ha-1 during Fall 1993. Pollen viability as indicated by the fluorescein diacetate method (FDA) was >85% and was not affected by field-applied B, however, in vivo pollen germination and tube growth were enhanced by foliar-applied B. More effect of applied B on in vivo growth appeared as pollen tubes progressed toward the ovary. For in vitro germination, foliar-applied B reduced bursting of tubes, and addition of B to the culture media significantly increased pollen germination and pollen tube growth.

SUMMARYSix pairs of isogenic lines of sorghum (Sorghum bicolor L. Moench) were sown in field plots in Montecillo, State of México (2240 m altitude), in 2005 and 2006. Crosses A (♀)×B (♂) were done in each pair. In A-lines, the length of pistil, stigma, style and ovary, as well as the ovary width, were measured. In B-lines, pollen diameter, viability (cytoplasm density) and production were evaluated. Pollen germination and pollen tube growth in the pistils of the A-lines, were quantified in vivo with aniline blue and epifluorescence 18 h after pollination (HAP), while fertilized pistils were counted at 96 HAP. Histological studies on both pollinated and non-pollinated pistils were performed in one male-sterile line. Seed yield, mean-seed weight, seeds per panicle and seed set (SS; seeds/flower/panicle) were determined at harvest. Pollen viability was the variable most related to pollen germination and pollen tube growth. Stigma receptivity was not associated with its morphology. Growth of the pollen tube in stigma, style and ovary was observed in the transmitting tissue 18 HAP, running parallel to the vascular tissue. Yield under chilling field temperatures (minimum average of 6 and 8°C) prevailing during flower development and pollination ranged from 7 to 12 g/panicle. The differences in seed production and SS among A×B crosses did not depend on the amount and viability of pollen.

Climate change may influence the composition of plant communities by affecting the reproduction, growth, establishment and local extinction of plant species. Predicting the effect of climate change may provide insight into the impact and relationship between weather pattern and flowering phenology in long term studies. Pollen viability is one important factor of reproduction. Pollen viability is essential for a good fruit set. The study is undertaken to evaluate the influence of temperature and rainfall fluctuation pattern on floral phenology and pollen viability in the restricted distribution of the plant Ehretia pubescens Benth. Field observation on floral phenology has revealed changes occurring in the pollen viability with the change of temperature and rainfall. The plants show drought resistant; however, it is observed that it blooms immediately after the rainfall. Change in the rainfall pattern results in change in flowering pattern. On the natural habitat fruit set is a good indicating of good germinability of pollen grain in vivo. In vitro pollen germinability is less efficient for this species as supported by the data.

Pollen germination timing has a paramount role in fertilization of a flower. Rapid germination and outgrowth of a pollen tube that penetrates the stigma is required. Physical and biological factors can affect pollen germination timing. The objective of this study was to determine if ACC oxidase antisense gene expression could influence in vitro pollen germination and in vitro pollen tube length growth. A transgenic (ACC oxidase antisense) `Galia' male parental line had a reduced fruit set compared to its wild type. Likewise, embryo abortion and empty seeds after self-pollination in a `Galia' male parental line were observed. Wild type and transgenic `Galia' male parental line melon plants were grown in a greenhouse according to the practices of Rodriguez (2003). Male flowers were collected from these plants between 10 to 12 am; pollen was obtained by dipping the anther in germination medium (10.25% sucrose, 0.031% calcium nitrate, 0.015% boric acid, 0.0075% KNO3, and 0.016% MgSO4) at 25 °C and analyzed immediately, either for total percentage of germination after 5 minutes of incubation or to measure pollen tube growth rate every 5 minutes during 1 hour. Each flower provided an average of 250 pollen grains. Assays were conducted by using the “Hanging Drop Method” (Okay and Ayfer, 1994). Percentage of pollen germination in WT `Galia' male parental line was greater than the transgenic line. Likewise, in vitro pollen tube growth in wild type `Galia' melon was greater than pollen from the transgenic line. Possibly the ACC oxidase antisense gene expression in `Galia' male parental line may have had an influence on the reduced fruit set observed.

The objective of this study was to evaluate the influence of two organic nanofertilizers, Lithovit and Nagro, on in vitro germination, pollen tube elongation and pollen grain viability of Pisum sativum L cv. Pleven 4. The effect of their application was high and exceeded data for the untreated control (44.2 and 47.23 % regarding pollen germination and pollen tube elongation, respectively), as well as the effect of the control organic algal fertilizer Biofa (17.5 and 27.9 %, respectively). Pollen grains were inoculated in four culture media. A medium containing 15% sucrose and 1% agar had the most stimulating impact on pea pollen grains. Pollen viability, evaluated by staining with 1% carmine, was within limits of 74.72-87.97%. The highest viability of pollen grains was demonstrated after the application of Nagro organic nano-fertlizer.

The causes of poor fruit set of cocoa (Theobroma cacao L.) in the greenhouse were studied by examining factors that may influence pollen germination. Hand pollination of cocoa flowers resulted in 45.8% fruit set when flowers were pollinated within 3 hours of anthesis. Pollen germination did not occur until about 6 hours after pollination. Later pollinations (7 to 9 hours after anthesis) or earlier pollinations (16 to 18 hours before anthesis) did not lead to fruit set. Cocoa pollen did not germinate in vitro unless the excised flowers were incubated for 6 hours at 25C in closed vials. During the incubation period, CO2accumulated to a final concentration of about 85 ml·liter-1 as a result of respiration. Ethylene production was not detectable. Incubation of flowers with a NaOH-saturated wick, to absorb CO2, prevented pollen germination in vitro. Incubation of flowers at 15C also prevented pollen germination in vitro at 25C. Hand pollination of flowers 7 to 9 hours after anthesis or 16 to 18 hours before anthesis using CO2-incubated pollen resulted in about 10% fruit set. Enclosed pollinations in vivo, in which CO2 was allowed to accumulate, resulted in nearly 100% fruit set. The initial failure to set fruit from hand pollinations may result from poor or slow pollen germination. Moreover, CO,-incubated pollen might be used to increase fruit set in cocoa by extending the effective pollination period.

The study was conducted on the in vitro pollen germination at 26?, 28?, 32? and 35?C for 24h of male parental lines, pollen tube growth in vivo in cross pollination of female and male parental lines that make couples in four hybrids: ZP 504 su (? ZPPL 51 ? ? ZPPL 67); ZP 677 (? ZPPL 17 ? ? ZPPL 201); ZP 704 (? ZPPL 109 ? ? ZPPL 79), ZP 611 k (? ZPPL 126 ? ? ZPPL 105), and the open pollination of female parental lines of the above mentioned hybrids. Pollen germination in vitro and pollen tube growth dynamics in vivo showed different genotypic specificities with the tests applied. The obtained results were discussed in the context of reproductive biology of ZP maize lines and aimed to create the preconditions for successful management and direction of the process in practice - seed production in certain environmental conditions.
