Fifty-one grapevine cultivars registered in the Czech Republic were a subject of RAPD analysis. Nine grapevine varieties which were supposed to be genetically distant, were analysed by means of 120 Operon primers during the initial screening of RAPD method. Sixteen primers producing reproducible and polymorphic bands were then used for amplification of all registered varieties. A dendrogram of genetic relationships and a methodology of identification of each variety were created. The resultant dendrogram mostly corresponds with known information about the origin and genetic relationships of the varieties. It is necessary to use the combination of RAPD markers for identification of each variety. It was possible to identify 26 out of 51 varieties registered in the Czech Republic by RAPD markers without any problems. Nine varieties were identifiable only on the basis of present and absent markers. Some varieties were very close relatives – they formed remote clusters. It was not possible to distinguish between them by RAPD method. Two varieties were not identifiable by the designed key.

Genetic analysis with RAPD markers has been extensively used to determine diversity among maize genotypes. The aim of the present study was to estimate genetic relationships among 70 genotypes, provided from SCDA Turda Cluj germplasm collection. RAPD analysis was performed with 14 decamer primers. These primers generated, among the studied genotypes, a number of polymorphic bands comprised between 13 bands (OPA 04) and 7 bands (OPAL 20). The highest numbers of polymorphic bands were obtained with primer OPA 04, respectively 13 bands, following by OPO 12 (12 polymorphic bands), OPAB 11 and OPA 17 (11 polymorphic bands). Lowest number was obtained with primer OPAL 20, respectively 7 polymorphic bands. Genetic distances were established using Nei-Li coefficient and UPGMA dendrogram was constructed with RAPDistance 1.04 software. The built dendrogram shows phylogenetic relationships between genotypes analyzed.

Genetic variability was detected in 15 varieties of triticale (XTriticosecale Wittmack., 2n = 6x = 42, BBAARR) registered in the Czech Republic by means of polymorphism of DNA using the RAPD method and the SSR method. For detection we used 80 RAPD primers. The lower reproducibility of the RAPD markers was resolved by means of repeated analyses (3–4 times). On the basis of statistical evaluation a dendrogram was set up, which allows highly significantly to differentiate the varieties Kolor, Modus and Tornado. The remaining 12 analysed varieties formed 4 clusters. In addition to the RAPD markers a protocol of detection of DNA polymorphism was elaborated and optimised with microsatellite (SSR) markers. For the analyses we used 2 SSR markers (1A chromosome [Xpsp2999] and 1B chromosome [Xpsp3000]), which have been discovered in wheat (T. aestivum L.). Basing on these two SSR markers the only variety Triamant was distinguished from the clusters of the other analysed varieties.

RAPD markers are used to study the genetic diversity of the main planting on 37 castor varieties widely cultivated in china according to the oil content and other characteristic of different castor varieties. Genetic distance of 37 Chinese castor varieties is studied by RAPD markers analysis. RAPD analysis shows that a total of 122 bands are amplified from random primers of 20 S series, including 71 polymorphic bands with polymorphic rate of 58.20%. 37 castor beans are divided into four major groups in the phylogenetic tree. One castor germplasm is included in1, 2, 3 groups respectively, and two sub-groups are included in the 4 major group.

Collard (Brassica oleracea L. var. acephala) is an important vegetable the southeastern U. S. There are few (about 10) commercial cultivars, half being open-pollinating (OP) lines, the remainder more recent F1 hybrids. There is a potential untapped B. oleracea germplasm pool in the form of collard landraces perpetuated by southeastern gardeners and farmers. To determine the amount of genetic variation among cultivars and also whether landraces represent unique genotypes, ten cultivars and eight lines or landraces were evaluated using RAPD analysis. Decamer primers were used to amplify total genomic DNA and to differentiate collard lines and other B. oleracea crop cultivars. Additionally, individuals of an OP collard cultivar and a land-race were analyzed to evaluate intra-line variation. Virtually all primers detected polymorphic bands among lines although some identified considerably more variants. Intra-line analysis indicated that OP lines are genetically broad-based populations. Many unique RAPD markers were identified in landraces indicating that the lines represent unique genotypes and that further line collection is warranted.

A total of 118 Pyrus sp. (pear) and cultivars native mainly to east Asia were subjected to randomly amplified polymorphic DNA (RAPD) analysis to evaluate genetic variation and relationships among the accessions. Two hundred fifty RAPD markers were scored from 20 decamer primers. RAPD markers specific to species were identified. Clustering analysis revealed two divisions: one comprising cultivars of P. communis L., and the other including all accessions of Pyrus native to east Asia. The grouping of the species and cultivars by RAPD data largely agrees with morphological pear taxonomy. However, some noted incongruence existed between two classification methods. Pyrus calleryana Dcne. clustered together with P. koehnei Schneid., P. fauriei Schneid. and P. dimorphophylla Makino. Pyrus betulaefolia Bge. clustered with P. ×hopeiensis Yu and P. ×phaeocarpa Rehd. A noncultivated clone of P. aromatica Kikuchi et Nakai grouped with P. aromatica cultivars. Pyrus hondoensis Nakai et Kikuchi and cultivars of P. ussuriensis Max. formed a single group. Some accessions from Korea (named Korean pear) had species-specific RAPD markers and comprised an independent group. Most of the Chinese white pears clustered together with most of the Chinese sand pears. Based on the present results, the new nomenclature P. pyrifolia var. sinensis (Lindley) Teng et Tanabe for Chinese white pear was suggested. Most accessions of Japanese pears fell into one main group, whereas pear cultivars from Kochi Prefecture of Japan subclustered with some Chinese sand pears and one accession from Korea. Our results infer that some local Japanese pear cultivar populations may have been derived from cultivars native to Kochi Prefecture in Shikoku region, and that the latter may have been introduced from ancient China and/or Korea.

Genetic relationships among 42 grape accession of at least 15 species were estimated and compared using RAPD and isozyme techniques. These accessions were either hybrids or wild collections of the Asiatic species, the American species, the European grape (V. vinifera), and muscadine grape (V. rotundifolia). A total of 196 RAPD fragments were generated from twenty 10-mer primers. The pairwise similarities among the accession ranged from 0.46 to 0.94. A dendrogram was generated based on the RAPD similarity coefficients. Species/accessions were basically grouped together in accordance with their geographic origins. The similarities and dendrogram resulted from the RAPD analysis were consistent with the ones generated from the isozyme data, and also consistent with the known taxonomic information. This result suggest that the RAPD method, like isozyme, is an useful tool for studying grape genetic relationship/diversity and origination.

Population and evolutionary analyses of pathogenic bacteria are frequently hindered by sampling strategies that concentrate on isolates from patients with invasive disease. This is especially so for the gram-negative diplococcus Neisseria meningitidis, a cause of septicemia and meningitis worldwide. Meningococcal isolate collections almost exclusively comprise organisms originating from patients with invasive meningococcal disease, although this bacterium is a commensal inhabitant of the human nasopharynx and very rarely causes pathological effects. In the present study, molecular biology-based techniques were used to establish the genetic relationships of 156 meningococci isolated from healthy young adults in the Czech Republic during 1993. None of the individuals sampled had known links to patients with invasive disease. Multilocus sequence typing (MLST) showed that the bacterial population was highly diverse, comprising 71 different sequence types (STs) which were assigned to 34 distinct complexes or lineages. Three previously identified hyperinvasive lineages were present: 26 isolates (17%) belonged to the ST-41 complex (lineage 3); 4 (2.6%) belonged to the ST-11 (electrophoretic type [ET-37]) complex, and 1 (0.6%) belonged to the ST-32 (ET-5) complex. The data were consistent with the view that most nucleotide sequence diversity resulted from the reassortment of alleles by horizontal genetic exchange.

Silybum marianum is an important medicinal plant used for hepatic disorders. Two varieties exist for this plant S. marianum var. album (white-flowered) and S. marianum var. purple (purple-flowered). A fast and simple method (RAPD) was developed for authentication of S. marianum varieties (purple- and white- flowered) at a DNA level. The two varieties were distinguished by polymorphic bands generated by four decamer primers, namely, OPP-10, OPG-03, OPG-01, and OPC-17. The present study aims to investigate the genetic variation in Cinnamomum tamala Nees. (Lauraceae) accessions collected from different locations of Uttarakhand Himalaya. The leaves samples were analyzed by using RAPD and ISSR markers. A total 22 primers were used for initial screening in order to select the ones giving good amplification. Seven primers (OPA-12, OPA-18, OPB-10, OPB-17, ISSR-21, ISSR-24 and ISSR-30) were found to be polymorphic in eight accessions. Based on combined profile of ISSR and RAPD markers the dendrogram was constructed by using UPGMA cluster analysis and all the accessions showed discrimination from one another. All the accessions were clustered into two major groups, one containing T1-T4 and second T5-T8. The percentage of polymorphic bands was 86.3 % for three ISSR markers and 70.5 % for four RAPD markers. This emphasize that ISSR markers were found to be the best for genetic variation in the species.

Morphological characterisation allows isolates of Colletotrichum gloeosporioides, Colletotrichum musae and Colletotrichum acutatum to be identified only to species level. Pathogenicity tests and random amplified polymorphic DNA (RAPD) markers distinguished a mango biotype of C. gloeosporioides from eight other isolates of C gloeosporioides obtained from five different fruit species. Using these procedures, it was also possible to distinguish C. acutatum and C. musae both from each other, and from the C. gloeosporioides isolates. In cross-infectivity studies, isolates of C. gloeosporioides displayed a wide host range with the exception of isolates from mango, which were highly virulent on mango only. Teleomorphic isolates of C. gloeosporioides were clustered together by RAPD analysis. This work has demonstrated the existence of a biotype of C. gloeosporioides which shows specialisation to mango.
