During 2014–2015, samples from ten 20–25 years old pine (Pinus ponderosa var. jeffreyi) trees showing wilt symptoms were collected from the Arborétum Mlyňany park. A disease was observed on 20% of the trees. The first symptoms are wilting, stunting, chlorosis, and discolouration of needles, which turned yellow on affected twigs, then red and finally they fell off. Isolations of the pathogen were done from the discoloured tissues of needles (twenty samples from each tree) on Potato Dextrose Agar. Colonies of the fungus (3–4 Petri dishes from each tree) were initially aerial, white or slightly violet, but with age they became red and red pigments were produced in agar. The observed micromorphological characteristics of the fungus, such as presence of simple and proliferating conidiophores with polyphialides, microconidia, macroconidial shape, and chlamydospore presence matched the description of Fusarium sporotrichioides. The identity of the fungus was confirmed by phylogenetic analysis of internal transcribed spacer (ITS) sequences. Sequence comparisons placed the fungus to the species F. sporotrichioides with similarity of 99.6% at the ITS sequence level.

AbstractPhylogenetic trees based on group I intron sequences and on internal transcribed spacer (ITS) sequences of mycobiont ribosomal genes were calculated and compared. Eight cetrarioid and four non-cetrarioid species of the Parmeliaceae were compared. The phylogeny based on group I intron sequences is partly congruent with the ITS sequence phylogeny. Group I intron sequences are presumably less informative for infragenic studies. The introns have a length of 214–233 nucleotides, and differ at up to 33% of the bases between species. All introns analysed are located between the positions 1516 and 1517 of the fungal 18S ribosomal RNA gene. Cetrarioid lichens form a non-homogeneous group within the Parmeliaceae according to both group I intron and ITS sequences.

To date, there are very few sequence data for Cyperaceae from mainland Southeast Asia. The aim of the present study was to contribute nuclear ribosomal internal transcribed spacer (ITS) sequences of selected species of Cambodian Cyperaceae to the overall phylogeny of the family. We generated ITS sequences of 38 accessions representing 26 species from Cambodia and used these sequences for phylogenetic analysis together with similar sequences from the National Center for Biotechnology Information GenBank. Our results corroborate recent phylogenetic work in the family and largely confirm established tribal relationships. The backbone of the phylogenetic tree of species-rich genera that have undergone rapid radiations is often weakly resolved (e.g. in Fimbristylis and in the C4 clade of Cyperus). Cryptic variation was revealed in the taxonomically difficult group of Fimbristylis dichotoma, with samples of this taxon appearing in two distinct clades within Fimbristylis. Further addition of geographically spread accessions of taxa will improve our understanding of the complex biogeographical history of the genera in the family. Eleocharis koyamae Tremetsb. & D.A.Simpson is proposed as a new name for E. macrorrhiza T. Koyama.

Three new species of Russula collected from southern China are proposed based on morphological characters and phylogenetic analysis of internal transcribed spacer (ITS) sequences. Russula bubalina sp. nov. is characterized by cinnamon buff to pink pileus with striate margin, interveined and forked lamellae, basidiospores with warty ornamentations not forming reticulum, hymenial cystidia becoming brown in sulphovanillin (SV) and slender terminal cells in suprapellis. R. pseudobubalina sp. nov. is closely related to Russula bubalina in macro-morphology, but it can be recognized by its unforked lamellae, shorter cheilocystidia, bigger basidia and basidiospores with lower ornamentations. Russula subatropurpurea sp. nov. can be recognized by the purplish brown pileus, mild-tasted and white context changing to light purplish red with FeSO4, white and forking lamellae without lamellulae, long hymenial cystidia becoming brown in SV. Both morphological and phylogenetic analysis consistently confirmed the distinct positions of three new species in subg. Heterophyllidia subsection Heterophyllinae.

The study investigated fungi associated with sea foods, fish (Pellonula leonensis) and crayfish (Parapandalus pritis) from different wetlands (Ibeno, Ikot Abasi, Eket and Itu) in Akwa Ibom State. Samples were collected and cultured on Potato Dextrose Agar (PDA) and the isolated fungi were identified using molecular technique. Based on PCR amplification, sequencing of the internal transcribed spacer and phylogenetic analysis, the fungi identified were: Aspergillus niger, A. felis, A. foetidus, A. aculeatus, A. japonicus, A. flavus, A. tamari, A. terreus, Penicilium citrinum, Candida tropicalis and Trametes polyzona. The most commonly isolated fungi from fish for the four locations were A. niger (52.8) in Itu, P. citrinum (51.7) in Ibeno, A. foetidus (53.5) in Ikot Abasi and A. aculeatus (51.6) in Eket. For samples of crayfish, the most commonly isolated fungi were A. niger (25.1) in Itu and A. foetidus (20.1) in Ikot Abasi. Generally, Ibeno recorded the highest frequencies of fungal isolates while Ikot Abasi had the least. Percentage occurrence of fungi isolated from fish samples were significantly higher (p<0.05) when compared with those isolated from crayfish samples. The presence of these fungi showed that sea foods were exposed to an increasing number of virulent infectious diseases in natural populations, and fungal-like diseases can cause some of the most severe die-offs and extinctions ever witnessed in wild species thus jeopardizing food security Keywords: Fungi, Sea food, Wetland, Aspergillus, Fish.

Species of Clavaria (Clavariaceae, Agaricales) collected from China were studied using morphological and molecular methods. Two species, C. griseolilacina and C. sinensis were described as new species; both possess branched and pale purple-tinged fruit bodies. However, the former possess a few branched, while the latter is profusely branched. The phylogenetic analysis based on the internal transcribed spacer (ITS) sequences confirmed the distinctions between these two new species and their close relationships with C. rubicundula and C. fumosa in the genus. An evolutionary tendency in genus Clavaria was discussed. Additionally, morphological features of the two new species were described in detail and were illustrated with line drawings and photographs. Holotypes of the new species are deposited in the Mycological Herbarium of Hunan Normal University (MHHNU). The ITS sequences generated in this study were deposited in GenBank. A key is provided for the known Clavaria species in China.

To find the differences in the internal transcribed spacer(ITS) sequences and provide scientific data for the authentication of Potentilla chinensis and its related species, we extracted the genome DNA from the leaves of 5 common Potetilla species in Jilin Province, amplified the ITS region using ITS universal primers of angiosperm, and sequenced the purified PCR products directly. Polymorphism of ITS sequences was found within P. chinensis and the sequence data suggested that our samples of this species might be related to hybridization. Other 4 species showed intraspecies-stability in ITS sequence. The ITS sequences of these 5 Potentilla species are significantly different. So ITS sequence analysis and other methods derived from it can be used in authentication of Potentilla.

The aim of this study was isolation and molecular identification of laccase-producing saprophytic/phytopathogen Basidiomycetes species from different geographic regions with dominant vegetation of <em>Pinus</em>, <em>Abies</em>, and <em>Quercus</em> spp. in the state of Michoacán, Mexico. Soil samples and visible mycelial aggregates were collected for fungal isolations. Soil samples were processed using a soil particle washing technique, where a selective Ascomycetes inhibitor and guaiacol, as an indicator of saprophytic Basidiomycetes growth, were used. Most of the isolates were obtained from samples collected in Parque Nacional, José Ma. Morelos (Km 23), Charo, Michoacán, Mexico. Based on sequence comparisons and phylogenetic analysis of internal transcribed spacer regions (ITS1-5.8S-ITS4) with respect to reference taxa, identification of saprophytic/phytopathogen Basidiomycetes species was carried out. In total, 15 isolates from 12 genera (i.e., <em>Bjerkandera</em>, <em>Coriolopsis</em>, <em>Ganoderma</em>, <em>Hexagonia</em>, <em>Irpex</em>, <em>Limonomyces</em>, <em>Psathyrella</em>, <em>Peniophora</em>, <em>Phlebia</em>, <em>Phlebiopsis</em>, <em>Trametes</em>, and <em>Trichaptum</em>) and one species from family Corticiaceae were identified. This study will be useful for further investigations on biodiversity of soil Basidiomycetes in different ecosystems. At present, these isolates are being used in our various lab experiments and can be useful in different industrial and bioremediation applications.
