Background:Rheumatoid arthritis (RA) is a systemic autoimmune disease characterized by the presence of rheumatoid factor (RF) and anticitrulline autoantibodies. Recent evidences suggest that impairment of neutrophil extracellular traps (NETs) could exert substantial influence on RA pathogenesis. The production of NETs depends heavily on the ROS generation. One of its mechanisms is xanthine oxidoreductase (XOR) mediated degradation of purine metabolites. Analysis of pro-oxidant activity of the enzymatic complex XOR and its constituents, xanthine oxidase (XO) and xanthine dehydrogenase (XDG), is an issue of considerable interest in this context.Objectives:Evaluation of XO and XDG activities in RF-positive and RF-negative RA using both plasma and lysed lymphocyte samples.Methods:The research was carried out in agreement with the WMA Declaration of Helsinki principles. Diagnosis of RA had been verified using ACR/EULAR 2010 criteria. Enzymatic activities in plasma and lymphocytes were measured spectrophotometrically and expressed as nmol/min/ml. Enzymatic activities in lymphocytes were also normalized to 1×107 cells/ml. Statististical tests were selected in line with common guidelines. Differences were considered significant when p<0.05. Reference ranges were calculated as means ±2SD.Results:75 adult RA patients (52 females and 23 males, mean age 43.9±0.97 years, mean disease duration 8.5±0.3 years) from the rheumatology unit of Volgograd Clinical Emergency Hospital #25 as well as 35 healthy controls were included in the study. RF-positive RA and RF-negative RA were observed in 49 (65.3%) and 26 (34.7%) patients, respectively. Reference ranges for plasma and lymphocyte XO activities were 2.60-3.96 and 14.2-27.8 nmol/min/ml, respectively. Similar ranges for XDG activities were 4.49-5.93 and 22.5-40.7 nmol/min/ml, respectively. Enzymatic profile of RA patients is characterized by significantly increased XO activity in plasma and decreased XO and XDG activities in lymphocytes (р<0.001). XO activity is increased (p<0.001), XDG activity is decreased (p<0.001) in blood plasma of patients with RF-negative RA, while the activity of both enzymes is decreased in lymphocytes (p<0.001). XO activity (p<0.001) and XDG activity (p<0.05) is increased in blood plasma, XO activity and XDG activity are decreased (p<0.001) in lymphocytes of patients with RF-positive RA. Plasma XO and XDG activities are also higher, and lymphocyte XO and XDG activities are lower in patients with RF-positive RA than in patients with RF-negative RA (р<0.001).Conclusion:Our study revealed the relationship between enzyme parameters and rheumatoid factor presence. More pronounced changes in the enzyme activities were observed in patients with RF-positive RA. These results demonstrate that activation of the xanthine oxidase/xanthine dehydrogenase enzyme complex is an substantial factor of induction and continuation of the autoimmune rheumatoid inflammation.Disclosure of Interests:None declared

Abstract The aim of this study was to determine whether the introduction of stress affects corticosterone and malonyl dialdehyde levels and antioxidant indices of the blood of turkey hens, and whether additives used in conjunction with stress, such as aloe extract supplemented with trans-resveratrol and vitamin C or the amidrazone derivative 5-oxo-1,2,4-triazine, can mitigate adverse changes that may occur in these parameters. The experiment was carried out on 360 turkey hens allocated randomly to 6 groups of 60 birds each. Groups C and C(+)stress were the control groups and did not receive any additive. Birds from groups A and A(+)stress were administered aloe extract with the addition of trans-resveratrol and vitamin C in the amount of 0.70 ml/kg body weight (BW)/ day. The turkey hens from groups T and T(+)stress received 5-oxo-1,2,4-triazine in the amount of 30 μg/kg BW/day. Blood tests included determination of the content of corticosterone and malonyl dialdehyde (MDA), total plasma antioxidant potential (FRAP) and vitamin C, as well as activity of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT ) and glutathione peroxidase (PGx). The blood serum samples were also analysed for levels of zinc (Zn), copper (Cu) and iron (Fe) by atomic absorption spectrometry (AAS). The stress factors applied resulted in a significant increase in the level of corticosterone and MDA and in SOD and CAT activity in the blood plasma of the turkey hens. A significant decrease in the level of vitamin C and total plasma antioxidant potential were also recorded in the birds treated with stress. The aloe preparation supplemented with trans-resveratrol and vitamin C caused a significant increase in vitamin C, FRAP, Fe and Cu, and a decrease in the corticosterone and MDA levels in the blood plasma of the turkey hens, while in the blood plasma of birds treated with 5-oxo-1,2,4-triazine a significant increase was noted in superoxide dismutase activity and a decrease in the malonyl dialdehyde level. In order to alleviate the negative effect of stress, supplementation of diets with aloe extract supplemented with resveratrol and vitamin C may be considered.

The state of the blood antioxidant system in the patients presenting with acromegaly. The objective of the present study was to evaluate the total antioxidant activity (TAA) of blood plasma, the levels of non-protein thiols and 2-thyobarbituric acid-active products (TBA-AP), superoxide dismutase (SOD) and catalase (CAT) activities as well as ceruloplasmin (CP) level in the patients presenting with acromegaly. It was shown that plasma TAA and SOD activity in this patients was on the average 20 and 30% lower respectively than in the control subjects. At the same time, the TBA-AP and CP levels increased by 50 and 40% respectively. These data suggest the development of oxidative stress in the acromegalic patients.

The stable metabolites rate of nitric oxide is reduced three times compared with the rate in the group of control animals in homogenates of rats’ gingival tissue with gastritis and duodenitis. Increasing of the catalase rate with simultaneous increasing of lipids and proteins oxidation was accompanied by decreasing activity of superoxide dismutase. Decreasing of the malonic dialdehyde content and indices protein molecules fragmentation in the blood plasma with simultaneous increasing of catalase and nitric oxide metabolites levels indicates the leading role of nitric oxide in antioxidant protection of the organism in conditions of disease.

The aim of present work was to assess the role of immunological disorders, hypoxia and lipoperoxidation in development and progression of erosive/ulcerative lesions of duodenum accompanied by chronic cerebrovascular insufficiency. We have studied a cohort of 125 patients with erosive and ulcerative lesions of duodenum associated with chronic disorders of cerebral circulation, aged from 48 to 74 years old. They underwent outpatient treatment and care at the Clinical Hospital No.5 (Saransk) over 2015-2018. A comparison group consisted of 39 patients with chronically impaired cerebral circulation, who did not show signs of erosive/ulcerative lesions of stomach and duodenum over the study period. Patients received conventional anti-ulcer therapies for 14 days. The cytokine profile, indexes of hypoxia, and serum markers of lipid peroxidation were evaluated. The study has shown an increase in the level of pro-and anti-inflammatory cytokines in cases with erosive/ ulcerative duodenal lesions associated with chronic impairment of cerebral circulation throughout the observation period, thus indicating to evolving imbalance of immunoregulatory system. The development of hypoxic changes in the blood plasma was observed, which was confirmed by increased contents of lactic and pyruvic acids, like as the hypoxia coefficient at all the observation terms. The lipid peroxidation processes were also activated in the course of evolving disorder, as judged by increased contents of diene conjugates, triene conjugates and the malonic dialdehyde levels upon admission, as well as on day 7 and 14 of observation, associated with decrease in the antioxidant potential of blood plasma, estimated by the superoxide dismutase index. A trend for a decrease in the indexes of diene conjugates, triene conjugates, malonic dialdehyde and an increased levels of superoxide dismutase was registered as late as after 30 days of observation. Hypoxia and lipid peroxidation may be viewed as predictors of chronic inflammation. Developing imbalance between pro-inflammatory, anti-inflammatory cytokines, and chronic hypoxia are of significant value for pathogenesis of erosive-ulcerative lesions of duodenum, being associated with chronic impairment of cerebral circulation. Under such conditions, oxidative stress is observed, with a shift of equilibrium towards pro-oxidants, along with exhausted potential of antioxidant defense system. In general, these disturbances determine the pathogenetic mechanisms of damage that occur in erosive and ulcerative lesions of duodenum, being associated with chronic cerebrovascular insufficiency, which have a mutually negative influence in the course of disease progression.
