Water stress induced proline accumulation and antioxidative enzymes in groundnut (Arachis hypogaea L.)

Thirty days grown seedlings of eight genotypes of groundnut viz. RHRG-6083, TAG-24, TG-60(LC), Karad-4-11, SB-XI, RHRG-6097, RHRG-6021, and RHRG-6055 were subjected to moisture stress by withholding irrigation for 15 days in pot culture experiment in order to evaluate its effect of on RWC , proline accumulation, soluble proteins, chlorophyll content and activities of superoxide dismutase (SOD), peroxidase (POX), glutathione reductase (GR) and lipid peroxidation (MDA).The levels of proline, soluble protein and the activity of all four antioxidative enzymes were found to be increased with MDA content in all genotypes of groundnut during water stress, however the chlorophyll and RWC content were found to be decreased. This study revealed that under water stress condition SB-XI, TAG-24 and RHRG-6021 showed higher increase of proline content and activities of antioxidative enzymes with lower reduction in RWC and chlorophyll. These three genotypes are stable during stress and seems to be promising for drought tolerance.

Abstract Growth of plants is severely reduced due to water stress by affecting photosynthesis including photosystem II (PSII) activity and electron transport. This study emphasised on comparative and priority targeted changes in PSII activity due to progressive drought in seven populations of Panicum antidotale (P. antidotale) collected from Cholistan Desert and non-Cholistan regions. Tillers of equal growth of seven populations of P. antidotale grown in plastic pots filled with soil were subjected progressive drought by withholding water irrigation for three weeks. Progressive drought reduced the soil moisture content, leaf relative water content, photosynthetic pigments and fresh and dry biomass of shoots in all seven populations. Populations from Dingarh Fort, Dingarh Grassland and Haiderwali had higher growth than those of other populations. Cholistani populations especially in Dingarh Grassland and Haiderwali had greater ability of osmotic adjustment as reflected by osmotic potential and greater accumulation of total soluble proteins. Maximum H2O2 under water stress was observed in populations from Muzaffargarh and Khanewal but these were intermediate in MDA content. Under water stress, populations from Muzaffargarh and Dingarh Fort had greater K+ accumulation in their leaves. During progressive drought, non-Cholistani populations showed complete leaf rolling after 23 days of drought, and these populations could not withstand with more water stress condition while Cholistani populations tolerated more water stress condition for 31 days. Moreover, progressive drought caused PSII damages after 19 days and it became severe after 23 days in non-Cholistani populations of P. antidotale than in Cholistani populations.

The aim of this research was to gauge the alternations in the lipid peroxidation and antioxidative enzyme activity in two cultivars (cv. RGS003 and cv. Sarigol) of canola under drought stress and drought tolerance amelioration by penconazole (PEN) and calcium (Ca). Plants were treated with different polyethylene glycol (PEG) concentrations (0, 5, 10 and 15%) without or with PEN (15 mg L–1) and Ca (15 mM). The Ca treatment prevented the negative effects of drought on fresh weight (FW) in RGS003 and Sarigol at 5 and 15% PEG respectively. Ca and PEN/Ca treatments caused significant induction in the proline content in Sarigol at 15% PEG; the latter treatment was accompanied by higher glycine betaine (GB), lower malondialdehyde (MDA) and growth recovery. Hydrogen peroxide (HO2) content in Sarigol was proportional to the severity of drought stress and all PEN, Ca and PEN/Ca treatments significantly reduced the H2O2 content. PEN and PEN/Ca caused alleviation of the drought-induced oxidative stress in RGS003. RGS003 cultivar exhibited significantly higher antioxidative enzymes activity at most levels of drought, which could lead to its drought tolerance and lower MDA content. In contrast to that of Sarigol, the activity of catalase and superoxide dismutase (SOD) increased with Ca and PEN/Ca treatments in RGS003 under low stress. The application of PEN and Ca induced significantly P5CS and SOD expression in RGS003 under drought stress after 24 h. Overall, these data demonstrated that PEN and Ca have the ability to enhance the tolerance against the drought stress in canola plants.

The objective of this study was to evaluate the competitiveness of two cultivars of upland rice drought-tolerant, cultured in coexistence with weed S. verticillata, under conditions of absence and presence of water stress. The experiment was conducted in a greenhouse at the Experimental Station of the Universidade Federal de Tocantins, Gurupi-TO Campus. The experimental design was completely randomized in a factorial 2 x 2 x 4 with four replications. The treatments consisted of two rice cultivars under two water conditions and four densities. At 57 days after emergence, were evaluated in rice cultivars and weed S. verticillata leaf area, dry weight of roots and shoots and total concentration and depth of roots. Was also evaluated in rice cultivars, plant height and number of tillers. Water stress caused a reduction in leaf area, the concentration of roots and vegetative components of dry matter (APDM, and MSR MST) of rice cultivars and Jatoba Catetão and weed S. verticillata. The competition established by the presence of the weed provided reduction of all vegetative components (MSPA, and MSR MST) of cultivars and Jatoba Catetão. It also decreased the number of tillers, the concentration of roots and leaf area. At the highest level of weed competition with rice cultivars, a greater decrease in vegetative components and leaf area of culture, regardless of water conditions.

Accurate information of crop growth conditions and water status can improve irrigation management. The objective of this study was to evaluate the performance of SAFYE (simple algorithm for yield and evapotranspiration estimation) crop model for simulating winter wheat growth and estimating water demand by assimilating leaf are index (LAI) derived from canopy reflectance measurements. A refined water stress function was used to account for high crop water stress. An experiment with nine irrigation scenarios corresponding to different levels of water supply was conducted over two consecutive winter wheat growing seasons (2013–2014 and 2014–2015). The calibration of four model parameters was based on the global optimization algorithms SCE-UA. Results showed that the estimated and retrieved LAI were in good agreement in most cases, with a minimum and maximum RMSE of 0.173 and 0.736, respectively. Good performance for accumulated biomass estimation was achieved under a moderate water stress condition while an underestimation occurred under a severe water stress condition. Grain yields were also well estimated for both years (R2 = 0.83; RMSE = 0.48 t∙ha−1; MRE = 8.4%). The dynamics of simulated soil moisture in the top 20 cm layer was consistent with field observations for all scenarios; whereas, a general underestimation was observed for total water storage in the 1 m layer, leading to an overestimation of the actual evapotranspiration. This research provides a scheme for estimating crop growth properties, grain yield and actual evapotranspiration by coupling crop model with remote sensing data.
