Fecundity and preferential oviposition by pulse beetle, Callosobruchus maculatus F on chickpea (Cicer arietinum L.) var dollar

In a laboratory experiment female Callosobruchus maculatus distributed eggs on healthy chickpea seeds in a manner that maximizes the amount of resources allocated to each offspring under favourable condition. The female preferred seeds having more quantity of resources to meet the nutrition of her offspring, seeds having healthy seed coat over damaged ones, fresh seeds over infested seeds for laying higher number of eggs under normal day light condition over the total dark. This was more so in presence of multiple copulating males over single one. Host deprivation did not have any influence on fecundity.

Los brúquidos se alimentan principalmente de semillas de leguminosas, entre las cuales se encuentran algunos cultivos de importancia económica como el frijol (Phaseolus vulgaris L.), garbanzo (Cicer arietinum L.), lenteja (Lens culinaris Medik) y tamarindo (Tamarindus indica L.) entre otros, considerando al primero como la fuente principal de proteína en la dieta humana; sin embargo, Vigna unguiculata (L.) Walp, por sus propiedades nutritivas podría sustituir al frijol, pero su principal problema es la presencia de plagas, en particular los brúquidos, los cuales ocasionan pérdidas  considerables en postcosecha, por lo que fue necesario conocer los parámetros poblacionales de Acanthoscelides obtectus (Say, 1831), Callosobruchus maculatus (Fabricius, 1775) procedente de dos hospederos diferentes y Zabrotes subfasciatus (Boheman, 1833) alimentados con V. unguiculata por medio de un estudio demográfico bajo condiciones controladas. Los parámetros poblacionales se estimaron con la técnica de tablas de vida de cohorte, la prueba de Log rank (p ≤ 0.05) y las tasas de reproducción e intervalos de confianza con la técnica no paramétrica de Traslape de Intervalos (p ≤ 0.10), para comparar tasas instantáneas de reproducción (rm), llamada también capacidad innata de incremento. Estos parámetros poblacionales fueron mayores en la emergencia de los adultos y tasas de reproducción de C. maculatus en sus dos modalidades por lo que esta especie puede ocasionar serios problemas cuando se presenta asociado con V. unguiculata; sin embargo, A. obtectus y Z. subfasciatus sólo si no tienen opción podríanalimentarse de V. unguiculata en condiciones de almacenamiento.

SDS-PAGE technique was used for the study of seed protein polymorphism among three genotypes of Cicer arietinum with different seed coat colour. A total of 24 polypeptide bands were recorded. Out of these 20 were common among all three genotypes and 4 (16.66%) were polymorphic. The data analysis using UPGMA clustering revealed that genotypes with C2 (dark brown) and C3 (black) were closer as compared to genotype with C1 (light brown) coat colour. Jaccard similarity coefficient value ranged from 0.87 to 0.92. The similarity matrix was subjected to UPGMA clustering to generate dendrogram. The most closely revealed genotypes were C2 (dark brown) and C3 (black) with the highest similarity index 0.92 whereas, C1 (light brown) showed minimum similarity index with C3 (black) genotype 0.87. Each of three genotypes of C.arietinum had some polypeptide bands which were peculiar to them only. This enabled distinguishing all three genotypes on the basis of specific polypeptide fragments using SDS-PAGE analysis.

Chickpea (Cicer arietinum L.) is one of the important plants in terms of nutrition. The yield and quality studies on chickpea, which has the highest production value among edible grain legumes, continue at full stem in our country. When it comes to quality, certain consumer expectations such as protein content, cooking time and seed coat ratio dominate the quality improvement. This study was conducted according to the experimental designs of randomized blocks under the Faculty of Agriculture and Natural Sciences, Uşak University and the laboratory conditions of Faculty of Agriculture, Ondokuz Mayıs University. In this study, a total of 30 chickpea genotypes were used, 13 of them have different varieties. Cooking Time (min), Water Absorption Index (%), Seed Coat Rate (%), Protein Rate (%) and Amylose Rate (%) values of the genotypes have been obtained. The statistically significant differences have been detected between the genotypes in terms of all parameters examined in the variance analysis conducted as a result of this study. Similarly, the result of this study has revealed the correlation between the properties. This study was obtained from a part of the master’s thesis.
