CRISPR/Cas9 is a valuable tool for both basic and applied research that has been widely applied to different plant species. In this chapter, we reviewed the application of CRISPR/Cas9 genome editing toolkit in Brassica crops. We also provided a case study in Brassica napus. Collectively, our results demonstrate that CRISPR/Cas9 is an efficient tool for creating targeted genome modifications at multiple loci in B. napus. These findings open many doors for biotechnological applications in oilseed crops.

Aureobasidium pullulans is a ubiquitous, polyextremotolerant, yeast-like ascomycete used for the industrial production of pullulan and other products and as biocontrol agent in the agriculture. Its application potential and its wide-spread occurrence make A. pullulans an interesting study object. The availability of a fast and efficient genome editing method is an obvious advantage for future basic and applied research on A. pullulans. In this study, we describe the development of a CRISPR/Cas9-based genome editing method using ribonucleoproteins (RNPs). We demonstrate that this method can be used for single and multiplex genome editing using only RNPs by targeting ura3 (encoding for orotidine-5'-phosphate decarboxylase), praics (encoding for phosphoribosyl aminoimidazole-succinocarboxamide synthase) and asl (encoding for arginine succinate lyase). We demonstrate the applicability of Trichoderma reesei pyr4 and Aspergillus fumigatus pyrG to complement the ura3 deficiency. Further, we show that the usage of RNPs can boost the homologous recombination rate up to nearly 100%, even when using only 20bp long homologous flanks. Therefore, the repair cassettes can be constructed by a single PCR, abolishing the need for laborious and time-consuming cloning. The here presented method allows fast and efficient genome editing for gene deletions, modifications, and insertions in A. pullulans.

In this commentary, I share my perspective on the goals of assessments in general, on linking assessments that were developed according to different specifications and for different purposes, and I propose several considerations for the authors and the readers. Nonsuicidal self-injury (NSSI) is direct, intentional harm to one’s own body performed without the intent to die. NSSI has a marked developmental onset reaching peak prevalence in adolescence. NSSI is present in the context of multiple psychological disorders and stands alone as a separate phenomenon. Research has accumulated over the past several decades regarding the course of NSSI. While great advances have been made, there remains a distinct need for basic and applied research in the area of NSSI. This chapter reviews prevalence rates, correlates and risk factors, and leading theories of NSSI. Further, it reviews assessment techniques and provides recommendations. Then, it presents the latest evidence-based treatment recommendations and provides a case example. Finally, cutting edge research and the next frontier of research in this area are outlined.
