Abstract Sanghuangporus baumii is a forest pathogenic fungus and also a medicine mushroom. In the wild, S. baumii parasitize on living host plants and there have been no reports of cultivating fruiting bodies under artificial conditions. In this study, we identified and isolated a S. baumii strain, and successfully cultivated the fruiting bodies on sawdust medium by optimizing culture conditions. The optimum medium, culture temperature and pH for mycelial growth of S. baumii were WBA and YPA, at pH 5.5–7.5 and 28℃, respectively. The contents of total flavonoids, total polysaccharides and total triterpenoids in S. baumii were compared with those in other two medicinal sanghuang, S. vaninii and S. sanghuang. The results showed that both the fruiting bodies and mycelia of S. baumii were rich in bioactive substances. The content of total flavonoids was higher in the fruiting bodies (34.91 mg/g), while the contents of total polysaccharides and total triterpenoids were higher in the mycelia (45.41 mg/g, 14.06 mg/g, respectively). In addition, the mycelia of S. baumii could be elicited to produce more bioactive substances. The use of sealing film in culture increased the polysaccharides content in mycelia by 51.8%, while the light increased the flavonoids content by 151.0%. The cultivation of fruiting bodies and the elicitation of bioactive substances from mycelia provide biological materials for the study and utilization of S. baumii.

AbstractExtracts of the fruiting bodies of the Oyster mushroom (Pleurotus ostreatus) grown on wood substrates (beech, oak, linden, walnut, poplar) and extracts of the fruiting bodies of the Oyster mushroom (Pleurotus pulmonarius) grown in nature on aspen wood were used to determine the total phenols, total flavonoids, lycopene and β-carotene. The content of individual antioxidants varies considerably depending, not only on the substrate, but also on the extracting agents. The highest content of total phenols and total flavonoids was found in methanol and water extracts of the fruiting bodies of the Oyster mushrooms grown on oak and linden substrates. The maximum content of lycopene and β-carotene was determined in acetone and n-hexane (ratio 4 : 6) extracts of the fruiting bodies of the Oyster mushroom grown on an oak block. The results obtained in this study demonstrated that the quantitative and also probably the qualitative composition of the antioxidants in the fruiting bodies of Oyster mushrooms depended considerably on the substrate composition.

Due to their intricate life histories and the unique wing patterns and colouring the butterflies of the genus Chrysoritis are of significant conservation and aesthetic value. Thisoverview probes into practical examples of butterfly life history research applicable to environmental management of this relatively well-known invertebrate group in South Africa. Despite the pioneer work on life histories of Chrysoritis in the past, more should be done to understand the life history of the butterflies in the wild, especially their natural host plants and the behaviour of adults and larvae. A system of voucher specimens of host plants should be introduced in South Africa. Although various host plant species in nature are used by the members of Chrysoritis, including the Chrysoritis chrysaor group, the choice of these in nature by each species is significant for conservation management and in the case of Chrysoritis aureus perhaps even as a specific characteristic.A revision of the ant genus Crematogaster will benefit the conservation management of Chrysoritis species since some of these ant species may consist of a number of specieswith much more restricted distributions than previously thought. Rigorous quantified tudies of population dynamics of Chrysoritis butterflies are absent and the introductionof such studies will benefit conservation management of these localised butterflies extensively.

ABSTRACT: This study evaluated the use of immersion water from peach palm leaves (PPLDA) as a component of the culture medium for the maintenance of Pleurotus spp. and the use of agricultural waste, peach palm leaves, as inoculum support for the fungi. The performance of the inoculum based on peach palm leaves (PPL) for the production of Pleurotus spp. fruiting bodies was compared with that using wheat grains (WG) as inoculum support. PPLDA culture medium (immersion water of peach palm leaves, dextrose, and agar) showed a lower radial velocity of mycelial growth for both fungi than that obtained with the culture medium WDA (wheat extract, dextrose and agar), commonly used as maintenance medium for Pleurotus spp. However, the type of inoculum support does not significantly influence the linear velocity of P. ostreatus mycelial growth, reaching 6.71 mm/day on wheat grains and 6.18 mm/day on peach palm leaves. Thus, when the inoculum based on peach palm leaves is utilized, the immersion water used for preparing this support can be used for preparing the PPLDA maintenance culture medium, diminishing the production costs of Pleurotus mushrooms. Data also showed that when Pleurotus sajor-caju was cultivated on peach palm leaves, using PPL as inoculum support, the fruiting bodies production parameters (Y = 47%, BE = 3% and Pr = 0.2 g/day) did not differ from that obtained using WG.

Macrolepiota procera, commonly called the Parasol Mushroom, is a delicious mushroom collected from the nature and commonly consumed by the public in many regions of Turkey. This study was conducted to determine the optimum culture conditions (pH, temperature, carbon and nitrogen sources) for mycelial growth of M. procera. Three pH values (pH 5.0, 5.5 and 6.0), four incubation temperatures (15, 20, 25 and 30°C), seven carbon (C) sources (dextrose, glucose, lactose, maltose, mannitol, sucrose and xylose) and six nitrogen (N) sources ((NH4)2HPO4, NH4NO3 and Ca(NO3)2, malt extract, peptone and yeast extract) were investigated. In the second step of the study, the effect of seven pH values (4.0, 4.5, 5.0, 5.5, 6.0, 6.5 and 7.0) on the mycelial colony diameter was examined at 20 and 25°C since these temperatures gave the best mycelial growth in the previously conducted temperature experiment. The best mycelial growth was determined at pH 6.0. The optimum temperature for mycelial growth of M. procera was found as 25°C. The use of glucose as carbon source and yeast extract and peptone as nitrogen source in the culture medium gave the best results for mycelial growth. Determining of optimum culture conditions for mycelial growth of M. procera will provide important contributions to the fortcoming studies on it’s commercially cultivation in Turkey.

Thirty six isolates of fungi obtained from anthracnose lesions of cashew and associated host plants in Brazil, were compared by their cultural, morphological and partial sequences of the 28S ribosomal DNA characters. They showed a high degree of cultural variability. The average mycelial growth rate on all tested media ranged from 10.2-13.3 mm/day between the isolates. Most of them produced perithecia (sterile and fertile) and some produced setae (sterile and fertile). All the isolates produced acervuli with predominantly cylindrical conidia (12.4-17.7 µmX 4.8-6.0 µm in width) with round ends, which became septate on germination, and produced unlobed or slightlylobed appressoria. Comparison of the D2 domain of the large subunit (LSU) rDNA sequences with those of other defined species of Colletotrichum and Glomerella grouped 35 of the isolates with known strains of C. gloeosporioides from different hosts (> 98.9% homology). The one exception (LARS 921) was identical to G. cingulata (LARS 238) from Vigna unguiculata.

AbstractFungal naphthoquinones, like red bikaverin, are of interest due to their growing applications in designing pharmaceutical products. Though considerable work has been done on the elucidation of bikaverin biosynthesis pathway in Fusarium fujikuroi, very few reports are available regarding its bioproduction in F. oxysporum. We are hereby proposing a putative metabolic pathway for bikaverin bioproduction in a wild F. oxysporum strain by cross-linking the pigment profiles we obtained under two different fermentation conditions with literature. Naphthoquinone pigments were extracted with a pressurized liquid extraction method, and characterized by HPLC–DAD and UHPLC-HRMS. The results led to the conclusions that the F. oxysporum LCP531 strain was able to produce bikaverin and its various intermediates, e.g., pre-bikaverin, oxo-pre-bikaverin, dinor-bikaverin, me-oxo-pre-bikaverin, and nor-bikaverin, in submerged cultures in various proportions. To our knowledge, this is the first report of the isolation of these five bikaverin intermediates from F. oxysporum cultures, providing us with steady clues for confirming a bikaverin metabolic pathway as well as some of its regulatory patterns in the F. oxysporum LCP531 strain, based on the previously reported model in F. fujikuroi. Interestingly, norbikaverin accumulated along with bikaverin in mycelial cells when the strain grew on simple carbon and nitrogen sources and additional cofactors. Along bikaverin production, we were able to describe the excretion of the toxin beauvericin as main extrolite exclusively in liquid medium containing complex nitrogen and carbon sources, as well as the isolation of ergosterol derivate in mycelial extracts, which have potential for pharmaceutical uses. Therefore, culture conditions were also concluded to trigger some specific biosynthetic route favoring various metabolites of interest. Such observation is of great significance for selective production of pigments and/or prevention of occurrence of others (aka mycotoxins).

Genus Porphyridium is a primitive single-celled red algae widely distributed in seawater, freshwater, and moist soil. It can synthesize bioactive substances such as phycoerythrin, extracellular polysaccharides and polyunsaturated fatty acids during the growth process. In this paper, the culture and bioactive substance yield of Porphyridium purpureum were studied by setting salinity, nitrogen-to-phosphorus ratio, and pH at different gradient levels. The results showed that the optimal conditions for the growth of P. purpureum were salinity 34 ppt, nitrogen-to-phosphorus ratio 169:1, and pH 8; the optimal conditions for obtaining the polysaccharides were salinity 17 ppt, nitrogen-to-phosphorus ratio 14:1, and pH 8; the optimal conditions for obtaining phycoerythrin were salinity 17 ppt, nitrogen-to-phosphorus ratio 68:1, and pH 8; the optimal conditions for obtaining the lipids were salinity 34 ppt, nitrogen-to-phosphorus ratio 1:1, and pH 8. In actual production applications, culture conditions should be set according to different product accumulation purposes in order to achieve the optimal production efficiency.
