Abstract This study aimed to investigate the short and long-term effect of early in life exposure to different feed sources on feed sorting and rumen fermentation of calves. Forty newborn female Holstein calves were randomly divided based on supplementation of concentrate (CON) or hay (HAY) only during the milk-feeding stage (d 1 to 56). After that, all calves were offered a total mixed ration (TMR) containing 43 % of forage and 57 % of concentrate until the end of the experiment (d 57 to 196). Rectal temperature, feed intake, body weight, body structural measurement, fecal and cough score were recorded ranges from day 1 to 196. Fresh and orts feed were sampled daily at the first two weeks (d 57-70) once TMR was offered and at the last week of the experiment (d 190 to 196) for analysis of feed sorting. Rumen fluid samples were collected at ranges from d 14-190 for detecting rumen pH and volatile fatty acids (VFA) concentrations. Our findings showed that early feed exposure did not influence calf health and growth performance but affected feed sorting in a short period. Upon transition to the same mixed diet, differences were found between treatments in the sorting of the different particle fractions. At week 9 and 10, calves early exposed to concentrates began to sort for fine particle fraction, and against the long particle fraction, whereas calves early fed hay sorted for the long particle fraction, and against the fine particle fraction. However, no carryover effect was found, the differences between treatments fade away when detecting feed sorting at the last week, all calves sorted for short and fine particle fractions, and sort against long and medium particle fractions. Although the pattern of feed sorting initially differed between treatments, the rumen pH and VFA concentration were not altered once all calves transitioned to a TMR. These results suggested that the feed familiarity established early in life would not affect diet selection and rumen fermentation in heifer later in life, the dietary experience after transition to same TMR would eventually override the effect of early feed experience.

The criteria used in comparing the utilization of grass silage by reindeer and sheep were rumen pH, ammonia, volatile fatty acids (VFA) and microbes. Rumen samples were taken before feeding, and 2 ½ and 5 ½ hours after the beginning of feeding. Rumen fermentation was lower in the reindeer than in the sheep and differed less between the three sampling times. In the reindeer/the pH of the rumen fluid averaged 6.94 and in the sheep 6.61. The average amounts of NH3—N were 17.0 and 24.2 mg/100 ml rumen fluid and those of total VFA 8.46 and 10.90 mmoles/100ml rumen fluid, respectively. The proportion of acetic acid in the VFA in the reindeer was 75.3 molar % and in the sheep 66.0 molar %, the corresponding values for propionic acid being 18.5 and 22.0 molar % and for butytic acid 4.2 and 8.8 molar %. The number of rumen ciliates in the reindeer averaged 87/mm3 rumen contents and in the sheep 314/ mm3. The numbers of bacteria were 16.0 X 106/mm3, respectively. The proportion of the total microbe mass in the reindeer rumen contents was 1.8 % and in the sheep 2.4 %. The proportions of bacteria in this mass were 87 % and 70 %, respectively. Profiles of rumen fermentation were examined in sheep supplemented with barley or barley plus virginiamycin for 7 weeks and challenged with 1.7 kg of barley with or without virginiamycin. Twenty-five sheep were housed in individual pens and fed chaff, up to a maximum of 1.4 kg/day. Sheep were allocated to one of three treatments: no grain supplement (n = 5), barley grain 700 g twice weekly (n = 10) or barley with virginiamycin (40 g/t grain; n = 10). After 7 weeks, five animals in each treatment group fed grain received 1.7 kg of barley and five received 1.7 kg of barley plus virginiamycin (40 g/t). Sheep supplemented with barley alone maintained a normal pattern of rumen fermentation in response to 1.7 kg of barley indicating adaptation of rumen microbes to grain feeding. In animals supplemented with barley on its own and then fed barley plus virginiamycin there were higher concentrations of L-lactic acid (P < 0.001) and ammonia (P < 0.01) and a lower rumen pH (P < 0.01) than sheep in other treatments at 9, 12 and 24 h following grain feeding. The number of protozoa was decreased (P < 0.001) 24 h following feeding in sheep supplemented with barley and challenged with barley plus virginiamycin compared to pre-feeding (0 h) counts, but not in other treatment groups. Incubations of rumen fluid with glucose from sheep supplemented with barley and barley plus virginiamycin indicated virginiamycin was still effective in reducing L-lactate production following 9 weeks of supplement feeding. The results indicate virginiamycin may disrupt rumen function in sheep already adapted to grain that has not been treated with virginiamycin.

The aim of the study was to determine the effect of different physical forms of starter feed on rumen fermentation indicators of calves after weaning and their weight gain. The experiment was performed with Czech Fleckvieh calves after weaning. The calves were fed ad libitum completely pelleted starter feed or texturized starter feed with chopped straw. The rumen fluid samples were collected after a month of feeding the starter feeds. The calves were weighed monthly. The pH, total acidity, total volatile fatty acids, acetate, propionate, butyrate, lactic acid, ammonia and the number of rumen ciliate protozoa were determined in the rumen fluid samples. The calves receiving the starter feed with straw showed significantly higher rumen pH (6.24 ± 0.51 vs. 5.58 ± 0.30), total volatile fatty acids (98.02 ± 20.46 vs. 61.40 ± 26.51 mmol/l), molar proportion of acetate (61.20 ± 4.87 vs. 50.53 ± 4.66%), and the acetate:propionate ratio (2.38 ± 0.53 vs. 1.34 ± 0.18) and lower molar proportion of propionate (26.55 ± 4.48 vs. 37.92 ± 3.58%) compared with the calves receiving pelleted starter feed. Average daily gain of the calves did not differ significantly. The feeding of starter feed with chopped straw compared with the pelleted starter feed led to better development of the rumen fermentation evaluated by rumen pH, by total volatile fatty acids production, and by the proportion and ratio of acetic and propionic acids. The feeding of starter feed with chopped straw reduced the occurrence of subacute ruminal acidosis in the weaned calves.

AbstractEffects of time of supplementation on forage intake, nutrient apparent digestibility and rumen fermentation were studied with 12 mature castrated male sheep (wethers) offered grass hay from 16.30 h to 09.30 h and supplemented with a cereal-based concentrate given at either 09.30 or 16.00 h. Voluntary intake of hay organic matter (OM) was decreased by feeding the concentrate (P< 0·01). Offering concentrate at 09.30 h after hay was available, increased intake of hay and total OM compared with offering it at 16.00 h before hay was available (P< 0·05). Daily pattern of hay intake was not changed when concentrate was offered at 09.30 h compared with feeding hay alone, but concentrate given at 16.00 h resulted in a lower hay intake between 16.30 and 18.00 h. Sheep offered concentrate at 09.30 h had higher rumen ammonia levels than those offered concentrate at 16.00 h, but there were no differences in the apparent digestibility of the nutrients, rumen pH and molar proportions of the main volatile fatty acids. There were also no differences between groups in the blood plasma concentrations of 3-hydroxybutyrate (30HB), nonesterified fatty acids (NEFA) and glucose. It is concluded that feeding supplement after rather than before a period of intake of forage or a bout of grazing may offer a means of minimizing reduction of forage intake as a consequence of feeding concentrate.

Feeding goats with calcium salts of fatty acids (CSFA) can supply ruminants with lipids, with minimal effects on ruminal fermentation and fiber digestibility. However, there is a shortage of information on the effect of CSFA on characteristics of rumen fermentation in grassland goats. Thus, the present study aimed to assess the addition of CSFA to concentrate on the parameters of rumen fermentation of grazing goats. Five rumen cannulated goats were distributed in a Latin square 5x5 design (treatments: 0%, 1.5%, 3.0%, 4.5% and 6.0% CSFA. The pH, ammonia N and volatile fatty acids (VFA) content were analyzed in the ruminal fluid at 0, 2, 4, 6 and 8 hours after concentrate supplementation. The pH and ammonia N concentration showed a linear effect with the addition of CSFA. There was no effect observed for the VFA molar concentration after grazing goats were fed with the experimental diet. In conclusion, further research is needed to investigate the addition of CSFA to goat diets because there is evidence that CSFA increases ruminal pH and decreases excess ruminal ammonia without changing the VFA concentration in the rumen fluid.

In a crossover trial, 4 cows were given concentrates containing rapidly and slowly degradable protein in combination with prewilted grass silage. Diets were given in 2 equal daily portions according to DCP and net energy requirements. The trial consisted of 3 main periods of 3 weeks each, with faeces, urine and milk collected and measured during the final week. On 1 day during this week samples of rumen fluid were taken and on 2 days blood samples were taken, directly before the morning feed and 1, 2, 3, 4 and 8 h later. N digestibity and N retention were the same on both rations. A concentrate with slowly-degradable protein resulted in a significantly higher rumen pH and acetate:propionate ratio. Ammonia concn. tended to be lower. Slowly-degradable concentrate protein led to a significantly higher concn. of urea and a lower concn. of insulin in blood. The concn. of several of the essential amino acids, and of the glucogenic amino acids and glycine was significantly lower. There were no significant effects of type of protein on milk yield or milk composition, but there was a tendency for these parameters to increase with ration containing slowly-degradable protein. Results suggest the ration containing slowly-degradable concentrate protein produced a more even rumen fermentation, which promoted a higher acetate:propionate ration in the rumen. This study evaluated the effects of intraruminal administration ofMegasphaera elsdeniion ruminal fermentation patterns, the profile of plasma metabolites, and milk yield and composition of mid-lactation dairy cows. Eight primiparous, ruminally cannulated Holstein cows were arranged in a paired 2×2 crossover design. Cows were randomly assigned to one of two treatments: 1) intraruminal inoculation of 35 ml suspension per day ofM. elsdeniiATCC 25940 (MEGA), containing 108cfu/ml of bacteria, dissolved in 35 ml of saline (0·15m), or 2) carrier alone (35 ml saline; CTR). Both postprandial and preprandial rumen volatile fatty acids (VFA) and plasma metabolite measurements were analysed. Postprandial VFA patterns were affected the most, with butyrate (P<0·01) and valerate (P<0·01) proportions increasing, and acetate (P<0·01), isobutyrate (P=0·05) and isovalerate (P<0·01) decreasing in MEGA cows. Preprandial data measured at various days showed that MEGA dosage tended to increase the molar proportion of propionate (P=0·09) and lower the acetate to propionate ratio (P=0·07) in the rumen fluid. There was no effect of treatment on rumen pH and on the concentration of lactate in the rumen as well as on selected preprandial plasma metabolites. Postprandial plasma concentrations of cholesterol tended to increase (P=0·07) in MEGA cows compared with CTR. Concentrations of non-esterified fatty acids (NEFA) in the plasma were lower in MEGA cows after the morning feeding (P<0·01). Sampling hour also affected plasma NEFA in this study. Plasma β-hydroxybutyrate (BHBA) were not affected by the treatment (P>0·05); however, after the morning feeding BHBA concentration was increased in both groups of cows. Dry matter intake and milk yield and composition were not affected by treatment. In conclusion, results indicate thatM. elsdeniihas the potential to modulate the rumen fermentation profile in mid-lactation Holstein cows, but these effects were only slightly reflected in changes in plasma metabolites and milk composition.

The effects of adding essential oils (EO) at different levels (125, 250, 500 mg/l) on rumen fermentation and biohydrogenation were examined in a rumen batch culture study. Treatments were: control without EO (CON), control with anise oil (ANO), cedar wood oil (CWO), cinnamon oil (CNO), eucalyptus oil (EUO), and tea tree oil (TEO). Essential oils, each dissolved in 1 ml of ethanol, were added to the culture flask containing 40 ml of buffer solution, 2 ml of reduction solution, 10 ml of rumen fluid, 25 mg of soybean oil, and 0.5 g of the diet. After 24 h of incubation in a water batch at 39&deg;C, three samples were collected from each flask and analyzed for ammonia-N, volatile fatty acids (VFA), and fatty acids (FA). Expect for CNO, the proportions of acetate, propionate, and acetate to propionate ratios were not affected (P &gt; 0.05) by EO addition. Addition of CWO, CNO, and TEO reduced total VFA concentrations (P &lt; 0.05) regardless of dose level. The ammonia-N concentration was greater in cultures incubated with EO regardless of dose level. Compared with the CON, the concentrations of C18:0 and trans C18:1 were reduced (P &lt; 0.05) with EO addition regardless of dose level. Compared with the CON, the concentration of linoleic acid was greater (P &lt; 0.05) when EO were added at 500&nbsp;mg/l. EO tested in this study had no effects on VFA profile but significantly reduced the formation of biohydrogenation products (C18:0 and trans C18:1).

In this work, 13 jet nebulizers, some of which in different configurations, were investigated in order to identify the biopharmaceutical constraints related to the quality attributes of the medicinal products, which affect their safety, efficiency, compliance, and effectiveness. The aerosolization parameters, including the aerosol output, aerosol output rate, mass median aerodynamic diameter, and fine particle fraction, were determined according to the European Standard EN 13544-1, using sodium fluoride as a reference formulation. A comparison between the aerosol output nebulization time and the fine particle fraction displayed a correlation between the aerosol quality and the nebulization rate. Indeed, the quality of the nebulization significantly increased when the rate of aerosol emission was reduced. Moreover, the performance of the nebulizers was analyzed in terms of respirable delivered dose and respirable dose delivery rate, which characterize nebulization as the rate and amount of respirable product that could be deposited into the lungs. Depending on which of these two latter parameters was used, the nebulizers showed different performances. The differences, in terms of the rate and amount of delivered aerosol, could provide relevant information for the appropriate choice of nebulizer as a function of drug product, therapy, and patient characteristics.
