Quality Assessment and Origin Identification on Four Species of Curcumae Radix by HPLC Assay and Fingerprint Analysis

Abstract BackgroundCurcumae Radix is a multi-source Chinese medicine called Yujin. It is derived from the root tubers of four Curcuma species including Curcuma wenyujin , C. kwangsiensis , C. phaeocaulis and C. longa. The identification of the root tubers of the four Curcuma species often caused confusions. MethodsIn this study, we developed HPLC/PDA methods to differentiate these four species of Curcumae Radix by HPLC assay and fingerprint analysis. The methods developed were also validated with their precision, repeatability and accuracy. ResultsCurdione could only be detected in C. wenyujin . For C. kwangsiensis , C. phaeocaulis , if the relative peak area value of peak 1 and germacrone peak of the samples was more than 1.024, it could be identified as C. phaeocaulis of Curcumae Radix. Curcumin, desmethoxycurcumin and bisdesmethoxycurcumin could only be detected in C. longa . We also developed a simple method to determine curcumin, desmethoxycurcumin and bisdesmethoxycurcumin in C. lon ga and germacrone in the other three species of Curcumae Radix. ConclusionsBecause of the definite difference of main ingredients between curcumin-free group (C. wenyujin , C. kwangsiensis and C. phaeocaulis ) and curcumin containing C. longa from this study, four Curcuma species of Curcumae Radix in the pharmacopeia should be separated into two monographs, with curcumin-free and germacrone containing species as one and curcumin containing C. longa as the second monograph. The HPLC/PDA methods of HPLC assay and fingerprint analysis developed in this study could be used for the quality control of these four species of Curcumae Radix. A simple and efficient fingerprinting method for chrysanthemum buds was developed with the aim of establishing a quality control protocol based on biochemical makeup. Chrysanthemum bud samples were successively extracted by water and alcohol. The fingerprints of the chrysanthemum buds samples were obtained using capillary electrophoresis and electrochemical detection (CE-ED) employing copper and carbon working electrodes to capture all of the chemical information. 10 batches of chrysanthemum buds were collected from different regions and various factories to establish the baseline fingerprint. The experimental data of 10 batches electropherogram buds by CE were analyzed by correlation coefficient and the included angle cosine methods. A standard chrysanthemum bud fingerprint including 24 common peaks was established, 12 from each electrode, which was successfully applied to identify and distinguish between chrysanthemum buds from 2 other chrysanthemum species. These results demonstrate that fingerprint analysis can be used as an important criterion for chrysanthemum buds quality control.

Tulisan ini membahas fenomena penyebaran kitab-kitab cetak keaga­maan di Jawa Barat, khususnya di Sukabumi dan Cianjur. Jenis kitab ini biasanya menggunakan tulisan Arab dengan bahasa Sunda dan menggu­nakan aksara Pegon. Kitab-kitab cetak dari jenis-jenis itu diproduksi dan direproduksi, dan masih digunakan sampai hari ini. Oleh karena itu, fenomena ini dapat disebut sebagai “living tradition”. Pengumpulan data dilakukan dengan melacak dan merekam kitab-kitab yang diproduksi (disalin atau dikarang) dan direproduksi (dicetak atau digandakan) dengan metode seder­hana, yakni fotocopi dan pencetakan tradisional seperti stensil, sablon, dan "cetak toko". Tulisan ini bertujuan untuk memetakan dan merevisi kategorisasi kitab-kitab tersebut dari kajian terdahulu berdasarkan jenis karya, seperti karangan asli, tuqilan, terjemahan, syarḥ (penjelasan), khulasah (ringkasan) yang lain, dan juga berdasarkan bahasa dan aksara yang digunakan. Selain itu, tulisan ini juga mengamati lembaga-lembaga atau individu-individu yang masih mereproduksi buku-buku (kitab-kitab) keagamaan sederhana seperti perusahaan percetakan atau pesantren yang menerbitkan kitab-kitab tersebut. Pada akhir artikel ini, ada beberapa saran dalam upaya untuk melestarikan kitab-kitab cetak dan karya-karya tersebut.Kata kunci: kitab, jenis karya, pencetakan tradisional, Sunda, Pegon, Jawa Barat This paper discusses the phenomenon of the spreading of religious printed books (kitabs) in West Java, especially in Sukabumi and Cianjur, which are characterized by the use of Arabic writing in Sundanese (or known as Pegon script). The printed books (Kitabs) of those types are produced and reproduced, and are still used to this day. Therefore, this phenomenon can be called as a “living tradition”. Data collection was conducted by tracing and recording religious books (kitabs) that are pro-duced (rewritten or compossed) and reproduced (printed or duplicated) with a simple method, known as photocopying and traditional printing such as stencils, screen printing, and “shop printing”. This paper aims to map and revise the categorization of these Kitabs from previous studies based on the types of works including original essays, tuqilan (quotations), translation, sharh (explanation), khulaṣah (summary), or the other, and also based on the language and the script used. In addition, this paper also observes the institutions or individuals that are still reproducing these printed religious books such as the printing company or pesantren that publish such kitabs. At the end of this article, there are some suggestions in attempts to preserve those printed kitabs and the works.Keywords: Kitabs, type of work, traditional printing, Sunda, Pegon, West Java

Background: Zishen Tongguan (ZSTG) capsules were prepared at the Affiliated Hospital of Nanjing University of Chinese Medicine and have been proven to be clinically effective for treating pyelonephritis and benign prostatic hyperplasia. However, the quality standards are not ideal; a comprehensive study of the “quality markers” (Q-markers), the chemicals inherent in traditional Chinese medicine and its preparations, has not been carried out. Experimental Methods: In this paper, a sensitive and specific ultra-high-performance liquid chromatographictandem mass spectrometry (UHPLC-MS/MS) method was developed for the simultaneous determination of eight potential Q-markers of ZSTG, including timosaponin A3, berberine, jatrorrhizine, phellodendrine, palmatine, mangiferin, neomangiferin, and timosaponin BII. A Kromasil 100-3.5 C18 column was used with a mobile phase of 0.2% formic acid with acetonitrile, and gradient elution at a flow rate of 0.2 mL/min was achieved in 13 minutes and used for separation. Detection was performed in positive/negative mode with multiple reaction monitoring (MRM). Results: The analytical method was validated in terms of the sensitivity, linearity, accuracy, precision, repeatability, stability and recovery. The method established here was successfully applied to study the potential Q-markers in 8 batches of commercial samples, which demonstrated its use in improving the quality control of ZSTG. Conclusion: The developed method had high repeatability and accuracy and was suitable for the simultaneous analysis of multiple Q-markers, which may provide a new basis for the comprehensive assessment and overall quality control of ZSTG.

Background and Objective: Kang Fu Xin liquid (KFX) is an official preparation made from the ethanol extract product from P. Americana. The present quality control method cannot control the quality of the preparation well. The aim of the present study is to establish a convenient HPLC method for multicomponents determination combined with fingerprint analysis for quality control of KFX. Methods: An HPLC-DAD method with gradient elution and detective wavelength switching program was developed to establish HPLC fingerprints of KFX, and 38 batches of KFX were compared and evaluated by similarity analysis (SA), hierarchical clustering analysis (HCA), and principal component analysis (PCA). Meanwhile, six nucleosides and three amino acids, including uracil, hypoxanthine, uric acid, adenosine, xanthine, inosine, tyrosine, phenylalanine and tryptophan in KFX were determined based on the HPLC fingerprints. Results: An HPLC method assisted with gradient elution and wavelength switching program was established and validated for multicomponents determination combined with fingerprint analysis of KFX. The results demonstrated that the similarity values of the KFX samples were more than 0.845. PCA indicated that peaks 4 (hypoxanthine), 7 (xanthine), 9 (tyrosine), 11, 13 and 17 might be the characteristic contributed components. The nine constituents in KFX, uracil, hypoxanthine, uric acid, adenosine, xanthine, inosine, tyrosine, phenylalanine and tryptophan, showed good regression (R2 > 0.9997) within test ranges and the recoveries of the method for all analytes were in the range from 96.74 to 104.24%. The limits of detections and quantifications for nine constituents in DAD were less than 0.22 and 0.43 μg•mL-1, respectively. Conclusion: The qualitative analysis of chemical fingerprints and the quantitative analysis of multiple indicators provide a powerful and rational way to control the KFX quality for pharmaceutical companies.

A technique for the determination of tannin content in traditional Chinese medicine injections (TCMI) was developed based on ultraviolet (UV) spectroscopy. Chemometrics were used to construct a mathematical model of absorption spectrum and tannin reference content of Danshen and Guanxinning injections, and the model was verified and applied. The results showed that the established UV-based spectral partial least squares regression (PLS) tannin content model performed well with a correlation coefficient ([Formula: see text]) of 0.952, root mean square error of calibration (RMSEC) of 0.476[Formula: see text][Formula: see text]g/ml, root mean square error of validation (RMSEV) of 1.171[Formula: see text][Formula: see text]g/ml, and root mean square error of prediction (RMSEP) of 0.465[Formula: see text][Formula: see text]g/ml. Pattern recognition models using linear discriminant analysis (LDA) and [Formula: see text] nearest neighbor ([Formula: see text]-NN) classifiers based on UV spectrum could successfully classify different types of injections and different manufacturers. The established method to measure tannin content based on UV spectroscopy is simple, rapid and reliable and provides technical support for quality control of tannin in Chinese medicine injections.
