In order to characterize the self-incompatibility system in Theobroma cacao, the levels of ethylene, indole-3-acetic acid (IAA), and abscisic acid (ABA) were determined after pollination with compatible and incompatible pollen and in unpollinated flowers. Pollen tube growth rates after incompatible and compatible pollinations were identical, and the majority of the pollen tubes reached the ovules between 12 and 20 hours after pollination. ABA levels rose in incompatibly pollinated flowers, and fell in compatibly pollinated flowers, prior to pollen tube—ovule contact. Ethylene evolution remained stable in compatibly pollinated flowers and rose in incompatibly pollinated flowers. IAA concentrations increased in compatibly pollinated flowers, and remained stable in incompatibly pollinated flowers after pollination and subsequent to pollen tube—ovule contact.

Examination of the behaviour of pollen on the style of Raphanus , following compatible and incompatible intraspecific pollinations, has revealed the self-incompatibility system in this species to be composed of at least three stages. The first, on which no information has been obtained in this study, involves the germination of the grain. The second stage concerns the ability of the pollen tube to penetrate the cuticle of the stigmatic papilla. It is possible that cutinase is deficient in incompatible pollen tubes but, in most instances, the outer layers of the stigmatic wall are penetrated. The third stage involves the interaction of substances secreted by the pollen tube with products of the stigmatic cytoplasm. The interaction is swiftly followed by the deposition, in the stigma, of a layered callosic body. This is formed immediately under the point of penetration and takes about 6 h to develop fully. Development of the pollen tube ceases as the first layers of callose are laid down. It is possible that the substances in the pollen responsible for the initiation of the second two stages are held in the tapetally synthesized tryphine, thus accounting for the sporophytic control of pollen compatibility in this species. The mature stigma contains large numbers of crystalline protein bodies, but it is not known whether they play any role in the self-incompatibility system.

The route of 'Orlando' tangelo (Citrus paradisi Macf. × C. reticulata Blanco) pollen tubes was traced and compared in self-incompatible pollinations and cross-compatible pollinations with 'Dancy' tangerine (C. reticulata Blanco). In both crosses, 'Orlando' pollen germinated in the stigmatic exudate and grew between the papillae on the stigma surface and inter-cellularly between the parenchymatous cells until reaching a stylar canal by 3 days. However, in the incompatible pollination, irregular deposition of callose occurred in the pollen tube walls as early as 1 day after pollination. By day 6, pollen tubes were in the upper portion of the ovary in the compatible pollination, whereas most pollen tubes from the incompatible pollination were still in the upper style. 'Orlando' pollen tube growth rate decreased substantially by day 3 in both the self-incompatible pollination and in vitro but increased rapidly after day 3 in the compatible combination. The generative cell divided between 1 and 3 days after pollination in the compatible cross. Generative cell division was observed by day 3 in only a few pollen tubes in the incompatible cross and in cultured tubes. Compatible pollen tubes grew slowly for the first 3 days after pollination, during which time generative cells divided and then grew rapidly until fertilization. In contrast, incompatible pollen tubes showed morphological features indicative of an incompatibility reaction by 1 day after pollination and grew slowly for a period of 6 days, and then ceased growth.

Pollen tube development in Orlando tangelo (Citrus paradisi Macf. × C. reticulata Blanco.) was compared within and between cross-compatible pollinations of Orlando pollen on Dancy tangerine (C. reticulata Blanco.) stigmas and self-incompatible pollinations on Orlando tangelo stigmas. Orlando and Dancy gynoecia were morphologically similar but differed slightly in stigma, style, and ovary lengths. Orlando pollen tube development was studied 1, 3, 6, 9, and 12 days after both cross- and self-pollination to record the number of pollen tubes at each of five levels: stigma surface, upper style, lower style, ovary, and entrance into ovules. In the incompatible cross (self-pollinated Orlando), the stigma was the primary region of pollen tube arrest. In the compatible cross (Orlando pollen on Dancy), some pollen tubes penetrated ovules between 9 and 12 days after cross pollination; however, other pollen tubes were arrested in the stigma. Pollen tubes that successfully penetrated ovules in the compatible cross differed morphologically from pollen tubes arrested in both the compatible and incompatible situations. Successful compatible pollen tubes were straight with thin-walled tips and regularly spaced callose plugs behind the growing tips. Many pollen tube abnormalities associated with the self-incompatible pollination of Orlando were also present among arrested pollen tubes from the compatible cross.

In many species of Paspalum, diploid (2x) cytotypes are usually outbreeders due to a self incompatibility system, while tetraploid (4x) conspecific counterparts are pseudogamous self-fertile apomicts. Intraspecific crossability between 2x female and 4x male cytotypes was investigated using classical methods of crossing in P. almum, P. brunneum, P. rufum, P. intermedium and P. quadrifarium. Expected triploid BII hybrids were obtained in P. intermedium (crossability: 0.004%) and in P. brunneum (crossability: 0.015%). In P. rufum, only tetraploid BIII hybrids were achieved with a crossability of 0.025% Looking for a better performance, in vitro ovary rescue was attempted 5 days after pollination in intraspecific 2x × 4x crosses of P. almum, P. quadrifarium, P. intermedium and P. rufum. The method was useful to recover some triploid BII hybrids in P. almum (success rate: 0.49%) and in P. rufum (0.59%), but failed in P. intermedium and P. quadrifarium. Pollen tube growth was usually inhibited at the stigma or style levels after self-pollination in 2x plants, confirming that diploids are self-incompatible. Pollen of 4x cytotypes germinated and penetrated the pistils of diploid conspecific cytotypes. This indicated that no incompatibility system exists in these species to keep the 2x cytotypes from hybridising with 4x conspecific cytotypes as pollen donors. However, after pollination of 2x cytotypes with pollen of 4x counterparts, most ovules showed embryo and endosperm development, but a few days later, endosperms collapsed and embryos stopped their development. As a result of these processes, sexual self-incompatible 2x cytotypes of Paspalum species produced very few triploid hybrids when pollinated with pollen of their apomictic 4x counterparts. Thus, the low diploid-tetraploid crossability was due to the existence of a post-zygotic abortive system and not caused by pre-fertilisation barriers.

Petunia has a monofactorial gametophytic self-incompatibility system which is characterized by a mechanism for pollen-tube rejection after incompatible pollination about half-way between stigma and ovule. From in vitro experiments it is known that pollen tube extension requires de novo synthesis of proteins on 80 S ribosomes. The synthesis of these structural proteins has no need of new mRNA formation. The situation in the style is quite different: both RNA and protein synthesis are positively affected by pollination. There is a qualitative difference in synthesis between self- and cross-pollinated styles. These findings suggest that inhibition or rejection of pollen tubes is ‘the normal reaction’, whereas unaffected penetration is based on the synthesis of special enzymes after compatible pollination which break down the existing incompatibility barrier.

Objetivo: Establecer una línea base como fundamento para la intervención e inducción de innovaciones a través de escuelas de campo y planes de negocios. Diseño/metodología/aproximación: El estudio se desarrolló en San José del Carmen, San Cristóbal de Las Casas, Chiapas, México. Se elaboró una encuesta como instrumento para recabar en campo información de familias de la localidad con la finalidad de conocer su situación actual. La localidad de trabajo se eligió utilizando como criterio las poblaciones que se encuentran dentro de Cruzada Nacional contra el hambre y en la clasificación de la pobreza extrema Resultados: La localidad de San José del Carmen es considerada como de alta marginación, la población es bilingüe, su principal lengua es el Tzotzil, el nivel de estudios promedio es hasta el tercer año de primaria, las principales actividades productivas son la siembra de maíz (Zea mays L.), frijol (Phaseolus vulgaris L.), papa (Solanum tuberosum L.) y haba (Vicia faba L.) que son utilizadas principalmente para autoconsumo.  No cuentan con agua potable ya que su sistema es a través de la recaudación de agua de lluvia o a través de un jagüey. Sus principales actividades económicas son la albañilería por parte de los hombres, y venta de productos, tales como el pozol (bebida energética a base de maíz y cacao (Theobroma cacao L.), tostadas y algunos vegetales por parte de las mujeres. Las tostadas son producidas en forma artesanal con un proceso de doble nixtamalización y vendida en bolsas de 20 tostadas cada una, a un costo de MX$10.00 en los mercados de San Cristóbal de las Casas, una a dos veces por semana alrededor de 40 bolsas por día de venta. Limitaciones del estudio/implicaciones: La afiliación zapatista de alguno de los pobladores y el hecho de que algunos habitantes no hablen español, fueron factores que limitaron desarrollar al 100% el estudio; sin embargo, se considera que se tienen los elementos necesarios para establecer la línea base de la situación de San José del Carmen. Conclusiones: El diagnóstico servirá como una herramienta para establecer una intervención a través de innovaciones y planes de negocios en la comunidad de estudio. Los pobladores cuentan con herramientas necesarias para obtener nuevos conocimientos que ayuden a un desarrollo de su producción. El idioma no deberá de ser una limitante para ello.
