Effect of Endogenous Expression of the Laminin α5 Chain on Chemotherapy Resistance of Colorectal Cancer Cells

The expression of the LAMA5 gene encoding the laminin α5 chain has been inhibited in the line HT29 cells. The treatment with 5-fluorouracil (5FU) and regorafenib (RF) showed that the knockdown of LAMA5 increases the sensitivity of the HT29 cells to 5FU by 2.2 times and does not affect their sensitivity to RF. The data of transcriptome analysis indicate that the observed changes in the sensitivity to 5FU may be caused by an activation of signaling pathways of the endoplasmic reticulum stress in the HT29 cells. In the case of RF, this effect can be compensated by a 2-fold decrease in the expression of the main RF transporter in HT29 cells. apoptosis, colorectal cancer, laminins, drag resistance, knockdown, regorafenib, endoplasmic reticulum stress, 5-fluorouracil, laminin α5 chain, endogenous expression, HT29, LAMA5, UPR.
