To improve reproductive performance in cattle, the accurate detection of estrus and optimization of insemination relative to ovulation are necessary. However, poor heat detection by farm staff leads to a decreased conception rate, thus inflicting economic damage to the beef and dairy industries. This study aimed to develop monoclonal antibodies (mAb) that can specifically bind to the bovine lactoferrin (bLF) protein, which we have previously demonstrated to be overexpressed in bovine cervical mucus during estrus. Female rats were intraperitoneally immunized with bLF protein as the antigen. Anti-bLF mAbs were then purified by affinity chromatography, and their binding affinity for the bLF antigen was examined using ELISA. We found a high binding affinity between mAbs and bLF. Finally, we developed a rapid bovine heat detection kit using the anti-bLF mAbs that we generated and tested on cervical mucus from 12 cows (estrous synchronization, n = 2; natural cycling, n = 10). We found that the kits accurately detected estrus. Overall, our fabricated heat detection kit based on rat anti-bLF mAbs could pave the way for the development of potent tools for heat detection devices for dairy cattle, thereby preventing economic loss.

The objective of the present study was to characterise follicular dynamics in Santa Ines (SI) during a 9-day protocol for oestrous synchronization and to evaluate the efficiency of the vaginal progesterone-releasing device Primer-PR® (Tecnopec, Brazil). Cyclic females were used (n = 10) at the ruminant sector of the veterinary hospital of the FMVZ–USP, in March 2009. The animals had body scores between 2.5 and 3 and were of proven fertility. Ultrasound examinations were performed using an ALOKA SSD-500 Scanner (Berger, Brazil) with a linear 5.0-MHz transducer, attached to a handle to allow safe intrarectal manipulation. Examinations were performed daily from 3 days (D-3) before the day of device placement (D0) until the day of device removal (D9), and twice daily from device removal until ovulation. All ewes received 0.03 mg of prostaglandin (D-cloprostenol, Prolise®, Tecnopec, Brazil) on D9. Ovulation was assessed by the disappearance of the growing larger follicle or follicles present in the previous examinations. Oestrous detection was performed using an intact male 3 times a day (at 10:00 a.m., 6:00 p.m., and 12:00 p.m.) from D9 until the last acceptant ewe, and blood samples were taken by jugular puncture for progesterone (P4) measurement by radioimmunoassay on solid phase (COAT-A-COUNT, Siemens, USA) from D-3 until the day of ovulation. Data were analysed by Shapiro–Wilk (PROC UNIVARIATE) using the SAS program (SAS Institute Inc., Cary, NC, USA, 2001) and shown as mean ± standard deviation. Plasma P4 concentrations between D0 and D9 were 6.56 ± 2.32 ng mL–1, peaking between D3 and D5 (8.07 ± 2.31 ng mL–1). Oestrous behaviour was shown 45.6 ± 12.71 h after Primer-PR® removal. The first and the last ewe that showed oestrous behaviour was at 30 h and 66 h after Primer-PR® removal, respectively, and the majority of ewes (50%) at 42 h. Oestrous lasted 26.40 ± 9.47 h, and the majority of ewes (70%) showed oestrous behaviour during 24 h. Ovulation occurred 73 ± 14.38 h after Primer–PR® removal and 1.3 ± 0.48 ovulations per animal were observed. From all growing presumptively dominant follicles observed, 92.3% of them ovulated. When double ovulations occurred (n = 3), the interval between first and second ovulation was 16 ± 6.93 h. Emergence of the ovulatory follicular wave occurred at 8.5 days + 16 h after Primer–PR® insertion. The follicles observed to continue growing had a diameter of 3.48 ± 0.28 mm when they were first detected and reached 5.63 ± 0.66 mm, with a growth rate of 0.73 ± 0.43 mm per day. A standard follicle wave within the 9-day protocol was not possible to determine, and the follicles receded in up to 4 days. In conclusion, placement of a Primer-PR® device for 9 days resulted in synchronous oestrus and ovulation in Santa Inês ewes. FAPESP, CAPES, Tecnopec.

The study of ovarian follicular dynamics (OFD) and other changes in the bovine reproductive tract has developedsignificantly over the past two decades, primarily due to the use of non-invasive investigative approaches suchas ultrasound. Advances in ultrasound examinations have provided a better understanding of ovarian activity,uterine involution, oviducts status, and other segments of the reproductive tract in the bovine postpartumperiod. Procedures including tracking of the reproductive tract have likewise aided in the development of newmethodologies and techniques to improve reproductive performance in cattle. The aim of this review was tosummarize knowledge regarding the reproductive tract in dairy cows during the postpartum period.

The objective of this experiment was to evaluate the role of a short-term increase in linolenic acid around the timed artificial insemination (TAI) on plasma fatty acid concentrations, estrous behavior, and pregnancy. Therefore, 96 cross-bred heifers (Initial BW=394±18.1 kg) fed chopped grass hay were allotted to one of three dietary treatments: no supplement (CON); 1.8 kg head−1 d−1 (DM basis) of a soybean-based pellet (SOY) high in linoleic acid (C18:2n-6); or 1.5 kg head−1 d−1 (DM basis) of a flaxseed-based pellet (FLX) high in linolenic acid (C18:3n-3). On day −9, relative to TAI, supplementation began, and heifers received a controlled internal drug-releasing (CIDR) device and gonadotropin releasing hormone (GnRH; 100 µg). On day −2, CIDRs were removed and heifers received 25 mg prostaglandin F2α (PGF2α). Heifers were bred by TAI (day 0) 54 h after CIDR removal and received GnRH. Estrous behavior was monitored throughout the feeding period by radiotelemetry. Supplementation ended on day 18 relative to TAI. Plasma concentrations of C18:2n-6 were greater by day 0 and day 18 (P<0.001) above that of CON for both SOY and FLX with SOY being greater than (P<0.001) FLX. Likewise, C18:3n-3 was also greater for supplemented heifers by day 0 and day 18 (P<0.001). The number of heifers that did not respond to estrous synchronization was not affected by dietary treatment (P=0.61). Conception to AI was not affected by treatment (P=0.24). Overall pregnancy rates tended to be lower for FLX (P=0.08) than SOY. Conception to a TAI program was not enhanced when oilseeds were fed to beef heifers during estrous synchronization and continuing past day 18 relative to artificial insemination.

Abstract This study investigated growth and reproductive performance of Bos indicus-influenced beef heifers provided a concentrate supplement at either a constant or stair-step supplementation. Sixty-four Brangus heifers were stratified by initial BW and age (255 ± 20 kg; 270 ± 22 d) and assigned to 1 of 16 bahiagrass pastures (4 heifers/pasture). Treatments were randomly assigned to pastures (8 pastures/treatment) and consisted of concentrate DM supplementation at: (1) 1.50% of BW from d 0 to 100 (CON) and (2) 1.05% of BW from d 0 to 49 and 1.95% of BW from d 50 to 100 (STP). Supplementation started on d 0 and continued until start of estrous synchronization on d 100. Intravaginal temperatures (16 heifers/treatment) were collected every 30 minutes from d 25 to 31 (September) and d 85 to 91 (November). All heifers were provided supplement at 1.50% of BW from d 101 to 211. Heifers were bred by AI from d 113 to 115 and Brangus bulls were placed with heifers from d 121 to 211. Data were analyzed using the MIXED procedure of SAS with treatment, time, and interaction as fixed effects. From d 0 to 49, ADG did not differ (P = 0.87); however, STP heifers had greater (P = 0.01; 0.73 vs. 0.56 kg/d) ADG from d 50 to 100 compared to CON heifers. Percentage of pubertal heifers, percentage of heifers in estrus, or reproductive tract scores did not differ between treatments (P ≥ 0.40). Intravaginal temperatures were greater (P &lt; 0.0001) for CON vs. STP heifers from d 25 to 31 but did not differ (P = 0.74) from d 85 to 91. Using a stair-step vs. constant supplementation amount (1.05 to 1.95% vs. 1.50% of BW) before the breeding season reduced intravaginal temperatures and increased overall ADG in Bos indicus-influenced beef heifers.
