The corrosion behavior of GW63 (Mg-6wt.%Gd-3wt.%Y-0.4wt.%Zr) alloys in 5% NaCl aqueous solution has been investigated by PARSTAT 2273 instrument. The Open Circuit Potential (ECORR) vs. time curve, cyclic polarization (Pitting Scans) curve and Electrochemical Impedance Spectroscopy (EIS) was measured for the GW63 alloys in as-cast and T6 heat treatment conditions. The EIS results indicated that the tendency of impedance variation for as-cast condition was monotonic decreasing, however, the tendency of variation for T6 condition was not completely monotonic but the total tendency was decreasing. The values of impedance of GW63 alloy at 0.1 Hz are about 103 ohm-cm2 for as-cast and T6 condition.

The purpose of the present study was to determine whether initial measures of impulse (i.e., area under the force-time curve) could be used to predict peak force (PF) in hand grip responses of 5- and 10-kg amplitude executed as rapidly as possible. 12 subjects performed 75 practice and 25 test contractions in each condition, with and without visual feedback. The time to peak (PT) for the 5-kg responses was 41 msec., while the 10-kg condition averaged 56 msec. Analyses demonstrated no effect for visual feedback. Correlational analyses of cumulative impulse sections from 5 msec., to PF within conditions showed that early measures of force within a response were not very good predictors of final force amplitude. Indeed, for both conditions upwards of 85% of a reponse needed to have occurred before 50% of PF variance could be predicted. Analyses across conditions demonstrated that 50% of PF variance could be predicted between 15 and 20 msec. PT was also an important predictor variable. By using PT along with cumulative impulse 50% of the variance in PF could be predicted prior to 10 msec., at 5 kg (25% of PT) and at about 15 msec., for 10 kg (27% of PT). Across conditions, 85% of the variance in PF was predictable by 5 msec. Such results suggest that either more refined response indices are needed in predicting response outcome or that even for very rapid responses some lower level “tuning” probably occurs after initiation.

Missense mutations in alpha-tropomyosin (TPM1) can lead to development of hypertrophic (HCM) or dilated cardiomyopathy (DCM). HCM mutation E62Q and DCM mutation E54K have previously been studied extensively in experimental systems ranging from in vitro biochemical assays to animal models, although some conflicting results have been found. We undertook a detailed multi-scale assessment of these mutants that included atomistic simulations, regulated in vitro motility (IVM) assays, and finally physiologically relevant human engineered heart tissues. In IVM assays, E62Q previously has shown increased Calcium sensitivity. New molecular dynamics data shows mutation-induced changes to tropomyosin dynamics and interactions with actin and troponin. Human engineered heart tissues (EHT) were generated by seeding iPSC-derived cardiomyocytes engineered using CRISPR/CAS9 to express either E62Q or E54K cardiomyopathy mutations. After two weeks in culture, E62Q EHTs showed a drastically hypercontractile twitch force and significantly increased stiffness while displaying little difference in twitch kinetics compared to wild-type isogenic control EHTs. On the other hand, E54K EHTs displayed hypocontractile isometric twitch force with faster kinetics, impaired length-dependent activation and lowered stiffness. Given these contractile abnormalities, we hypothesized that small molecule myosin modulators to appropriately activate or inhibit myosin activity would restore E54K or E62Q EHTs to normal behavior. Accordingly, E62Q EHTs were treated with 0.5μM mavacamten (to remedy hypercontractility) and E54K EHTs with 0.5 μM danicamtiv (to remedy hypocontractility) for 4 days, followed by a 1 day washout period. Upon contractility testing, it was observed that the drugs were able to reverse contractile phenotypes observed in mutant EHTs and restore contractile properties to levels resembling those of the untreated wild type group. The computational, IVM and EHT studies provide clear evidence in support of the hyper- vs. hypo-contractility paradigm as a common axis that distinguishes HCM and DCM TPM1 mutations. Myosin modulators that directly compensate for underlying myofilament aberrations show promising efficacy in human in vitro systems.

ABSTRACT Amikacin and gentamicin pharmacokinetic behaviors after nebulization were determined by comparing plasma and pulmonary epithelial lining fluid (ELF) concentrations in rats after intratracheal and intravenous administrations. ELF areas under concentration-time curve were 874 and 162 times higher after nebulization than after intravenous administration for amikacin and gentamicin, respectively. Even if both molecules appear to be good candidates for nebulization, these results demonstrate a much higher targeting advantage of nebulization for amikacin than for gentamicin.
