Total Carotenoids Content and Assessment of Carotene Profile by High- Performance Liquid Chromatography in Selected Vegetables of Bangladesh with Special Reference to Some Unconventional Varieties

Background: Carotenoids are natural plant pigments and precursors of vitamin A, which provide a health benefit, protect against chronic degenerative diseases, and contribute to immune functions. Utilization and identification of foods with a high content of carotenoids received greater attention nowadays. Objective: The present study was aimed at evaluating total carotenoids content (TCC) and carotene composition of unconventional leafy vegetables growing in Bangladesh. Materials and Methods: The samples were collected from different locations of Bangladesh and mixed together to ensure sample representativeness. Acetone–petroleum ether extraction followed by spectrophotometric measurement was utilized for quantification of TCC. High-performance liquid chromatography (HPLC- DAD) was used to separate and quantify carotene components. Results and Discussion: The TCC values ranged from 129.38 μg/100 g edible portion (EP) in Helencha to 12803.31 μg/100 g EP in Roktodrone. The study findings also revealed that the selected plant samples exhibited high content of β-carotene ranging from 120.99 μg/100 g EP in Parul to 11301.16 μg/100 g EP in Roktodrone. It was also observed that most of the vegetables analyzed did not have α-carotene, lycopene, β-cryptoxanthin or it was in such a small quantity that could not be detected with HPLC. Conclusion: It can be suggested that regular intake of these vegetables may provide pro-vitamin A. In order to address the vitamin A deficiency and to maintain biodiversity, the analysis of carotenoids composition of these unconventional plant foods is to be initiated and make it available to the mass population. It would also fill up the data gap in the existing food composition table of Bangladesh.

Vitamin A deficiency is still one of the major public health problems in least developed countries. Fortification of vegetable oils is a strategy implemented worldwide to prevent this deficiency. For a fortification program to be effective, regular monitoring is necessary to control food quality in the producing units. The reference methods for vitamin A quantification are expensive and time-consuming. A rapid method should be useful for regular assessment of vitamin A in the oil industry. A portable device was compared to high-performance liquid chromatography (HPLC) for three plant oils (rapeseed, groundnut, and soya). The device presented a good linearity from 3 to 30 mg retinol equivalents per kg (mg RE.kg- 1). Its limits of detection and quantification were 3 mg RE.kg- 1 for groundnut and rapeseed oils and 4 mg RE.kg- 1 for soya oil. The intra-assay precision ranged from 1.48 % to 3.98 %, considered satisfactory. Accuracy estimated by the root mean squares error ranged from 3.99 to 5.49 and revealed a lower precision than HPLC (0.4 to 2.25). Although it offers less precision than HPLC, the device estimates quickly the vitamin A content of the tested oils from 3 or 4 to 15 mg RE.kg- 1.

Vitamin A deficiency continues to be a global public health problem. Fortification of oil with vitamin A is considered a cost-effective, feasible strategy to prevent this problem but quality control poses a challenge to program implementation. To overcome this, we have validated a newly developed device that quantitatively measures the content of retinyl palmitate in refined palm oil, is simple to use, and yields immediate results.Linearity of analysis ranged from 2.5 - 30 mg retinol equivalents (RE)/ kg of palm oil, with 2.5 mg RE/kg being the determination limit; inter- and intra-assay precision ranged from 1.4 - 7.1 %. Comparison with a high-performance liquid chromatography method showed high agreement between the methods (R2 = 0.92; Limits of Agreement: -1.24 mg to 2.53 mg RE/kg), and further comparisons illustrate that the new device is useful in low-resource settings. This device offers a field- and user-friendly solution to quantifying the vitamin A content in refined palm oil.

This study investigated the α- and β-carotene content and provitamin A value of four leafy vegetables sold at local and street markets in Viçosa, MG, Brazil, in the spring and winter of 2002. Carotenoids were analyzed by high-performance liquid chromatography. α-Carotene was detected in all samples sold during spring, but was only present in a few samples of smooth and curly lettuce and kale in winter. β-Carotene was found in marked quantities in all leafy vegetables analyzed. Duncan's test (α = 5%) showed significantly higher α-carotene content in curly lettuce and vitamin A value in large-leaved watercress in the spring. Mean β-carotene content and vitamin A value were 7544, 8751, 2584, 2792, 8193, and 5338 μg/100 g and 666, 760, 227, 238, 698, and 460 μg RAE/100 g in large-leaved and hydroponic watercress, smooth and curly lettuce, kale and spinach, respectively. All leafy vegetables analyzed represent important sources of provitamin A and supply an important part of the daily requirements of children and adults.

Broccoli is a source of bioactive compounds that provide an important nutritional value. The content of these compounds can vary depending on agronomic and environmental conditions, as well as on elicitation. In this study, three crop trials were carried out to evaluate the effects of the cultivation season, the application of different dosages of methyl-jasmonate (MeJA) on the overall quality and on the total content of bioactive compounds of ‘Parthenon’ broccoli cultivated under the field conditions of southeastern Spain. Color parameters, chlorophyll content, total phenolic compounds, total flavonoids and antioxidant activity were measured to evaluate the overall quality. Moreover, individual carotenoids, phenolic compounds and glucosinolates were evaluated by high performance liquid chromatography with diode array detection (HPLC-DAD) and high performance liquid chromatography equipped with diode array detector coupled to mass spectrometer using electro spray ionization (HPLC-DAD-ESI/MSn). The content of total carotenoids, phenolic compounds and glucosinolates were higher in autumn compared with spring, showing increases of 2.8-fold, 2-fold and 1.2-fold, respectively. Moreover, a double application of MeJA increased the contents of total carotenoids, phenolic compounds and glucosinolates by 22%, 32% and 39%, respectively, relative to the untreated samples. Considering our results, the controlled and timely application of 250 µM MeJA to the aerial parts of the plants four days before harvest, on two consecutive days, seems to be a valid agronomic strategy to improve the health-promoting capacity of Parthenon broccoli, without compromising its overall quality.

Tingginya tingkat konsumsi minyak goreng oleh Masyarakat Indonesia menjadikan minyak goreng digunakan sebagai food vehiclepada program fortifikasi vitamin A.Program tersebut dilakukan untuk mengatasi Kekurangan vitamin A (KVA). KVAsangat mempengaruhi kelangsungan hidup anak dan standar upaya penyelamatan proses kehamilan dan persalinan. Namun pemanasan yang terjadi selama proses penggorenganpada minyak dapatmerusak jumlah vitamin A yang difortifikasi. Tujuan penelitian ini untuk membandingkan nilai retensi vitamin A di dalam kentang yang digoreng dengan menggunakan minyak goreng curah dan minyak goreng kemasan fortifikasi dengan pengaruh suhu dan waktu penggorengan. Penelitian ini menggunakan metode eksperimental disain rancangan acak lengkap (RAL) dengan dua faktor, suhu (140dan 170oC) dan waktu penggorengan (10 dan 15 menit) tanpa pengulangan.Sampel penelitian yang digunakan adalah kentang beku, minyak goreng curah dan minyak goreng kemasan fortifikasi.Nilai retensi vitamin A kentang dan minyak goreng diukur dengan menggunakan metode High Performance Liquid Chromatography (HPLC).Analisis perbedaan dan interaksi variabel diolah dengan ANOVA dua arah. Berdasarkan luas area puncak vitamin A pada kromatogram sebelum dan setelah penggorengan, ditemukan kecenderungan penurunan persentase retensi vitamin A dari minyak goreng kemasan fortifikasi dibandingkan minyak goreng curah.Nilai retensi vitamin A pada kentang yang digoreng dengan minyak goreng curah adalah 217-251%,sedangkan retensivitamin A kentang yang digoreng dengan minyak goreng kemasan fortifikasi adalah 48-131%. Analisis ANOVA dua arah tanpa pengulangan untuk persen retensi vitamin A menunjukkan bahwa (p<0,05) variabel jenis minyak goreng berpengaruh secara nyata terhadap retensi vitamin A. Retensi vitamin A pada kentang goreng yang digoreng dengan minyak goreng curah jauh lebih tinggi dibanding minyak goreng kemasan fortifikasi
