Purwoceng (Pimpinella pruatjan Molk or Pimpinella alpine KDS) belongs to the Apiaceae family, is an endemic plant in mountainous areas such as the Dieng Plateau, Pangrango Mountain, and mountainous areas in East Java. Parts of the plants, especially roots are efficacious as aphrodisiacs, diuretic drugs and tonics. Based on CITES, this plant belongs to the category of endangered species, so its presence is very rare in nature. This scarcity occurs due to the purwoceng exploitation in nature as traditional herbal medicine without any cultivation or rejuvenation. In situ conservation efforts are almost impossible because the original habitat of this plant has become extinct. Thus, ex situ conservation is more suitable to be applied, by using in-vitro culture techniques. Therefore, a research study is needed on the development of tissue culture techniques in the production of the seeds. Methods of data collection in this study was conducted by literature studies. Therefore, ex-vitro root induction techniques are applied currently, namely rooting induction. The content of the chemical compounds and various properties make purwoceng as a commodity in the raw materials of drugs that have great potential to promote industrial players in the economic field. The genus Juniperus (of the Cupressaceae family) is the second most prevalent group of conifers on Earth. Juniper species are widely dispersed in the Northern Hemisphere, in Europe and Asia, and in Africa and Central America. Juniper species are resistant to dry climates and can adapt to difficult environmental conditions. Most juniper species are important in both ecological and economic terms. However, today, many forests in which junipers occur are being reduced in size due to both natural causes (fires, for example) and human activity (uncontrolled exploitation of forests, etc.). Also, climate changes may have adversely affected the range of populations of different juniper species. For this reason, some juniper species are now categorized as rare or endangered, and require immediate protective action. Therefore, there is an urgent need to develop effective strategies for ex situ conservation, including reliable procedures for Juniperus sp. reproduction for future reintroduction and restoration programs. The conservation strategies used until now with traditional forestry techniques (seed propagation, rooted cuttings, grafting) have not been satisfactory in many cases. Thus, increasing attention is being paid to the possibilities offered by in vitro culture technology, which enables the conservation and mass clonal propagation of different coniferous tree species. In this mini-review, we summarize the current state of knowledge regarding the use of various methods of the propagation of selected Juniperus species, with a particular emphasis on in vitro culture techniques.

Mini tomato cv. Micro-Tom is a plant favored by researchers because it has a shortlife cycle and small in size so that it becomes a practical plant and is suitable for representingresearch on tomato plants or other plants of the Solanaceae family. Unlike ordinary tomatoplants which are large, take up space to grow, and sometimes have a long life cycle, makingordinary tomato plants a plant that is not so easy to handle. For that reason, the mini tomato isnow described as a laboratory tomato. The aim of research is prepared mini tomato forgenomic study. The research was conducted at Politeknik Negeri Lampung Tissue CultureLaboratory from April to September 2020. The material used for this research was mini tomatoseeds. The explants used for the multiplication in this study were the cotyledons. These studiesused macro elements, microelements, tomato vitamins, CaCl2, and FeSO4. EDTA to makeMurashige and Skoog (MS) media, co-cultivation media, and callus induction media, shootinduction media, and root induction media for generated tomato plants. We successfullydeveloped a mini tomato until final root induction. The result of these studies will use forgenomic study (next research).

The aim of this study was to evaluate the effect of 6-benzylaminopurine (BAP) separately and in interaction with auxins on the change in the qualitative and quantitative composition of flavonoids in the raw materials of regenerating plants Iris sibirica L. Cambridge grade in comparison with aeroponic and intact raw materials using the method of high-performance liquid chromatography. Raw materials of I. sibirica Cambridge variety obtained in vitro culture had a richer qualitative composition of flavonoids than intact plants. The dependence of the accumulation of flavonoids on the concentration of 6-benzylaminopurine in nutrient media was noted. The presence of 13 compounds was observed in extracts of 70% ethyl alcohol from regenerating plants grown at the lowest concentration of BAP (1.0 µM) within the experiment. In quantitative terms, the flavonoid apigenin was maximally determined on a medium with BAP 1 µM, and kaempferol - on media with BAP 5.0 µM, supplemented with auxins. For a medium with 7.5 µM BAP, the lowest variety of compounds was observed (9) and the lowest kaempferol content. Auxins influenced the synthesis of flavonoids. The amount of flavonoids in all variants of the experiment increased by an average of 13% in the presence of auxins. The stages of the technological process of obtaining raw materials I. sibirica Cambridge variety on the basis of clonal micropropagation and cultivation in aeroponics conditions allowed to obtain raw materials that do not contain heavy and toxic metals, are not infected with pathogens and pests. With 1 m2 of useful area of aeroponics for 1 year, it is possible to collect 5 times more raw materials than with field cultivation. According to the qualitative composition of phenolic compounds, aeroponic raw materials are identical to intact plants.

Epigenetic variation, and particularly DNA methylation, is involved in plasticity and responses to changes in the environment. Conservation biology studies have focused on the measurement of this variation to establish demographic parameters, diversity levels and population structure to design the appropriate conservation strategies. However, in ex situ conservation approaches, the main objective is to guarantee the characteristics of the conserved material (phenotype and epi-genetic). We review the use of the Methylation Sensitive Amplified Polymorphism (MSAP) technique to detect changes in the DNA methylation patterns of plant material conserved by the main ex situ plant conservation methods: seed banks, in vitro slow growth and cryopreservation. Comparison of DNA methylation patterns before and after conservation is a useful tool to check the fidelity of the regenerated plants, and, at the same time, may be related with other genetic variations that might appear during the conservation process (i.e., somaclonal variation). Analyses of MSAP profiles can be useful in the management of ex situ plant conservation but differs in the approach used in the in situ conservation. Likewise, an easy-to-use methodology is necessary for a rapid interpretation of data, in order to be readily implemented by conservation managers.

Context: Orchid produces a huge number of minute seeds but the seeds can not germinate easily in nature due to the lack of endosperm in the seeds is an incompatibility barrier that limits its propagation in nature. Objectives: To develop in vitro culture techniques for quick propagation of Vanda tessellate, a commercially important orchid species. Materials and Methods: Shoot tips were used as experimental materials. The explants were surface sterilized and the shoot tips were excised. The isolated shoot tips were cultured in MS medium supplemented with different concentration and combinations of auxin and cytokinin. Results: The combination of 1.5 mgl-1 NAA and 1.0 mgl-1 BAP was proved to be the best medium formulation for multiple shoot formation as well as maximum shoot elongation. The single shoots were isolated from the multiple shoots and subcultured in MS medium having NAA and IBA individually and in combinations for root induction. Maximum root induction was obtained in MS agarified medium having 0.5 mgl-1NAA and 1.0 mgl-1IBA. The well rooted plantlets were hardened successfully in the potting mixture containing coconut husk, perlite, charcoal, brick pieces in the ratio of 2:1:1:1 and eventually established under natural condition.Conclusion: An efficient regeneration protocol for micropropagation in V. tessellata through shoot tip culture has been established.Key words: Shoot tip; micropropagation; orchid.DOI: 10.3329/jbs.v17i0.7122J. bio-sci. 17: 139-144, 2009

An experiment was conducted to study the in vitro culture of pointed gourd. Cotyledon rescued from physiologically matured seeds (PMS) and immatured seeds (IMS) of pointed gourd were used as explants. Cotyledon excised from PMS responded very well in all culture conditions. Plant regenerated from cotyledon of PMS ranged from 38 to 96% in different hormonal formulations of culture media. Highest percentage of shoot regeneration was observed in MS + 1.0 mg/l BAP and lowest in MS + 2.5 mg/l BAP. No plant regeneration was observed in cotyledon from immatured seeds. The highest percentage of root induction (99%) was achieved in half MS medium supplemented with 0.5 mg/l NAA. The regenerated plantlets were successfully established in earthen pot.
