SummaryWe have evidence that ristocetin and botrocetin mediate binding of von Willebrand Factor (vWF) to platelet glycoprotein lb (GPIb) through two distinct domains on the vWF molecule. This was established by using monoclonal antibodies (MAbs) to vWF and synthetic peptides derived from the sequence of vWF. MAb 322 and MAb NMC/vW 4 both recognize native vWF as well as fragments containing the GPIb-binding domain of vWF, obtained with the following enzymes: trypsin (116 kDa), V-8 pro tease (Spill, 320 kDa) and V-8 protease plus subtilisin (33-28 kDa). Nevertheless, the lack of reciprocal displacement between the two MAbs in experiments of competitive inhibition for binding to vWF demonstrate that their respective epitopes are separate. Both MAbs inhibit 125I-vWF binding to platelet membrane GPIb and vWF-dependent platelet agglutination induced by ristocetin. However, only MAb NMC/vW4 inhibits these functions in the presence of botrocetin and when ristocetin-induced platelet agglutination is inhibited by MAb 322, botrocetin is still able to restore the agglutination. The involvement of two distinct domains of vWF for binding to GPIb in the presence of ristocetin or botrocetin was confirmed in experiments of binding of 125I-vWF to platelets using as competitor synthetic peptides corresponding to the GPIb binding domain of vWF (Cys 474 to Pro 488 and Ser 692 to Pro 708). At a final concentration of 2.5 mM both peptides inhibit more than 90% of the binding of vWF to ristocetin-treated platelets but are unable to modify this binding in the presence of botrocetin. In conclusion our data suggest that botrocetin and ristocetin involve distinct sites on vWF for binding to GPIb.

Enormous successes have been obtained against the control of major epidemic diseases, such as SARS, MERS, Ebola, Swine Flu in the past. Dynamic interplay of biological, socio-cultural and ecological factors, together with novel aspects of human-animal interphase, pose additional challenges with respect to the emergence of infectious diseases. The important challenges faced in the control and prevention of emerging and re-emerging infectious diseases range from understanding the impact of factors that are necessary for the emergence, to development of strengthened surveillance systems that can mitigate human suffering and death. The aim of the current study is to assess the awareness of symptomatic differences between viral diseases like COVID-19, SARS, Swine flu and common cold among dental students that support the prevention of emergence or re-emergence. Cross-sectional type of study conducted among the undergraduate students comprising 100 Subjects. A questionnaire comprising 15 questions in total were framed, and responses were collected in Google forms in SPSS Software statistical analysis. The study has concluded that dental students have an awareness of the symptomatic differences between infectious viral disease. The study concluded that the awareness of symptomatic differences between viral diseases like COVID-19, SARS, Swine flu, Common cold is good among the dental students who would pave the way for early diagnosis and avoid spreading of such diseases. A further awareness can be created by regular webinars, seminars and brainstorming sessions among these healthcare professionals.

Background: Stray and pet dogs constitute the dog population in Bangladesh without any established dog statistics. Diseases of dogs are not only associated with morbidity and mortality in dogs but also associated with human health problems as zoonotic diseases. Inland reports on the prevalence of dog diseases are limited in Bangladesh and there is need to investigate diseases in both stray and pet dog populations. Objectives: The main objective is to determine the prevalence of Canine distemper (CD), Canine adenovirus- 1 (CAV-1), CAV-2, Canine influenza (CI) and Dirofilaria immitis (Heart worm infection = HWI) infection in stray dogs in Bangladesh. Materials and Methods: Blood, ocular and nasal samples were collected from each of 30 randomly caught stray dogs (11 male and 19 female) and of different ages, including growing (n = 3) and adult (n = 27) dogs in the district of Mymensingh in Bangladesh from January to June 2010. These samples were tested by using Antigen Test Kits (RapiGEN Inc, Korea). Results: All the 30 stray dogs showed negative results to CD, CAV-1 and CAV-2, whereas an overall high prevalence of CI (4/30; 13.33%) and HWI (15/30; 50.0%) were recorded. The higher prevalence of CI was recorded in growing (n = 2/3; 66.67%) than adult (n = 2/27; 7.41%) but it did not differ significantly (p > 0.05) between male (n = 2/11; 18.18%) and female (n = 2/19; 10.53%) dogs. Significantly (p < 0.05) higher prevalence of D. immitis infection was recorded in adult (n = 15/27; 55.56%) than growing (0/3; 0.0%), male (n = 8/11; 72.73%) than female (n = 7/19; 36.84%) dogs. Conclusions: Further studies on CI and D. immitis are necessary on a large population of stray and pet small animals along with humans to ascertain their importance on health and zoonotic significance in Bangladesh.
