The purpose of this research was to determine the effect of progesterone to estradiol levels and histology of uterus, to test the effect of estrous cycle to uterus histology of females’ mice after treated by progesterone in birth control pills. This research used Completely Randomized Design (CRD). Females Mus musculus Balb-C and synthetic progesterone (cyproterone acetate) contained in ‘Diane-35’ birth control pills were used in this study. The research consisted of two treatment groups, i.e. group K had no treatment and group P treated by a solution of Diane-35 at a dose of 2.6 mg per 5 mL distilled water. Results on the effects of progesterone on endometrial thickness suggested that there were significant differences between treatment groups. Meanwhile, the effect of progesterone on estradiol levels indicated that there were no significant differences among treatment groups. This means that the thickness of the endometrium was strongly influenced by the presence of a synthetic progesterone hormone and induced endometrium growth thicker. The levels of estradiol are not significantly different due to a negative feedback mechanism of Follicle Stimulating Hormone (estrogen effect) and Luteinizing Hormone (progesterone effect). The greatest thickness of endometrium of estrous cycle was treated by progesterone, which was 0.2500 mm2.

Abstract. The levels of oxytocin receptor (OTR), cytosolic progestin receptor (cPR), cytosolic and nuclear estrogen receptor (cER, nER) were measured in the endometrium of 28 heifers that had been slaughtered on a defined day of the estrous cycle, In an additional, trial endometrial tissue obtained from 78 heifers or cows at the abattoir was analyed for OTR. OTR was absent during the luteal phase (after day 6), but a minor elevation was observed after day 15. OTR increased rapidly after luteolysis on days 17–18 reaching a maximum during estrous on day 21, and decreased again during days 1–6. cER and cPR were different to OTR but followed a similar pattern with maximal levels during days 1–8 of the estrous cycle. At day 12 both receptors were minimal and increased again towards day 21. nER was maximal at day 19–21 coinciding with maximal estradiol levels and estrous. Our data indicate that owing to an increasing sensitivity of the endometrium to progesterone and estradiol after day 12, these steroids may be mainly responsible for the initiation of first PGF2α surges and luteolysis. Oxytocin seems to be of minor importance at this stage owing to low sensitivity of the endometrium for oxytocin.

Penelitian ini bertujuan untuk mengetahui peran polisakarida krestin (PSK) dengan waktu pemberian yang berbeda terhadap hitung jenis leukosit mencit yang diinfeksi Mycobacterium tuberculosis. Penelitian ini menggunakan 30 ekor mencit betina dewasa jenis Mus musculus strain BALB/C, berumur 8-10 minggu, berat badan berkisar 25-30 g. Polisakarida krestin (PSK) diisolasi dari Coriolus versicolor yang diperoleh dari alam. Infeksi menggunakan Mycobacterium tuberculosis strain H37Rv (ATCC 27294 T). Hewan percobaan dikelompokkan menjadi 6 kelompok sebagai berikut: kelompok I, hanya diberi akuades; kelompok II, hanya pemberian PSK; kelompok III, hanya dengan infeksi Mycobacterium tuberculosis; kelompok IV, pemberian PSK sebelum infeksi Mycobacterium tuberculosis; kelompok V, pemberian PSK sesudah infeksi Mycobacterium tuberculosis; kelompok VI, pemberian PSK sebelum dan sesudah infeksi Mycobacterium tuberculosis. Pemberian PSK dilakukan selama 7 hari berturut-turut melalui gavage. Infeksi Mycobacterium tuberculosis dilakukan sebanyak 2 kali dengan selang waktu 1 minggu melalui intraperitoneal. Hitung jenis leukosit dilakukan dengan mengelompokan masing-masing jenis leukosit dalam 100 sel leukosit pada apusan darah, dan data hasil pengamatan dianalisis dengan uji Kruskal-Wallis, kemudian untuk mengetahui signifikansi dilanjutkan dengan uji Mann-Whitney. Secara keseluruhan penelitian menunjukkan presentase jenis leukosit dengan jumlah tertinggi adalah neutrofil. Pada kelompok VI presentase monosit dan neutrofil meningkat melebihi normal, sedangkan presentase limfosit menurun, dan presentase basofil dan eosinofil tidak mengalami perubahan. Kesimpulan penelitian ini adalah PSK meningkatkan jumlah leuksosit mencit jenis neutrofil dan monosit pada waktu sebelum dan sesudah infeksi Mycobacterium tuberculosis

<p>Mycobacterium tuberculosis is a major infection agent of tuberculosis that is controlled by the response of cell-mediated immunity. It is macrophages and cytolytic T lymphocytes. Activated macrophages will produce free radicals. Excessive free radicals cause tissue damage. Polysaccharide krestin contains β-glucan. It is a scavenger of free radicals. This research aimed to identify the influence of polysaccharide krestin from C. versicolor on nitrite and malondialdehyde concentrations of mice serum exposed by M. tuberculosis. Nitrite concentration was determined by nitrite assay. Malondialdehyde concentration was determined by TBARS assay. The result showed that adding polysaccharide krestin before exposure (P1) and adding polysaccharide krestin before-after exposure (P3) had the best potential to decrease nitrite concentration. Nitrite concentrations of P1 and P3 were 1.364 ± 0.523 M and 1.456 ± 0.712 M respectively. Meanwhile, P1 group and adding polysaccharide krestin after exposure (P2) had the best potential to decrease malondialdehyde concentration. Malondialdehyde concentrations of P1 and P2 were 1125.86 ± 97.96 µM and 953.86 ± 328.16 µM respectively. Their nitrite and malondialdehyde concentrations decreased, compared to K and K- groups. The research conclusion was that adding polysaccharide krestin before exposure could decrease both nitrite and malondialdehyde concentrations.</p><p><strong>How to Cite</strong></p><p>Wahyuningsih, S., Pramudya, M., &amp; Sugiharto, S. <p><em>Pseudomonas aeruginosa </em>is an opportunistic bacterium that causes infections in human. The wall cell of its bacteria contains lipopolysaccharide as virulency factors to protect it from human immunity. Lipopolysaccharide can inhibit phagocytosis in the body. Polysaccharide krestin (PSK) from <em>Coriolus versicolor </em>extract contains β-glucan that can increase phagocytic activity and capacity. This research aimed to identify the effect on polysaccharide krestin from <em>C. versicolor</em> extract on phagocytic activity and capacity of mice exposed by <em>P. aeruginosa</em>. The design of this research was experimental design. There were six treatment groups. The phagocytic activity and capacity were counted on slide smears of mice peritoneal fluid. The data was analyzed by using one way ANOVA. The results of the phagocytic activity and capacity showed that PSK was added before exposure (P1) or after exposure (P2) or before-after exposure (P3) had potential to increase phagocytic activity and capacity.  The conclusion of the research was that adding polysaccharide krestin either before exposure or after exposure or both of them could increase phagocytic activity and capacity. The benefits of this research to development of science are expected to reduce human infection and to utilize natural ingredients as immunomodulator.</p><p><strong>How to Cite</strong></p><p>Wahyuningsih, S. The research was conducted to determine the effect of insulin-like growth factor-I (IGF-I) derived from pregnant crossbred mare serum (PMS) on estrous cycle and litter size in mice (Mus musculus). The research was arranged by Completely Randomized Design (CRD) with seven treatments and five replications. The treatments were control (CO)= physiological NaCl, treatment (T) 1 = 10 ng/ml of IGF-I pregnant mares crossbred serum (PMS), T2 = 20 ng/ml of IGF-I PMS, T3 = 40 ng/ml of IGF-I PMS, T4 = 10 ng/ml of IGF-I recombinant mouse, T5 = 20 ng/ml of IGF-I recombinant mouse, and T6 = 40 ng/ml of IGF-I recombinant mouse. Serie I the subject of this research were 35 female mice. Observed variables included estrous cycle phases that proestrus, estrus, metestrus, and diestrus phase. Serie II for the objects of this research were 35 female mice and 21 male mice in the same treatment on litter size in mice. The data were analyzed by Analysis of Variance (ANOVA), followed by HSD (Honestly Significant Difference) test. The result showed that the addition of IGF-I PMS did not significantly affect (p>0.05) on proestrus, metestrus, and diestrus phase. The addition of IGF-I PMS 40 ng/ml was extended (p<0.05) the estrus phase. It showed that there was not significantly difference (p>0.05) between the effect of IGF-I PMS and IGF-I recombinant mouse with the mice estrous cycles. The result of IGF-I derived from PMS and IGF-I recombinant mouse on litter size in mice did not significantly difference (p>0.05). Sex hormones, including estradiol, play important physiological roles in bone metabolism. The purpose of this study was to investigate whether there is estrous-cycle-dependent variation in orthodontic tooth movement, and, if so, to determine the mechanism. Ten-week-old female Wistar rats were used. They received repeated orthodontic force during specific phases in the estrous cycle. Tooth movement in animals that received force principally in estrus was about 33% greater than that in animals that received such force principally in pro-estrus (p < 0.05). Serum estradiol levels also varied according to the estrous cycle, with a peak during pro-estrus and a nadir during estrus, and were inversely related to tooth movement. Furthermore, there were negative correlations between estradiol and both serum TRAP activity and pyridinoline (r = -0.42, p < 0.05; r = -0.59, p < 0.001). These results suggest that cyclic changes in the estradiol level may be associated with the estrous-cycle-dependent variation in tooth movement through its effects on bone resorption.
