Background: Currently, there is a trend towards using natural and ethnopharmacological species with therapeutic potential. This investigation evaluated the antifungal activity of two species in the Ecuadorian Andes, which are used in treating dermatomycosis: Ambrosia arborescens Mill. (Marco) and Aristeguietia glutinosa Lam. (Matico). Methods: We worked with seven concentrations (100 to 700ppm) of Ambrosia arborescens Mill. extract and ten concentrations (0.5 to 5%) of essential oil (EO) of Aristeguietia glutinosa Lam. on Trichophyton mentagrophytes ATCC 9533, Trichophyton rubrum ATCC 28188, Microsporum canis ATCC 36299 and Candida albicans ATCC 10231. The methodology used was a modified version of the Kirby-Bauer method, using diffusion in agar wells. Results: The Tukey test, after the one-way Anova, determined effective concentrations of EO: 5% for Trichophyton mentagrophytes, 4.5% for Trichophyton rubrum, 5% for Microsporum canis and 2% for Candida albicans. In the extracts, the concentration of 700ppm was used for Trichophyton mentagrophytes, Trichophyton rubrum, and 600ppm for Microsporum canis and Candida albicans. Conclusions: The evaluation of the antifungal activity of the Ambrosia arborescens extract showed inhibition in the studied dermatophytes in each one of the planted concentrations (100 to 700ppm). The evaluation of the antifungal activity of Aristeguietia glutinosa EO showed inhibition in the studied dermatophytes in each of the planted concentrations (0.5 to 5%).

Acalypha wilkesiana (AW), a popular medicinal plant has been used in traditional medicine to treat a variety of skin disorders including pityriasis versicolor and seborrheic dermatitis. As a prelude to clinical trials in humans, an experimental study was carried out to determine the spectrum of antifungal activity of 2 variants of the Acalypha wilkesiana plant. Materials and Methods: The ethanol extract and herbal cream formulation of the dried leaves of 2 cultivars (Macrophylla & Hoffmani) of Acalypha wilkesiana were investigated for in-vitro antifungal activity by disc diffusion and micro-broth dilution techniques. Organisms tested were typed cultures of Malassezia furfur, Candida albicans and Trichophyton rubrum; and clinical strains of Microsporum canis and Epidermophyton floccosum. Results: Both cultivars (Macrophylla and Hoffmanii) of the plant showed good activity against all the fungi tested except Microsporum canis (8.0±0.00; 7.00±0.00 mm). The greatest activity was observed against Trichophyton rubrum (22.0±0.00; 24.00±0.00 mm) while Candida albicans showed the least activity (15.0±0.00; 18.00±0.57 mm). The Minimum Inhibitory Concentration (MIC) of the crude extract ranged between 0.25 and 8 mg/ml for all organisms, while that of the herbal cream was 0.31-8mg/ml. The lowest MIC was seen with Candida albicans for both varieties of the plant. The Acalypha wilkesiana Hoffmanii demonstrated a greater activity against Candida albicans and Malassezia furufur than the A. wilkesiana (Macrophylla). Conclusion: This study reveals Acalypha wilkesiana leaf extract has potential for development as a cream that can be used to treat superficial fungal skin infections.

Antifungal activities of Eugenia umbelliflora Berg. (Myrtaceae) were tested in vitro against a panel of standard and clinical isolates of human fungal pathogens (dermatophytes and opportunistic saprobes). Methanol extracts of leaves and fruits of E. umbelliflora were separately prepared and partitioned, to yield dichloromethane (DCM), ethyl acetate (EtOAc) and aqueous fractions (Aq). Three compounds (1-3) were obtained from the DCM extract using chromatographic procedures. Antifungal assays were performed using agar dilution techniques. Both extracts (fruits and leaves), their DCM and EtOAc fractions, and compound 2 (betulin and betulinic acid) presented selective antifungal activity against dermatophytes (Epidermophyton floccosum, Microsporum canis, Microsporum gypseum, Trichophyton rubrum, Trichophyton mentagrophytes), with MIC values between 200 and 1000 μg/mL, and interestingly, inhibited 4/5 species with MIC values of ≤500 ≤g/mL. The aqueous fractions of fruits and leaves, and compounds 1 (α, β amyrin) and 3 (taraxerol) were inactive up to the maximum concentrations tested (1000 μg/mL).

A onicomicose é definida como uma doença fúngica caracterizada pelo processo infeccioso da região é ungueal e, é responsável por desencadear uma má qualidade de vida aos pacientes e apesar de serem consideradas micoses superficiais, são difíceis de diagnosticar e tratar, com isso, tem impulsionado a realização de pesquisas na tentativa de obter outras opções terapêuticas além dos medicamentos sintéticos disponíveis na área da saúde. Diante do exposto, o presente estudo teve por finalidade avaliar a ação in vitro dos óleos essenciais de Copaifera officinalis, Eugenia caryophylata, Melaleuca alternifolia, Rosmarinus officinalis e Thymus vulgaris frente a fungos causadores de onicomicose. Para a pesquisa da atividade antifúngica in vitro dos óleos essenciais selecionados foi utilizado o método teste de placa de ágar com orifício frente às cepas de Candida albicans (ATCC 10231), Microsporum canis (ATCC 14055) e Trichophyton rubrum (ATCC 28189), nas proporções de 100%, 25% e 5% sendo a leitura das placas e interpretação dos resultados avaliados a partir da observação do halo de inibição, considerando a área sem crescimento detectável a olho nu. Observou-se que os óleos essenciais de Eugenia caryophylata, Melaleuca alternifolia, Rosmarinus officinalis e Thymus vulgaris apresentaram atividade antifúngica com formação de halos sobre as espécies fúngicas leveduras e dermatófitos, sendo que o óleo de Copaifera officinalis, apresentou apenas uma fraca atividade em relação as espécies fúngicas testadas. Portanto, conclui-se que os óleos essenciais analisados apresentaram atividade antifúngica, abrindo a perspectiva de novos estudos para caracterização de seus princípios antimicrobianos e alternativas ditas naturais para tratamento dessa podopatia.

Los dermatofitos causan infecciones que en ocasiones no responden adecuadamente al tratamiento antifúngico implementado, sin embargo, se han publicado muy pocos estudios de sensibilidad a nivel mundial. El objetivo de este trabajo fue evaluar in vitro, la sensibilidad de hongos dermatofitos aislados de muestras clínicas frente a tres antifúngicos de uso habitual. Se trabajó con 50 cepas de las especies: Trichophyton mentagrophytes, Trichophyton rubrum, Microsporum canis, Nannizzia gypsea, Trichophyton tonsurans y Epidermophyton floccosum. Los antifúngicos probados fueron terbinafina, itraconazol y fluconazol. Para el estudio de la sensibilidad se desarrolló la técnica de microdilución en caldo según el documento M38-3ra ed. del Clinical and Laboratory Standards Institute. El inóculo fue ajustado a 103 UFC/ml. Se incubaron a 35° C con lecturas diarias hasta las 96 horas. Terbinafina fue la de mayor actividad antifungica (CIM entre 0,03 y 0,50 μg/ml), seguida por itraconazol (CIM entre 0,12 y 4 μg/ml). Fluconazol mostró la menor actividad antifúngica (CIM de 8 a > 64 μg/ml). Se concluye que la terbinafina presenta la mayor actividad antifúngica in vitro frente a las cepas de dermatofitos estudiadas.

ABSTRACT R126638 is a novel triazole with in vitro activity similar to that of itraconazole against dermatophytes, Candida spp., and Malassezia spp. In animal models of dermatophyte infections, R126638 showed superior antifungal activity. R126638 inhibits ergosterol synthesis in Candida albicans, Trichophyton mentagrophytes, Trichophyton rubrum, and Microsporum canis at nanomolar concentrations, with 50% inhibitory concentrations (IC50s) similar to those of itraconazole. The decreased synthesis of ergosterol and the concomitant accumulation of 14α-methylsterols provide indirect evidence that R126638 inhibits the activity of CYP51 that catalyzes the oxidative removal of the 14α-methyl group of lanosterol or eburicol. The IC50s for cholesterol synthesis from acetate in human hepatoma cells were 1.4 μM for itraconazole and 3.1 μM for R126638. Compared to itraconazole (IC50 = 3.5 μM), R126638 is a poor inhibitor of the 1α-hydroxylation of 25-hydroxyvitamin D3 (IC50 > 10 μM). Micromolar concentrations of R126638 and itraconazole inhibited the 24-hydroxylation of 25-hydroxyvitamin D3 and the conversion of 1,25-dihydroxyvitamin D3 into polar metabolites. At concentrations up to 10 μM, R126638 had almost no effect on cholesterol side chain cleavage (CYP11A1), 11β-hydroxylase (CYP11B1), 17-hydroxylase and 17,20-lyase (CYP17), aromatase (CYP19), or 4-hydroxylation of all-trans retinoic acid (CYP26). At 10 μM, R126638 did not show clear inhibition of CYP1A2, CYP2A6, CYP2D6, CYP2C8, CYP2C9, CYP2C10, CYP2C19, or CYP2E1. Compared to itraconazole, R126638 had a lower interaction potential with testosterone 6β hydroxylation and cyclosporine hydroxylation, both of which are catalyzed by CYP3A4, whereas both antifungals inhibited the CYP3A4-catalyzed hydroxylation of midazolam similarly. The results suggest that R126638 has promising properties and merits further in vivo investigations for the treatment of dermatophyte and yeast infections.

Resumen:Objetivo: Determinar la frecuencia de las dermatomicosis en personas de diferentes instituciones de atención social en la ciudad de Manizales durante el año 2011. Método: Mediante la toma de muestras de los sitios que presentaban algún tipo de lesión sospechosa de ser una micosis cutánea, se hizo un análisis directo con KOH y cultivo en medios de Saboureaud y Mycosel. Se aplicó un instrumento de recolección de información para establecer  factores asociados con la presencia de estos microorganismos.Resultados: Los hongos levaduriformes encontrados con mayor frecuencia fueron: Candida albicans, Trichosporon sp, y los mohos saprofitos Penicillium sp, fusarium sp; seguido de hongos dermatofitos como: Trichophyton mentagrophites, Trichophyton rubrum, Epidermophyton floccosum y Microsporum gypseum. Las lesiones secas y descamativas se encontraron con mayor frecuencia. El compartir baños y vivir en hacinamiento y el uso de elementos comunes fueron los factores asociados más importantes en este estudio.Conclusiones. Las dermatomicosis son frecuentes en poblaciones vulnerables y se asocian a diferentes factores muy similares a los encontrados en otros estudios de igual naturaleza.    Palabras clave: dermatomicosis, onicomicosis, jóvenes, ancianos, factores asociados. Summary:Objective: To determine the frequency of ringworm in people of different social care institutions in the city of Manizales in 2011.Method: Using the sampling sites that had some kind of suspicious lesion from a cutaneous mycosis, direct analysis with KOH and culture media was Saboureaud and Mycosel. An instrument of data collection was used to establish factors associated with the presence of these microorganisms.Results: The yeast found most frequently were: Candida albicans, Trichosporon sp and Penicillium molds saprophytes sp, Fusarium sp, followed by dermatophyte fungi such as Trichophyton mentagrophytes, Trichophyton rubrum, Epidermophyton floccosum and Microsporum gypseum. Dry scaly lesions were found more frequently. The shared bathrooms and living in overcrowded and the use of common elements were the most important in this study associated factors.Conclusions: Dermatomycoses are common in vulnerable populations and are associated with different very similar to those found in other studies of the same nature factors. Keywords: ringworm, onychomycosis, young, old, associated factors.

Trabajo de investigación desarrollado con el objetivo de conocer la frecuencia de la dermatofitosis humana en una comunidad con desigualdad social. Hipótesis: la dermatofitosis humana es una afección fúngica muy frecuente en comunidades con desigualdad social. Zona de estudio: comunidad con desigualdad social periurbana de la ciudad de Ayacucho. Tipo de estudio: no experimental. Diseño de estudio: descriptivo-transversal. Muestra: no probabilística, individuos con signos de afecciones compatibles a micosis superficial. Metodología: muestras de escamas de piel, pelos, fragmentos de uñas de pies y manos, escamas de planta, espacios interdigitales y otras partes de cuerpo, fueron tomadas con una hoja de bisturí de filo romo o con el borde de un portaobjetos previa desinfección con alcohol al 70% y colocadas en sobres de papel oscuro etiquetados, en el laboratorio de Epidemiología y Micología de la Universidad Nacional de San Cristóbal de Huamanga, se realizó la observación directa con KOH al 20% y la siembra en placas Petri conteniendo Agar Sabouraud y Agar selectivo para hongos patógenos, después de una incubación a 25ºC por hasta 14 días; las colonias coincidentes con dermatofitos fueron observados al microscopio para identificarlos. Resultados: se ha encontrado que 85/153 (55,5%) presentaron diversas formas de dermatofitosis, no se observó preferencia de la dermatofitosis humana con relación al género (p> 0,05), los factores asociados a la dermatofitosis determinados estadísticamente (p< 0,05) fueron la higiene, el piso de la vivienda y la crianza de animales, se han identificado las especies de Trichophyton rubrum, Trichophyton mentagrophytes, Microsporum canis y Microsporum gypseum.
