Introduction. TStandard sperm examination, performed according to WHO guidelines, remains the main method of assessment of male fertility. At the same time, this research method has a number of significant limitations, including a low predictive value regarding the outcomes of assisted reproductive technologies (ART) programs and the outcome of naturally occurring pregnancies. The limitations of standard sperm examination dictate the need for additional methods for assessing male fertility. The most promising and widely used test is the assessment of the level of sperm DNA fragmentation. Aim. To evaluate the associations between the levels of sperm DNA fragmentation and the age of the patients along with the following parameters used as a part of standard sperm analysis: semen volume, total number of spermatozoa, percentages of progressively-mobile, non-progressively-mobile and immobile forms, percentages of morphologically normal forms and the forms bearing head defects within the structure of total number of morphological anomalies, as well as semen leukocyte count. Materials and methods. Study materials were the examination results from 121 males aged from 21 to 53 years old (mean age 32.7±4.5 years old), undergoing an examination within the Clinical Institution «Mother and Child – Yaroslavl» during a time period from January 2019 until April 2020. Standard sperm analysis procedures were carried out according to latest edition of WHO Guidelines (2010). The determinations of germ cell DNA fragmentation levels were performed using the TUNEL method. Results. The test results have revealed a weak negative relation between the sperm DNA fragmentation (%) and the percentage of progressively-mobile forms (%) – r = -0.26 (p <0.01). The correlation of sperm DNA fragmentation with age and other parameters was considered statistically insignificant. The second stage of the analysis have demonstrated that the increased degree sperm DNA fragmentation is 1.8-fold more often found in the patients having signs of astenozoospermia (23.6%) comparing to the patients showing normal degree of spermatozoa mobility (13.1%), (p <0.05). Conclusions. The level of sperm DNA fragmentation correlates with the percentage of progressively-mobile forms of spermatozoa (negative relation) and no correlations were found with other semen parameters and with the age. The rates of increased levels of sperm DNA fragmentation are 1.8-fold more often found in astenozoospermia patients comparing to the patients showing normal degrees of spermatozoa mobility.

For conception and the development of healthy embryos, sperm DNA integrity is crucial. According to a growing body of studies, there is a strong correlation between sperm DNA damage and male infertility. Among the new medicines being developed in the medical field, the application of Platelet Rich Plasma (PRP) in human reproduction has yet to be examined. A total of 100 semen samples were used in the current experimental investigation. From November 2020 to June 2021, the research was conducted at the High Institute for Infertility Diagnosis and Assisted Reproductive Technologies. Masturbation was used to get an ejaculated semen sample. After semen analysis, the samples were separated into two equal parts, one without autologous PRP and the other with 2% autologous PRP, with the DNA fragmentation assessed using the Sperm Chromatin Dispersion Test. There was highly significant reduction in DNA fragmentation index (p < 0.001). The mean sperm DNA integrity was reduced after adding PRP (33.85±16.73 vs 38.55±16.64), Mean (± SE). PRP has been shown to improve human sperm DNA integrity.

The article provides an overview of studies of antioxidant properties of the components contained in AndroDoz, as well as the results of using AndroDoz biocomplex, due to which infertile men showed an increase in concentration, motility and percentage of sperm with a normal form in the ejaculate, a decrease in sperm DNA fragmentation and an increase in pregnancy rates after using assisted reproductive technologies, as well as getting pregnant naturally. Due to absence of adverse events and side effects, AndroDoz can be recommended to a wide range of patients with pathozoospermia and idiopathic infertility.

The article provides an overview of the most influential papers on the topic of “male infertility”. The selection criteria were the practical significance of the article and the impact factor of the journal in which it was published, according to SCImago Journal Rank (SJR). As a result, we created the list of 10 papers published in the I quarter (January – March) of 2020. The review included articles on the following issues: the effectiveness and safety of antioxidants, zinc and folic acid in the treatment of male infertility; the effect of obesity on sperm DNA-fragmentation; the possibility of obtaining of spermatogonia in children and adolescents with Kleinfelter syndrome; comparison of intracytoplasmic sperm injection (ICSI) and intracytoplasmic morphologically selected spermatozoa injection (IMSI); the reaction of the female reproductive system to the seminal fluid; viral profile in seminal fluid of men receiving the antiretroviral therapy, the effect of sleep duration on fertility and the risk of diabetes mellitus in children, conceived using assisted reproductive technologies.

AIM: This study aimed to assess the difference in sperm concentration, total motility, progressive motility, and deoxyribonucleic acid (DNA) fragmentation index (DFI) before and after processing with 5 and 10 min swim-up (SU). METHODS: Fifty patients who met the study inclusion criteria from June 2020 to October 2020 were subjected to routine semen and sperm DNA fragmentation analysis. Each of the samples was then divided into three tubes, one as control and the others were processed using the SU method with 5 and 10 min centrifugation time, respectively. After being processed, the samples were subjected again to routine semen and sperm DNA fragmentation analysis. The results were being compared among three groups. RESULTS: The sperm concentration after 5 and 10 min SU (27.78–39.79 and 35.36–51.09, respectively; p < 0.05) was significantly higher compared to fresh semen (24.85–32.33). The total motility before and after 5 and 10 min SU were 43.78–51.08, 97.66–98.20, and 97.86–98.20, respectively. The progressive motility after 5 and 10 min SU (0–41 and 0–54, respectively) was significantly higher than fresh semen (0–24; p < 0.05). The DFI was significantly better after 5 min SU (3.82–6.98) compared to fresh semen and after 10 min SU (13.48–19.04 and 1–25, respectively; p < 0.05). CONCLUSION: Prolonged centrifugation time may yield a higher number of sperm concentration and motility, but it may also lead to a higher DFI. Hence, a shorter centrifugation time should be used for a better semen quality intended for assisted reproductive technologies.


The effect of the level of DNA fragmentation in the sperm on the development of early embryos in IVF programs for men with low fertility was investigated. A significant negative association between the level of DNA fragmentation in the sperm and the total blastocyst formation rates and a percent of blastocysts with high quality morphology in patients with impaired fertility is proved (rs = -0,59; rs= -0,56). Evaluation of semen quality in men with low fertility is a necessary step before the use of assisted reproductive technologies. The connection between the levels of sperm DNA fragmentation and the peculiarities of the early development of embryos obtained in programs of extracorporal fertilization has been proved. Assessing the quality of sperm in men with reduced fertility is a necessary measure before using auxiliary reproductive technologies. According to literature, the morphology of the blastocyst does not always depend on chromosomal changes in the embryos. Consequently, it is promising to carry out pre-implantation genetic diagnostics of embryos obtained in programs of extracorporal fertilization in patients with high levels of DNA fragmentation in spermatozoa to study the effect of this violation on the karyotype of embryos.
