This study was conducted in Grand station of Diwania/ Taj Al-Nahrain company- Al-Qadisiya Province for 84 days to investigate the effect of level of addition of commercial baker's yeast Saccharomyces cerevisiae,0, 15 and 30 g/cow/day to concentrate diet on rumen fermentation characteristics  in German Holstein Friesian lactating cows.  Nine multiparous Holstein cows in their mid-lactation were used. Daily feeding program applied in that station included offering 1 kg of concentrate diet per 3-4 kg milk produced divided into morning and evening meals, 10 kg of green forage, 10 kg whole plant corn silage and free choice of rice straw. Silage was characterized with accepted quality and fermentation parameters, values of pH, concentrations of ammonia nitrogen (NH3-N) and total volatile fatty acids (TVFA) were, 3.5, 0.98% of total nitrogen and 2.55 mM/DM respectively. Results revealed that addition of yeast at level of 15 and 30 g/cow/day reduced (P<0.01) concentration of NH3-N from 18.14 to 13.92 and 14.21 mg/100 ml respectively. Significant (P<0.01) increase in concentration of TVFA from 7.32 to 9.39 mM /100 ml was also observed due to addition of yeast at level of 15 g/cow/day. However, ruminal pH values were not affected by addition of yeast

The effects of glycerol with fat sources as a feed alternative were investigated in the ration for Nellore steers (Bos indicus). Eight cannulated steers at 30 months of age with initial bodyweight of 554 ± 36.0 kg were used in a double 4 by 4 Latin square design with four consecutive 16-day periods. Treatments were three different diets with lipid sources (soybean, soybean oil and calcium salts of fatty acids) and one diet control without lipid sources. All diets formulated contained 10% crude glycerol and 5% ether extract with a forage:concentrate ratio of 35:65; corn silage was used as forage. The ruminal liquid was sampled for 24 h and ruminal fermentations were monitored by measuring pH, concentrations of ammonia nitrogenand volatile fatty acids in rumen fluid. Urine samples were obtained from the total collection for 24 h for estimation of rumen microbial protein supply using urinary purine derivatives. Our results showed that the use of lipid sources combined with glycerol did not induce significant changes in rumen pH, acetate molar proportion, ruminal microbial protein or dry matter intake. Although the acetate molar proportion was kept constant within normal parameters, the propionate molar proportion was increased by the diet containing lipid sources. Moreover, we found that there was a negative effect of lipid sources on crude protein and neutral detergent fibre corrected for ash and protein intake, and one positive effect on ether extract intake. We believe that association between fat and glycerol may affect rumen fermentation parameters through reducing fibre intake and increasing propionate production and ammonia nitrogen.

Abstract This study determined the buffering capacity of buffer agents and its effect on in vitro and in vivo rumen fermentation characteristics and bacterial composition of a high-concentrate fed Hanwoo steers. Both BC0.9% and BC0.5% had significantly highest buffering capacity, pH, and ammonia-nitrogen (NH3-N) than BC0.3% and CON at 24 h incubation. Individual and total volatile fatty acids (VFA) were significantly lowest in CON. Phylum Bacteroidetes dominated all treatments but a higher abundance of Firmicutes in BC0.5% than others. Ruminoccocus bromii and Succiniclasticum ruminis were dominant in BC0.5% and Bacteroides massiliensis in BC0.3%. The normalized data of relative abundance of observed OTUs’ representative families have grouped the CON with BC0.3% in the same cluster, whereas BC0.5% and BC0.9% were clustered separately which indicates the effect of varying buffering capacity of buffer agents. Principal coordinate analysis (PCoA) on unweighted UniFrac distances revealed close similarity of bacterial community structures within and between treatments and control, in which BC0.9% and BC0.3% groups showed dispersed community distribution. Overall, the increasing buffering capacity enhances rumen fermentation parameters and affects rumen microbiome by altering bacterial community through distinct structure between high and low buffering capacity, thus an important factor to prevent ruminal acidosis during a high-concentrate diet.

This study aimed to explore the rumen bacterial community of yak in response to dietary supplements during the cold season. In addition, the rumen fermentation products were also analyzed. Twenty-one female domestic yaks were randomly divided into three groups i.e., pure grazing (GG) group, grazing plus oats hay supplement (OG) group, and grazing plus concentrate supplement group (CG). Rumen contents were collected after 90 days to assess rumen fermentation parameters and bacterial community. The GC group presented higher concentrations of ammonia nitrogen (P < 0.001), and total volatile fatty acids (TVFA) (P < 0.001), and lower rumen pH (P < 0.001) compared to other experimental groups. The CG group displayed higher proportions of propionate, butyrate, isobutyrate, and isovalerate while lower A/P ratio compared to other experimental groups. Shannon, Chao1, and ACE values were significantly lower in the OG group compared to GG and CG groups. Anosim test showed significant differences in bacterial community structure between groups but the PCA plot was not very informative to see these differences. Bacteroidetes, Proteobacteria, and Firmicutes were the three dominant phyla in all groups. The genera Oscillospira was more abundant in GG and OG groups. Higher relative abundance of Ruminococcus and Clostridium was observed in the GG group, while Ruminobacter, Corynebacterium, and Selenomonas were more abundant in the CG group. These findings will help in improving our understanding of rumen bacteria in yaks in response to changes in diet.

Motile Aeromonas Septicaemia disease (MAS) attacking tilapia has increased in recent years as a consequence of intensive aquaculture activities, which led to losses in aquaculture industry. The agent causing MAS disease is Aeromonas hydrophila. The disease can be controlled with the β-glucan. As immunostimulants, β-glucans can also increase resistance in farmed tilapia. Studies on the use of β-glucan extracted from baker's yeast Saccharomyces cerevisiae was intended to evaluate the non-specific immune system of tilapia that were challenged with Aeromonas hydrophila. The method used was an experimental method with a completely randomized design consisting of four treatments with three replicats. The dose of β-glucan used as treatments were 0 mg.kg-1 fish (Control), 5 mg.kg-1 fish (B), 10 mg.kg-1 fish (C) and 20 mg.kg-1 fish (D), each treatment as injected three times at intervals of 3 days, the injection volume of 0.5 ml/fish for nine days and resistance surveillance for seven days. The results showed that the difference in the amount of β-glucan and the frequency of the injected real influence on total leukocytes, phagocytic activity and resistance. Total leukocytes, phagocytic activity and resistance to treatment was best achieved by the administration of C a dose of  10 mg.kg-1 of the fish© Penyakit Motil Aeromonas Septicaemia (MAS) yang menyerang ikan nila mengalami peningkatan selama beberapa tahun terakhir sebagai konsekuensi dari kegiatan akuakultur intensif, yang menyebabkan kerugian dalam industri budidaya. Agen utama penyebab penyakit MAS adalah Aeromonas hydrophila. Untuk mengendalikan penyakit tersebut dapat dilakukan dengan pemberian β-glukan. Sebagai imunostimulan, β-glukan juga dapat  meningkatkan resistensi pada ikan nila yang dibudidayakan. Pengkajian mengenai pemanfaatan β-glukan yang diekstrak dari ragi roti Saccharomyces cerevisiae dimaksudkan untuk menguji sistem imun non spesifik ikan nila yang diuji tantang dengan bakteri Aeromonas hydrophila. Metode yang digunakan yaitu metode eksperimen dengan rancangan acak lengkap yang terdiri dari empat perlakuan dan tiga ulangan. Dosis β-glukan  yang digunakan sebagai perlakuan sebesar 0 mg.kg-1 ikan (Kontrol), 5 mg.kg-1 ikan (B), 10 mg.kg-1 ikan (C) dan 20 mg.kg-1 ikan (D), masing-masing perlakuan diinjeksi sebanyak 3 kali dengan interval waktu 3 hari selama 9 hari, volume injeksi 0,5 mL/ekor ikan dan pengamatan resistensi selama tujuh hari. Hasil penelitian menunjukkan perbedaan jumlah β-glukan dan frekuensi pemberian yang diinjeksikan memberikan pengaruh nyata terhadap total leukosit, aktivitas fagositosis dan resistensi. Total leukosit, aktivitas fagositosis dan resistensi terbaik dicapai pada perlakuan C dengan dosis 10 mg.kg-1 ikan©
