This study aimed to explore the rumen bacterial community of yak in response to dietary supplements during the cold season. In addition, the rumen fermentation products were also analyzed. Twenty-one female domestic yaks were randomly divided into three groups i.e., pure grazing (GG) group, grazing plus oats hay supplement (OG) group, and grazing plus concentrate supplement group (CG). Rumen contents were collected after 90 days to assess rumen fermentation parameters and bacterial community. The GC group presented higher concentrations of ammonia nitrogen (P < 0.001), and total volatile fatty acids (TVFA) (P < 0.001), and lower rumen pH (P < 0.001) compared to other experimental groups. The CG group displayed higher proportions of propionate, butyrate, isobutyrate, and isovalerate while lower A/P ratio compared to other experimental groups. Shannon, Chao1, and ACE values were significantly lower in the OG group compared to GG and CG groups. Anosim test showed significant differences in bacterial community structure between groups but the PCA plot was not very informative to see these differences. Bacteroidetes, Proteobacteria, and Firmicutes were the three dominant phyla in all groups. The genera Oscillospira was more abundant in GG and OG groups. Higher relative abundance of Ruminococcus and Clostridium was observed in the GG group, while Ruminobacter, Corynebacterium, and Selenomonas were more abundant in the CG group. These findings will help in improving our understanding of rumen bacteria in yaks in response to changes in diet.

Abstract This study determined the buffering capacity of buffer agents and its effect on in vitro and in vivo rumen fermentation characteristics and bacterial composition of a high-concentrate fed Hanwoo steers. Both BC0.9% and BC0.5% had significantly highest buffering capacity, pH, and ammonia-nitrogen (NH3-N) than BC0.3% and CON at 24 h incubation. Individual and total volatile fatty acids (VFA) were significantly lowest in CON. Phylum Bacteroidetes dominated all treatments but a higher abundance of Firmicutes in BC0.5% than others. Ruminoccocus bromii and Succiniclasticum ruminis were dominant in BC0.5% and Bacteroides massiliensis in BC0.3%. The normalized data of relative abundance of observed OTUs’ representative families have grouped the CON with BC0.3% in the same cluster, whereas BC0.5% and BC0.9% were clustered separately which indicates the effect of varying buffering capacity of buffer agents. Principal coordinate analysis (PCoA) on unweighted UniFrac distances revealed close similarity of bacterial community structures within and between treatments and control, in which BC0.9% and BC0.3% groups showed dispersed community distribution. Overall, the increasing buffering capacity enhances rumen fermentation parameters and affects rumen microbiome by altering bacterial community through distinct structure between high and low buffering capacity, thus an important factor to prevent ruminal acidosis during a high-concentrate diet.

The effects of glycerol with fat sources as a feed alternative were investigated in the ration for Nellore steers (Bos indicus). Eight cannulated steers at 30 months of age with initial bodyweight of 554 ± 36.0 kg were used in a double 4 by 4 Latin square design with four consecutive 16-day periods. Treatments were three different diets with lipid sources (soybean, soybean oil and calcium salts of fatty acids) and one diet control without lipid sources. All diets formulated contained 10% crude glycerol and 5% ether extract with a forage:concentrate ratio of 35:65; corn silage was used as forage. The ruminal liquid was sampled for 24 h and ruminal fermentations were monitored by measuring pH, concentrations of ammonia nitrogenand volatile fatty acids in rumen fluid. Urine samples were obtained from the total collection for 24 h for estimation of rumen microbial protein supply using urinary purine derivatives. Our results showed that the use of lipid sources combined with glycerol did not induce significant changes in rumen pH, acetate molar proportion, ruminal microbial protein or dry matter intake. Although the acetate molar proportion was kept constant within normal parameters, the propionate molar proportion was increased by the diet containing lipid sources. Moreover, we found that there was a negative effect of lipid sources on crude protein and neutral detergent fibre corrected for ash and protein intake, and one positive effect on ether extract intake. We believe that association between fat and glycerol may affect rumen fermentation parameters through reducing fibre intake and increasing propionate production and ammonia nitrogen.

Abstract Background: Rumen bacterial community is mainly affected by the type of diet consumed by the host animals. High concentrate diet increases the abundance of lactic acid producers and utilizers due to high level of non-structural carbohydrates thus reducing the number of fiber-degrading bacteria because of drastic decrease in pH. Dietary buffers are essential in regulating rumen pH through the compounds responsible in resisting drastic decrease in pH once cattle were fed with high-concentrate diet. However, no study has evaluated the effects of buffering capacity and efficiency in alleviating chronic acidosis in rumen. Ruminal metataxonomic and fermentation characteristics analyses were conducted to evaluate the effect of different buffering capacities on in vitro and in vivo experiments in high-concentrate fed Hanwoo steers. Results: Results revealed that BC0.9% and BC0.5% had similar and significant effect (P < 0.05) on in vitro ruminal fermentation at 3 to 24 h incubation. Both BC0.9% and BC0.5% had significantly highest (P < 0.05) buffering capacity, pH, and ammonia-nitrogen (NH3-N) than BC0.3% and CON at 24 h of incubation. Individual and total volatile fatty acids (VFA) were significantly lowest in CON. Increasing buffering capacity concentration showed linear effect on pH at 6 to 24 h while total gas and NH3-N at 3 and 12 h. This study was conducted in Grand station of Diwania/ Taj Al-Nahrain company- Al-Qadisiya Province for 84 days to investigate the effect of level of addition of commercial baker's yeast Saccharomyces cerevisiae,0, 15 and 30 g/cow/day to concentrate diet on rumen fermentation characteristics  in German Holstein Friesian lactating cows.  Nine multiparous Holstein cows in their mid-lactation were used. Daily feeding program applied in that station included offering 1 kg of concentrate diet per 3-4 kg milk produced divided into morning and evening meals, 10 kg of green forage, 10 kg whole plant corn silage and free choice of rice straw. Silage was characterized with accepted quality and fermentation parameters, values of pH, concentrations of ammonia nitrogen (NH3-N) and total volatile fatty acids (TVFA) were, 3.5, 0.98% of total nitrogen and 2.55 mM/DM respectively. Results revealed that addition of yeast at level of 15 and 30 g/cow/day reduced (P<0.01) concentration of NH3-N from 18.14 to 13.92 and 14.21 mg/100 ml respectively. Significant (P<0.01) increase in concentration of TVFA from 7.32 to 9.39 mM /100 ml was also observed due to addition of yeast at level of 15 g/cow/day. However, ruminal pH values were not affected by addition of yeast

AbstractEffects of time of supplementation on forage intake, nutrient apparent digestibility and rumen fermentation were studied with 12 mature castrated male sheep (wethers) offered grass hay from 16.30 h to 09.30 h and supplemented with a cereal-based concentrate given at either 09.30 or 16.00 h. Voluntary intake of hay organic matter (OM) was decreased by feeding the concentrate (P< 0·01). Offering concentrate at 09.30 h after hay was available, increased intake of hay and total OM compared with offering it at 16.00 h before hay was available (P< 0·05). Daily pattern of hay intake was not changed when concentrate was offered at 09.30 h compared with feeding hay alone, but concentrate given at 16.00 h resulted in a lower hay intake between 16.30 and 18.00 h. Sheep offered concentrate at 09.30 h had higher rumen ammonia levels than those offered concentrate at 16.00 h, but there were no differences in the apparent digestibility of the nutrients, rumen pH and molar proportions of the main volatile fatty acids. There were also no differences between groups in the blood plasma concentrations of 3-hydroxybutyrate (30HB), nonesterified fatty acids (NEFA) and glucose. It is concluded that feeding supplement after rather than before a period of intake of forage or a bout of grazing may offer a means of minimizing reduction of forage intake as a consequence of feeding concentrate.

Background and Aim: Crude glycerin is changed to propionate in the rumen, while chitosan can be used as a feed supplement to increase propionic acid concentration and decrease methane (CH4) production. We hypothesized that supplementation with a combination of a high level of crude glycerin with chitosan could have a beneficial effect on ruminal fermentation and mitigate CH4 production. This study aimed to explore the combined effects of crude glycerin and chitosan supplementation on nutrient digestibility, rumen fermentation, and CH4 calculation in native Thai bulls. Materials and Methods: Four 2-year-old native Thai bulls, weighing 150±20 kg, were kept in a 2×2 factorial arrangement in a 4×4 Latin square design. Factor A represented the incorporation of crude glycerin at 10.5% and 21% of the dry matter (DM) of a total mixed ration (TMR), and factor B represented the supplementation of chitosan at 1% and 2% DM of a TMR. Results: Increasing levels of crude glycerin at 21% decreased DM intake by 0.62 kg/day compared with 10.5% crude glycerin (p<0.05), whereas nutrient digestibility did not change (p>0.05). The incorporated crude glycerin and supplemented chitosan levels did not affect the pH, temperature, concentrations of ammonia-nitrogen, microbial population, and blood urea nitrogen (p>0.05). Supplemented chitosan and incorporated crude glycerin did not show any interaction effects on the molar portions and total volatile fatty acids (VFAs), except estimated CH4. Increasing the incorporated crude glycerin levels increased propionate and decreased the ratio of acetate to propionate ratio, whereas levels of butyrate, acetate, and total VFAs were unchanged. The combination of crude glycerin at 21% in the TMR with chitosan at 2% reduced CH4 estimation by 5.08% compared with the other feed treatment. Conclusion: Increasing incorporated crude glycerin levels in a TMR significantly elevated the propionate concentration, whereas combining 21% crude glycerin in the TMR diet with 2% chitosan supplementation could depress CH4 estimation more effectively than adding one of these supplements alone.

The goals of the current study were to investigate the effects of including chicory (Cichorium intybus L.) into the traditional feeding regime of ryegrass/white clover (Lolium perenne L./Trifolium repens L.), and time of its allocation on milk production, rumen fermentation, and FA composition of milk and rumen digesta of dairy cows. Nine groups of four cows were allocated one of three replicated feeding regimes: (1) ryegrass/white clover only (RGWC), (2) ryegrass/white clover + morning allocation of chicory (CHAM), and (3) ryegrass/white clover + afternoon allocation of chicory (CHPM). One cow per group had a rumen cannulae fitted. Treatment did not affect total grazing time or estimated dry matter intake, but cows ruminated more when fed RGWC than chicory. Allocating chicory in the afternoon elevated milk production compared with RGWC and CHAM. Milk from cows grazing chicory contained greater concentrations of polyunsaturated FA (PUFA) such as C18:3 c9, 12, 15 and C18:2 c9, 12 than those on RGWC. As with milk, rumen digesta concentration of PUFA increased when cows grazed on chicory rather than RGWC, which corresponded with lower concentrations of intermediate vaccenic and biohydrogenation end-product stearic acid for cows grazing on chicory. Mean ruminal pH was lower for cows offered chicory than those on RGWC, reflecting greater rumen concentrations of volatile fatty acids (VFA) for cows fed chicory. Allocating chicory during the afternoon is a useful strategy that can translate to improved milk production. The lower rumen pH, lower concentration of vaccenic and stearic acids, and elevated concentration of PUFA in the rumen of cows fed chicory suggest reduced biohydrogenation and may explain the elevated concentration of PUFA in the milk of cows fed chicory compared with those fed RGWC.

SUMMARYA comparative study of the effect of dietary nitrogen (N) content [Low: 11·0; Medium-Low (MLow): 16·7; Medium-High (Mhigh): 23·1; High: 29·2 N g/kg dry matter (DM)] on apparent digestibilities, rumen fermentation and N balance was conducted in coarse wool Tibetan sheep and Gansu Alpine fine-wool sheep at Wushaoling in the northeast of the Qinghai-Tibetan Plateau. It was hypothesized that responses would differ between breeds and that responses would favour Tibetan over fine-wool sheep at low N intakes. Eight wethers [four Tibetan sheep and four fine-wool sheep, 20–24 months old; body weight ± standard deviation was 52 ± 3·2 kg] were used in two concurrent 4 × 4 Latin square designs. Dry matter, organic matter, neutral detergent fibre and acid detergent fibre digestibilities were higher in Tibetan than fine-wool sheep when fed the Low, MLow and High N diets while N retention was higher when the animals were fed the Low and MLow N diets. Tibetan sheep had a higher rumen pH than fine-wool sheep; however, total volatile fatty acids were similar between breeds. Molar proportions of acetate were higher but propionate and butyrate lower in Tibetan than fine-wool sheep. In addition, Tibetan sheep had higher concentrations of ruminal free amino acid-N and soluble protein-N than fine-wool sheep. Plasma and saliva urea-N concentrations were higher in Tibetan than fine-wool sheep when supplied with the Low N diet. It was concluded that Tibetan sheep were better able to cope with low N feed than fine-wool sheep because of the higher N retention and higher DM and fibre digestibilities with Low and MLow diets.

AbstractThe effect of including 14 g urea with either 75 g dry matter (DM) from sugar cane molasses (UM) or Colombian rice polishings (RP) at three levels, 68 (RP1), 137 (RP2) or 203 (RP3) g DM on grass hay DM intake and on rumen fermentation was investigated. An incomplete Latin-square design was used and each experimental period was divided into 12 days for adaptation to each diet followed by 9 days in metabolism cages when all measurements were made using five adult sheep.Pooled mean values for rumen metabolites (five samples per day) were calculated. Rumen pH was not affected by the nature of the supplements. Concentrations of volatile fatty acids (VFA) (UM 92·8, RP1 84·2, RP2 86·4, and RP3 84·0 (s.e.d. 3·4) mmol/l) and lactate (UM 2·0, RP1 1/6, RP2 1/7, RP3 1/8 (s.e.d. 0·014) mmol/l) (UM v. RP, P < 0·05 and P < 0·10 respectively) were lower when RP were given, while concentration of branched and longer chain VFA (26·5, 34·0, 31·1 and 33·5 (s.e.d. 1·6) mmol/mol total VFA, UM v. RP, P < 0·01) and ammonia (98, 131, 141, 137 (s.e.d. 16·1) mg/l, UM v. RP, P < 0·05) were increased. Numbers of rumen protozoa (1·6, 3·2, 2·7, 3·3 (s.e.d. 0·75) × 105 per ml, UM v. RP, P < 0·20) tended to be higher 2 h after feeding when RP rather than UM were given. However, hay DM intake (1050, 960, 960, 880 (s.e.d. 45·3) g/day, UM v. RP, P < 0·05), and microbial protein supply (11·7, 9·3,11·1,10·8 (s.e.d. 0·59) g N per day, UM v. RP, P < 0·05), estimated from urinary purine excretion were reduced by feeding RP instead of UM.At the levels of inclusion tested RP did not increase the efficiency of rumen fermentation and were not as effective a supplement with urea as was molasses for a low-quality forage diet.

Abstract This study investigated the effects of substitution of whole corn silage (CS) with Broussonetia papyrifera silage (BPS) in different ratios on the serum indicators, hindgut fermentation parameters (pH, ammoniacal nitrogen, and volatile fatty acids), and fecal bacterial community of Holstein heifers. Sixteen heifers (8-month-old, 220±30 kg) were randomly divided into four treatments according to different BPS substitution ratios of feed basis (0%, 25%, 50%, and 75%). The experiment consisted of a 7-day preliminary feeding period and a 30-day experimental period. On the last day of the trial, the blood samples were collected from caudal vein, and the feces samples were collected from rectum. With the increasing of BPS content, the concentration of malondialdehyde (MDA) and interleukin-1β (IL-1β) in serum decreased (P<0.05), and the immunoglobulin A (IgA) and IL-4 content of serum increased (P<0.05); and the hindgut pH value increased (P<0.05). 16S rDNA sequencing found that the dominant phyla were Firmicutes, Bacteroidetes, and Verrucomicrobia; and the dominant genera were Ruminococcaceae_UCG-005, Ruminococcaceae_UCG-010, and Rikenellaceae_RC9_gut_group. Linear Discriminant Analysis Effect Size (LEfSe) analysis found 12 differential operational taxonomic units (OTUs) which have strong correlation with some serum and hindgut indicators. Phylogenetic Investigation of Communities by Reconstruction of Unobserved States (PICRUSt) found that BPS have impacts on the pathways, such as carbohydrate transport and metabolism, and promotes amino acid transport and metabolism. To sum up, inclusion of BPS in heifer diets can affect the fecal bacterial community, and further improve serum anti-oxidant and immune indicators in Holstein heifers.
