Aktivitas Sitotoksik dan Antiangiogenesis Ekstrak Fraksi Umbi Mentimun Papasan (Coccinia grandis (L.) Voigt) Terhadap Sel Kanker Payudara T47D Dan Chorio Allantoic Membrane (CAM) Embrio Ayam yang Diinduksi bFGFPenelitian ini bertujuan untuk mengetahui aktivitas sitotoksik dan antiangiogenesis ekstrak dan fraksi umbi mentimun papasan (Cocconia grandis (L.) Voigt) terhadap sel kanker payudara T47D dan Chorio Allantoic Membrane (CAM) embrio ayam yang diinduksi protein bFGF. Ekstrak diperoleh melalui metode maserasi dengan pelarut etanol 96%. Ekstrak etanol selanjutnya difraksinasi dengan metode partisi cair-cair. Fraksi yang diperoleh dilakukan analisis kualitatif dengan metode kromatografi lapis tipis (KLT). Dari analisis KLT menunjukan bahwa terdapat beberapa golongan senyawa aktif yaitu alkaloid, flavonoid, tanin, saponin dan terpen. Uji sitotoksik dilakukan menggunakan sel kanker payudara T47D dan sel VERO dengan metode MTT assay kemudian dibaca absorbansinya pada ELISA reader dengan parameter pengamatan nilai IC50. Uji antiangiogenesis dilakukan menggunakan metode Chorio Allantoic Membrane (CAM) embrio ayam yang diinduksi protein bFGF dengan parameter pengamatan jumlah pembuluh darah baru. Hasil penelitian menunjukan bahwa ekstrak, fraksi n-heksan, fraksi etil asetat, dan fraksi air memiliki nilai IC50 berturut-turut adalah 500,167; 353,131; 109,975; dan 303,990 µg/mL. Fraksi etil asetat memiliki nilai IC50 paling kecil sehingga dilanjutkan ke uji antiangiogenesis. Fraksi etil asetat memiliki aktivitas antiangiogenesis terhadap Chorio Allantoic Membrane(CAM) dengan konsentrasi efektif sebesar 219,95 µg/mL ditandai dengan persentase penghambatan angiogenesis sebesar 56,78%.Kata Kunci:  Antiangiogenesis; Chorio Allantoic Membrane (CAM); mentimun papasan (Cocconia Grandis (L.) Voigt) Base Towards T47D Breast Cancer Cells and Chorio Allantoic Membrane (CAM) of Chicken Embryo Induced by bFGF ABSTRACTThis study aims to identify the cytotoxic activity and antiangiogenesis of extract and fraction of (Coccinia Grandis (L.) Voigt) base towards T47D breast cancer cells and Chorio Allantoic Membrane (CAM) of chiken embryo induced by bFGF. The extract was extracted throught maceration method with ethanol 96% soluble. The ethanol extract was then fractionated by the liquid-liquid partition method. The fractions obtained were analyzed qualitatively by the thin layer chromatography (TLC) method. The TLC analysis shows that there are several classes of active compounds, namely alkaloids, flavonoids, tannins, saponons and terpenes. The ethanol extraction was then fractionated using the liquid-liquid extraction. The cytotoxic test was conducted using the T47D breast cancer cells and VERO cells with the MTT assay method which their absorbance were read on ELISA reader with the value of observation parameters IC50. The antiangiogenesis test was conducted using the Chorio Allantoic Membrane (CAM) method of chicken embryo induced with bFGF protein with the new blood vessels count as the observation parameters. The results showed that the extract, n-hexane fraction, ethyl acetate fraction, and water fraction had IC50 values of 500.167; 353.131; 109.975; and 303.990 µg/mL, respectively. Ethyl acetate fraction has the smallest IC50 value so it is continued to the antiangiogenesis test. Ethyl acetate fraction had an antiangiogenesis activity towards Chorio Allantoic Membrane (CAM) with the effective concentration 219,95 µg/mL marked by the angiogenesis inhibitors percentage of 56,78%.Keywords: Antiangiogenesis; Chorio Allantoic Membrane (CAM); Coccinia grandis (L.) Traditionally, some People from bangka belitung has consumed Eriocaulon cinereum R.Br as an anticancer drug. Aims of this study is to determine the potential of cytotoxicity of Fraction of Ethyl Acetate and Dichlorometane against T47D cell. Fractionation using Vacuum Liquid Chromatography (VLC) method. MTT assay method was used on evaluating the citotoxic activity. The fraction of dichloromethane performed high activity as a chemopreventive agent with IC50 score was 131,921 µg/ml to T47D and 413,042 µg/ml cells to vero cell whereas for ethyl acetate fraction performed lower activity as chemopreventive agent with IC50 score was 531,808 µg/ml to T47D and 679,114 µg/ml to vero cell). Phytochemical tests showed that the dichloromethane fraction contains phenolic, steroid and terpenoid compounds. While ethyl acetate fraction contains alkaloids, flavonoids and terpenoids. These compounds have activity on cancer cells through apoptotic mechanisms. The results showed the excellent potential of this plant in killing breast cancer cells.

Cancers are diseases that can cause death and breast cancer is one of the prevalence cancers. Conventional treatments have been used to treat cancer. However, these treatments produced inefficient effects and low survival rate. Therefore, most cancer patients began to consume complementary and utilized alternative treatments to alleviate their pain. This study is to identify the cytotoxicity effect of methanolic leaves extract Typhonium flagelliforme and Clinacanthus nutans on breast cancer cells (MDA-MB-231). MTT assay has been used to determine the cytotoxicity effect of both plants on MDA-MB-231 cells and CHO cells (non-cancerous cell), which acted as the positive control cells. Result revealed that T. flagelliforme extract has shown higher cytotoxic effect on MDA-MB-231cells (IC50: 0.11 mg/mL) compared to C. nutans extract (IC50: 0.17 mg/mL). Then, the IC50 values of T. flagelliforme and C. nutans extract on CHO cells were 0.10 mg/mL and 0.24 mg/mL respectively. Based on these results, T. flagelliforme represented higher toxicity effect on CHO compared to the MDA-MB-231 cells. Thus, in future CHO cell (as a positive control) can be replaced with a normal breast cell line such as HCC1395 (epithelial mammary duct of normal breast cells) to understand clearly the toxicity effect of T. flagelliforme extract towards the normal breast cell. Moreover, identification of potential compounds that can inhibit MDA-MB-231 cells growth  is also important for future research.
