Research on indirect organogenesis of cucumber apple (Cucumis sp.) was conducted from March to May 2020. This study aims to find the best combination between Benzyl Amino Purine (BAP) and Naphthalene Acetic Acid (NAA) on the growth of cucumber apple (Cucumis) sp.) on Gamborg media (B5) by organogenesis. The callus used came fromcucumber seeds explants with a concentration of NAA 2,0 mg/l + BAP 0,2 mg/l using Murashige and Skoog media. The research method used is an experimental method with nonparametric statistics with 24 treatments repeated three times and analyzed descriptively using the Kruskal Wallis test. The results showed that the best treatment of cucumber apple shoots was the combination of NAA and BAP, namely the B4N2 treatment (2,5 ppm BAP + 0,20 ppm NAA) with shoot height (0,45 cm). At a single concentration, a concentration of 1 ppm BAP was able to provide a faster effect on shoot formation time, namely at 24 days after initiation (hsi) and the best response to shooting height (0,5cm).

This study presented the elimination of the glochids in the areoles of Opuntia ficus-indica cladode induced the shoot formation from the explants containing the shoot apical meristem (SAM) on Murashige and Skoog medium (MS) supplemented with 6-benzylaminopurine (BA) 5 mg/L. Morphological and physiological changes in shoot formation process were analyzed. This process involved the constant stages: activation of cell division, initiating of meristematic region; formation of shoot primordium and shoot with leaves. The SAM position in the upper front of the cladode gave the highest shoot productivity. This position had the photosynthesis rate, respiration rate, endogenous indole acetic acid (IAA) and zeatin activity higher than the others. Difference of auxin concentrations increased the amount of shoots and shoot height. The maximum number of shoots per explant was obtained on MS medium supplemented with BA 5 mg/L and 1-naphthalene acetic acid (NAA) 0.5 mg/L. The elimination of SAM by removing the surface of the shoot gave the highest number of the shoot. The correlation of the glochids in the areoles, explant position, plant hormones, photosynthesis rate respiration rate and shoot formation were discussed.

Salah satu alternatif untuk perbanyakan anggrek hitam (Coelogyne pandurata Lindl.) adalah melalui multiplikasi tunas anggrek secara in vitro. Tujuan penelitian adalah untuk membuktikan pengaruh pemberian NAA (Naphthalene Acetic Acid) dan ekstrak biji jagung (Zea mays) terhadap multiplikasi tunas anggrek hitam. Penelitian dilakukan di Laboratorium Kultur Jaringan Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Tanjungpura Pontianak. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola faktorial dengan dua faktor perlakuan. Faktor pertama adalah NAA terdiri dari 5 taraf konsentrasi yaitu A0 (0 M/ kontrol) A1 (10-7 M), A2 (10-6 M), A3 (5x10-7 M) dan A4 (5x10-6 M ) dan faktor ekstrak biji jagung (B) dengan 5 taraf konsentrasi yaitu B0 (0%), B1 (2,5%), B2 (5%); B3 (7,5%) dan B4 (10%). Pemberian kombinasi NAA dan ekstrak biji jagung berpengaruh nyata terhadap semua parameter yaitu jumlah tunas, jumlah daun, dan tinggi tunas. Hasil terbaik rerata jumlah tunas pada perlakuan A4+B4 yaitu 5x10-6M NAA+10% ekstrak biji jagung. Hasil terbaik pada rerata jumlah daun pada perlakuan A2+B2 yaitu 5x10-7M NAA+5% ekstrak biji jagung dan hasil terbaik pada rerata tinggi tunas pada perlakuan A1+B1 yaitu 10-7M NAA+2,5% ekstrak biji jagung.Kata Kunci: multiplikasi; tunas anggrek hitam; ekstrak biji jagung; NAA. ABSTRACTGenerative reproduction of orchid plants it takes a requires the help of mycorriza mushrooms for seed germination, whereas conventional propagation business takes a long time to obtain large quantities of plants. One alternative to the propagation black orchids (Coelogyne pandurata Lindl.) is required through tissue culture techniques. The purpose of this study is to find the influence and concentration corn seed extract (Zea mays) and NAA (Naphthalene Acetic Acid) on the multiplication black orchids. This research was conducted in the tissue culture laboratory Biology Department Faculty of Mathematics and Natural Sciences Tanjungpura University Pontianak. The study used a Complete Randomized Design (RAL) of factorial patterns with two treatment factors. The first factor is that the NAA consists of 5 concentration levels  A0 (0 M) A1 (10-7 M), A2 (10-6 M), A3 (5x10-7 M) and A4 (5x10-6 M ) and the second factor is that corn seed extract of 5 levels concentratio B0(0%), B1 (2,5%), B2 (5%); B3 (7,5%) and B4 (10%). The administration NAA and corn seed extract in combination has a real effect on all parameters namely the number shoots, the number leaves, and the height shoots. The best results where the average number of shoots in the treatment of A2+B2 namely 5x10-6M NAA + 10% corn seed extract. The best results average number of leaves in the treatment  A2+B2 namely 5x10-7M NAA + 5% corn seed extract and in the best results for shoot height in the treatment of A1+B1 namely 10-7M NAA + 2.5% corn seed extract.Keywords: Multiplication; black orchid’s shoot; corn  seed extract; NAA

ABSTRACT  The objective of the study was to obtain the best 2,4-D concentration on callus induction of the banana flowers in banana propagation using indirect organogenesis method. Kesuna, local banana cultivar obtained from Sembung Gede, Tabanan was used as explant material. Callus induction was performed using 2,4-Dichlorophenoxyacetic acid with concentration of 0; 0.5; 1.0; 1.5 and 2.0 ppm. Each treatment was represented by 3 bottles and each bottle was planted with 3 explants, so each treatment was represented by 9 explants of banana flowers. The results showed that the concentration of 2.0 ppm 2.4-D induced callus with the fastest time and gave the highest percentage of the explants producing callus. The calluses were subsequently subcultured into regeneration medium using 0.5 mg/L Benzylaminopurine (BAP) and 0.005 mg/L Napthaleneaceticacid (NAA). The calluses were subsequently sub-cultured into a regeneration medium using 0.5 ppm (BAP) and 0.005 ppm Naphthalene acetic acid (NAA) to induce shoots and roots and performed plantlets.   Keywords: 2,4-Dichlorophenoxyacetic acid, banana’s flowers, callus

An efficient protocol is outlined for rapid and mass micropropagation of Ficus carica L. (fig). Shoot tips (5 mm) were obtained from mother plants stock grown on half strength Murashige and Skoog (½ MS) medium with the addition of 30 g/L sucrose. For shoot multiplication Benzyl amino purine (BAP) and kinetin produced differences number of new shoot per plant and shoot height. BAP at 0.4 mg/L in combination with 0.2 mg/L indole-3-butyric-acid (IBA) produce maximum in vitro propagation rate, with 4.2 shoots per ex-plant. Root initiation was experimented on MS medium containing different concentrations of mg/L, IBA, IAA (Indole-3-acetic-acid) (IAA) or Naphthalene acetic acid (NAA). Highest number of root (4.3) was resulted when 1.5 mg/L IBA was used. After acclimatization in a mixture of (1 soil: 1 perlite: 1 peat) survival rate of 80% was achieved. For in vitro conservation of F. carica was experimented as microshoots were stored for 40 weeks on MS medium containing different sucrose concentration. Medium supplemented with 3% sucrose gave the highest regrowth (89%) at 24 ± 2 °C. Culture grew slowly when transferred to new fresh medium after the storage periods.

Protocols of axillary bud multiplication were established for Dodonaea viscosa (Sapindaceae). Murashig & Skoog’s medium with 0.2 mg/l BAP with 0.01 Naphthalene acetic acid induced high rate of shoot induction and an average of five shoots per node. Subsequent culture enhanced the number of shoots. Callus initiated from the basal cut end explants differentiated in to more than 10 shoots on MS Medium with 0.4mg/l Benzylaminopurine and 0.02mg/l Indole Butric Acid. Eighty per cent of the rooted shoots survived when transferred to greenhouse and subsequently to the field.

We have initiated in vitro propagation the Caryopteris mongolica Bge. from seeds. Seeds were surface sterilized by immersing in 70% ethanol for 90 sec, 10% hydro peroxide for 10 minutes and 5% sodium hypochlorite for 20 minutes. Finally, rinsed with sterile distilled water for 4 times. Sterilized seeds were germinated on MS medium supplemented with 0.5, 1, 2 mg/l gibberillic acid, respectively. Nodal segment and cotyledon explants from 4 weeks old seedling were cultured on the MS basal medium supplemented with benzylaminopurine(BAP) 1 mg/l, BAP 2 mg/l and Indole acetic acid(IAA) 0.3 mg/l, BAP 3 mg/l, Kinetin(KIN) 1 mg/l, KIN 2 mg/l and Naphthalene acetic acid(NAA) 0.3 mg/l, KIN 3 mg/l and NAA 0.4 mg/l concentrations, respectively. After 3 weeks, measured of shoot height and bud number and 4 weeks later subcultured in a same medium. High frequency multiple buds were formed on MS medium supplemented with BAP 3mg/l, from cotyledon explants, but KIN 3 mg/l and NAA 0.4 mg/l was best version of nodal segment explants. All shoots were cultured in the ½ MS basal without any growth regulators for induction of root. Research with the title shoot multiplication shoots tomatoes (Lycopersicumesculentum mill) using the Benzyl Amino Purine (BAP) dan Naphtalene Acetic Acid(NAA) in vitro have been conducted at the Laboratory of Biotechnology Faculty ofAgriculture, Islamic University of Riau, Pekanbaru. This research has been carried outfor three months carried out startingfrom November 2010 to February 2011. This studyaims to determine the effect of a single interaction between the administration and plantgrowth regulators BAP and NAA on shoot multiplication of in tomato shoots vitro.Rancangan used in this study was completely randomized design (CRD) in factorialwhich consist of two factors. The first factor is factor B (concentration of BAP) withfour standard treatments are: B0 (0 ppm), B1 (1 ppm), B2 (2 ppm), B3 (3 ppm).Thesecond factor is the factor (the concentration of NAA) with four standard treatments,including: N0 (0 ppm), N1 (0.1 ppm), N2 (0.5 ppm), and N3 (1 ppm), to obtain 16combined treatment with three replications. Parameters observed, namely: age emergedshoots, number of shoots, shoot height, the percentage of growing shoots, roots andgrowing percentage of the number of explants forming callus. The data was statisticallyanalyzed the results of observations, when the F calculated is greater than the F table,followed by a further test of honest real difference (HRD) 5%. From the results ofresearch in the interaction of BAP and NAA effect on the parameters of high-shoots bytreatment tebaik B1N0 namely 6.16 cm. BAP singly significantly affect the parametersage appears buds (days) and the percentage grows shoots with the best treatment B2 (2ppm), shoot height with the best treatment B0 (0 ppm), the number of shoots (the fruit)with the best treatment B3 (3 ppm). singly whereas NAA significantly affect theparameters age appears shoots (day) and high-shoots with the best treatment N0 (0ppm), and the percentage grows roots with the best treatment N3 (1 ppm).

Globba leucantha var. bicolor Holttum is a part of Zingiberaceae plant that is not yet widely known but can be used as a medicinal material as well as an ornamental plant. So far Globba plant propagation has not been widely done and has relied solely on from its habitat. Research aims at acquiring Naphthalene Acetic Acid (NAA) concentrations and optimal kinetin for the multiplication of ginger plant shoots (Globba leucantha var. bicolor Holttum) on in vitro culture. The research method uses experimental methods with non-parametric statistics and 24 repeated treatments 3 times as well as being analyzed descriptively using the Kruskal Wallis Test. The treatment used NAA with concentrations of 0 mg/l, 0.1 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l, and 2 mg/l combined with Kinetin with concentrations of 0 mg/l, 0.1 mg/l, 1 mg/l and 10 mg/l. The results showed that the growth of Globba plant's best shoots at N3K3 treatment (NAA 1 mg/l + Kinetin 10 mg/l) with a shoot emergent time of 21.58 HSI, a percentage of shoot forming 66.67%, a shot count of 0.89 buds per explant, and a shoot length of 0.18 cm. Additionally at the same time also formed Globba plant roots with the best treatment of N5K2 (NAA 1 mg/l + Kinetin 10 mg/l) with a time of appearing rooted 25.67 HSI and a root count of 7.78 roots per explant.

Basils are mainly used as food flavoring and source of aromatic oil, but the leaves have also been used for herbals and cosmetics. Basil is propagated by seeds but the germination is often low in the field. This study aims to investigate the effect of different concentrations of NAA (Naphthalene Acetic Acid) and BAP (Benzyl Amino Purine) to induce the adventitious shoot proliferation of three species of basil (Ocimumsp. L.) in vitro. The research was conducted at the Tissue Culture Laboratory II of the Department of Agronomy and Horticulture, Bogor Agricultural University (IPB) from November 2016 till October 2017. This research consisted of three separate experiments with one basil species for each experiment, Lemon Basil, “Greek Minette”, and “Thai Siam Queen”. The experiments were organized in a completely randomized block design with two factors: NAA concentrations (0, 0.5, and 1 mg.L-1) and BAP (0, 1, 2, and 3 mg.L-1) using Murashige and Skoog (MS) medium supplied with 25 ml.L-1 coconut water. The three basil species responded differently to NAA and BAP in the media. The optimum concentration of NAA and BAP to induce callus growth in Lemon Basil was 0.5 mg.L-1 and 3 mg.L-1, respectively, with BAP alone showing the best effect on proliferation. In “Greek Minette”, however, the addition of NAA and BAP had no significant effect on the callus growth, proliferation, height, and number of leaf, node and root per explant. Media without growth regulators provide the best results in increasing height, explant proliferation rate, number of nodes and shoots, and percentage of proliferating shoots of “Thai Siam Queen”.Keywords: cytokinin, “Greek Minette”, Lemon Basil, “Thai Siam Queen”, synthetic auxin

The influence of growth regulators, explants and their interactions on in vitro shoot bud formation from callus was studied in <I>Dianthus caryophyllus</I> L. The leaf and internode explants were cultured on Murashige and Skoog (MS) medium containing different concentrations of growth regulators. The highest callus induction was observed with 2 mg/l 2,4-dichlorophenoxy acetic acid (2,4-D) and 1 mg/l benzyl adenine (BA). Out of twenty seven shoot regeneration media tested, only 2 mg/l thidiazuron (TDZ) and zeatin alone or in combination with naphthalene acetic acid (NAA) and/or indole acetic acid (IAA) could differentiate calli. The highest average number of shoots was observed with 2 mg/l TDZ and 1 mg/l IAA. Significant differences were observed in calli producing shoots and number of shoots per callus in the explants of leaf and internode. The shoots were elongated and multiplied on MS medium supplemented with 1 mg/l BA and solidified with 1% agar. The shoots were rooted and hardened with 76% survival success in pots after six weeks of transfer to the pots.
