Basils are mainly used as food flavoring and source of aromatic oil, but the leaves have also been used for herbals and cosmetics. Basil is propagated by seeds but the germination is often low in the field. This study aims to investigate the effect of different concentrations of NAA (Naphthalene Acetic Acid) and BAP (Benzyl Amino Purine) to induce the adventitious shoot proliferation of three species of basil (Ocimumsp. L.) in vitro. The research was conducted at the Tissue Culture Laboratory II of the Department of Agronomy and Horticulture, Bogor Agricultural University (IPB) from November 2016 till October 2017. This research consisted of three separate experiments with one basil species for each experiment, Lemon Basil, “Greek Minette”, and “Thai Siam Queen”. The experiments were organized in a completely randomized block design with two factors: NAA concentrations (0, 0.5, and 1 mg.L-1) and BAP (0, 1, 2, and 3 mg.L-1) using Murashige and Skoog (MS) medium supplied with 25 ml.L-1 coconut water. The three basil species responded differently to NAA and BAP in the media. The optimum concentration of NAA and BAP to induce callus growth in Lemon Basil was 0.5 mg.L-1 and 3 mg.L-1, respectively, with BAP alone showing the best effect on proliferation. In “Greek Minette”, however, the addition of NAA and BAP had no significant effect on the callus growth, proliferation, height, and number of leaf, node and root per explant. Media without growth regulators provide the best results in increasing height, explant proliferation rate, number of nodes and shoots, and percentage of proliferating shoots of “Thai Siam Queen”.Keywords: cytokinin, “Greek Minette”, Lemon Basil, “Thai Siam Queen”, synthetic auxin

Coleus tuberosus, also known as black potato, is one of the Indonesian local tubers consumed as a carbohydrate substituent. Therefore, this study aimed to examine the effect of processing and cooling methods on the in vitro digestibility of black potato starch. Furthermore, two factors Randomized Block Design with a 2x3 experimental design was used, which consisted of processing methods (boiling, roasting, and microwave) and cooling at room temperature and 4 °C for 24 hours with 3 repetitions. Black potato flour was compared with the raw form, by assessing some parameters, namely Resistant Starch (RS), Slowly Digestible Starch (SDS), Rapidly Digestible Starch (RDS), and Glycemic Index (GI). Also, the analysis of total starch, moisture, and color was performed, hence raw black potatoes generally have 10% resistant starch (%wb). Different treatments of cooking and cooling had a significant effect (α = 0.05) on moisture content, total starch, RS, RDS, SDS, GI, brightness (L), and yellowness (b). Black potatoes subjected to the processing method followed by cooling had lower RDS and increased RS content. Furthermore, refrigeration at 4°C for 24 hours reduced the digestibility of black potato starch more than cooling at room temperature. Contrarily, microwaved black potato cooled at room temperature showed a higher digestion rate compared to the raw counterpart. Conclusively, processing followed by cooling reduces the GI and increases the RS content of Coleus tuberosus.

Two different methods of in vitro multiplication of Rauwolfia serpentina from nodal explants were compared viz. multiplication via callus morphogenesis and that via shoot proliferation from axillary buds. The second method was found to be far better. The optimum shoot proliferation occurred on Murashige and Skoog (MS) medium supplemented with 1 mg/L naphthalene acetic acid (NAA) and 2 mg/L of benzyl aminopurine (BAP). The best rooting of shoots occurred on MS medium containing 4% sucrose and 1 mg/L of NAA. Solid and liquid MS media were found to be similar in supporting shoot proliferation. The plants produced were successfully hardened and established in soil. An easy, reliable and reproducible protocol was developed for in vitro micropropagation of Rauwolfia serpentina from nodal explants.

The research was conducted at the cutting red bettle (Piper crocatum Ruiz and Pav.) to determined the effect of (1) the difference growth of cuttings of red bettle which are given IBA and without IBA, (2) the concentration of NAA on growth of cutting red bettle, (3) the concentration of NAA on growth of cutting red bettle on each given IBA. The treatment was arranged in factorial (2 x 4) in randomized block design with three replication. The first factor were without IBA (A) and the given of 1000 ppm IBA (A1). The second factor were the concentration of NAA consists of: 0 ppm (B), 1000 ppm (B1), 2000 ppm (B2), and 4000 ppm (B). The results showed that the NAA concentration of 4000 ppm produced the most number of roots on either at the node or at the base of cutting. Planting cutting red bettle which given IBA 1000 ppm was able to accelerated the time leaves open and increased the number of cutting which germinate. The mixtured of giving IBA 1000 ppm and the concentration of NAA 4000 ppm, produced the most number of cutting that germinated.

Three spineless phenotypes of Acacia wrightii G. Bentham ex A. Gray were identified with aesthetic landscape potential. Experiments in seed, cutting, grafting, and tissue culture propagation were undertaken to perpetuate this desired spineless phenotype. Germination percentages for mechanically scarified seeds ranged from 33% to 94%, however yield of spineless seedlings was low (0% to 34%). Sulfuric acid scarification for 10, 20, 30, or 60 minutes hastened and unified germination compared to nontreated seeds by 7 to 8 days. Vegetative propagation was successful for softwood cuttings. Rooting measures increased with auxin (2:1 indole butyric acid to naphthalene acetic acid) concentrations from 0 to 15000 mg·L–1, with maximum rooting percentage (70%), root number (9.2), and root length (12.4 cm) per softwood cutting at 15000 mg·L–1 auxin 8 weeks after treatment. Rooting was not successful for semi-hardwood or hardwood cuttings. Whip-and-tongue or T-bud grafting was not successful. Tissue culture of shoots from in vitro germinated seedlings indicated that shoot proliferation was greatest in Murashige and Skoog (MS) medium with 15 μm zeatin. The number of shoots that rooted in vitro increased with increasing concentrations of indole-3-butyric acid from 0 to 25 μm.

Research with the title shoot multiplication shoots tomatoes (Lycopersicumesculentum mill) using the Benzyl Amino Purine (BAP) dan Naphtalene Acetic Acid(NAA) in vitro have been conducted at the Laboratory of Biotechnology Faculty ofAgriculture, Islamic University of Riau, Pekanbaru. This research has been carried outfor three months carried out startingfrom November 2010 to February 2011. This studyaims to determine the effect of a single interaction between the administration and plantgrowth regulators BAP and NAA on shoot multiplication of in tomato shoots vitro.Rancangan used in this study was completely randomized design (CRD) in factorialwhich consist of two factors. The first factor is factor B (concentration of BAP) withfour standard treatments are: B0 (0 ppm), B1 (1 ppm), B2 (2 ppm), B3 (3 ppm).Thesecond factor is the factor (the concentration of NAA) with four standard treatments,including: N0 (0 ppm), N1 (0.1 ppm), N2 (0.5 ppm), and N3 (1 ppm), to obtain 16combined treatment with three replications. Parameters observed, namely: age emergedshoots, number of shoots, shoot height, the percentage of growing shoots, roots andgrowing percentage of the number of explants forming callus. The data was statisticallyanalyzed the results of observations, when the F calculated is greater than the F table,followed by a further test of honest real difference (HRD) 5%. From the results ofresearch in the interaction of BAP and NAA effect on the parameters of high-shoots bytreatment tebaik B1N0 namely 6.16 cm. BAP singly significantly affect the parametersage appears buds (days) and the percentage grows shoots with the best treatment B2 (2ppm), shoot height with the best treatment B0 (0 ppm), the number of shoots (the fruit)with the best treatment B3 (3 ppm). singly whereas NAA significantly affect theparameters age appears shoots (day) and high-shoots with the best treatment N0 (0ppm), and the percentage grows roots with the best treatment N3 (1 ppm).

Abstract The availability of nitrogen and sulfur in the rumen should be synchronized to generate microbial protein synthesis. This study aimed to evaluate rumen degradable protein and sulfur supplementation on in vitro digestibility and ruminal fermentation. The experimental design was a 4 x 3 factorial randomized block design. Factor 1 was RDP levels (%CP) (R1= 60%; R2= 55%; R3= 50%; R4= 45%), and factor 2 was sulfur supplementation (S1= 0%; S2= 0.1%; S3= 0.2%). Data were analyzed with ANOVA followed by the Duncan test. The result showed that sulfur supplementation improved DMD and OMD without changes in rumen pH. NH3 concentration was influenced by RDP levels. Treatment R1 had the highest NH3 concentration (7.03 mM). There was an interaction of two factors on total VFA concentration. The combination of R1 and S2 resulted in the highest total VFA concentration than others. The total bacteria population increased with increasing RDP levels. Protozoa population was affected by RDP levels and sulfur supplementation. This study concluded that a combination of 60% RDP and 0.2% sulfur supplementation resulted in the best ration to improve ruminal fermentation and digestibility.

Research on indirect organogenesis of cucumber apple (Cucumis sp.) was conducted from March to May 2020. This study aims to find the best combination between Benzyl Amino Purine (BAP) and Naphthalene Acetic Acid (NAA) on the growth of cucumber apple (Cucumis) sp.) on Gamborg media (B5) by organogenesis. The callus used came fromcucumber seeds explants with a concentration of NAA 2,0 mg/l + BAP 0,2 mg/l using Murashige and Skoog media. The research method used is an experimental method with nonparametric statistics with 24 treatments repeated three times and analyzed descriptively using the Kruskal Wallis test. The results showed that the best treatment of cucumber apple shoots was the combination of NAA and BAP, namely the B4N2 treatment (2,5 ppm BAP + 0,20 ppm NAA) with shoot height (0,45 cm). At a single concentration, a concentration of 1 ppm BAP was able to provide a faster effect on shoot formation time, namely at 24 days after initiation (hsi) and the best response to shooting height (0,5cm).

ABSTRAK. Penggunaan umbi benih untuk bahan perbanyakan bawang merah mempunyai beberapa masalah, antara lain ketidaktersediaan benih bermutu, produktivitas rendah, dan mahal. Salah satu alternatif teknologi yang potensial untuk dikembangkan sebagai benih ialah penggunaan biji botani atau true shallots seed (TSS).  Penelitian dilakukan untuk memproduksi benih bawang merah TSS di dataran tinggi melalui peningkatan pembungaan dan viabilitas serbuk sari menggunakan BAP dan boron.  Penelitian dilakukan di Kebun Percobaan Balai Penelitian Tanaman Sayuran Lembang, Jawa Barat (ketinggian 1.250 m dpl.) dari Bulan Agustus 2011 sampai dengan Februari 2012. Rancangan percobaan yang digunakan yaitu acak kelompok faktorial dengan tiga ulangan.  Perlakuan terdiri atas dua faktor, yaitu aplikasi benzil amino purin (BAP) 0, 50, 100, 150, dan 200 ppm dan boron 0, 1, 2, 3, dan 4 kg/ha.  Aplikasi  BAP diberikan tiga kali pada umur 1, 3, dan 5 minggu setelah tanam (MST), dan boron pada umur 3, 5, dan 7 MST. Hasil penelitian menunjukkan bahwa aplikasi BAP dapat meningkatkan pembungaan, viabilitas serbuk sari bawang merah, dan produksi benih TSS tetapi tidak meningkatkan mutu benih, sedangkan aplikasi boron efektif meningkatkan semua variabel yang diamati termasuk mutu benih TSS. Konsentrasi BAP yang optimum untuk menghasilkan produksi benih TSS ialah 37,5 ppm, sedangkan dosis boron yang optimum untuk menghasilkan bobot benih per plot yang tinggi dengan mutu benih sesuai standar sertifikasi mutu yaitu 2,88 kg/ha.  Hasil yang diperoleh pada perlakuan boron memberikan peningkatan sebesar 165,69% daripada kontrol. Hasil penelitian ini memberikan informasi teknologi produksi TSS yang dapat dikembangkan untuk memproduksi benih TSS bermutu tinggi. <br /><br />ABSTRACT. Rosliani, R, Palupi, ER, and Hilman, Y 2012. The use of bulb for propagation material of shallots has several problems including unavailability of quality seeds, low productivity, and expensive. One of the potential alternative technologies to be developed as seed is using true shallots seed (TSS). The aimed of research was to produce TSS in the highlands through increased flowering and pollen viability by using benzyl amino purine (BAP) and boron. The study was conducted at the Experimental Field, Indonesian Vegetable Research Institute (IVEGRI) in Lembang, West Java (altitude 1,250 m asl.), from August 2011 to February 2012. The factorial experiment was arranged in a randomized block design with three replications. The treatments consists of two factors, namely the application of BAP 0, 50, 100, 150, and 200 ppm and boron (0, 1, 2, 3, and 4 kg/ha). Benzyl amino purine application was given three times at  1, 3, and 5 weeks after planting (WAP), and boron at  3, 5, and 7 WAP. The results showed that application of BAP improved flowering, pollen viability, and seed production of TSS but did not improve seed quality, while boron application effectively increased all variables including the seed quality of TSS. The efficient concentration of BAP to improve TSS seed weight per plot was 37.5 ppm, while the optimum concentration of boron to improve TSS seed production with the good seed quality (according to the certification standards of seed quality) was 2.88 kg/ha. The yield obtained in the treatment of boron gave an increase of 165.69% compared to the control. The results provide information about TSS production techniques that can be developed to produce  the high seed quality of TSS.<br /><br />

This study examined the effects of plant growth regulators, explant types, and their orientations on in vitro shoot proliferation of Casuarina cunninghamiana Miq. and also the subsequent rooting ability of shoots. Results showed that shoot proliferation occurred only in shoot tip explants cultured vertically on Murashige and Skoog (MS) medium supplemented with 2 or 4 μM thidiazuron (TDZ). Neither 6-benzylaminopurine alone nor in a combination with 1-naphthalene acetic acid (NAA) or gibberellic acid had any effect on shoot proliferation. TDZ at 4 μM resulted in the greatest percentage of axillary bud sprouting (70%) and mean number of sprouts per explant (2.3). Additionally, no shoot proliferation was observed from detipped or single-node explants or from horizontally placed shoot tip explants when cultured on the same TDZ-containing medium. The induced shoots produced adventitious roots on MS medium supplemented with 2.5, 5, or 10 μM indole-3-butyric acid (IBA), not with indole-3-acetic acid and NAA. Although the mean number of roots per explant was not significantly different between 2.5 and 5 μM IBA, the highest rooting percentage (68%) and mean length of roots per explant (0.7 cm) was achieved at 5 μM IBA. The current study provided preliminary information toward commercial in vitro propagation of Casuarina cunninghamiana male plants.

The study aimed to determine the effect of the combination of city waste compost and cattle manure on the growth and yield of pakcoy plants (Brassica rapa, L. Chinensis) in Aluvial, Indramayu Regency. The study was conducted at the Experimental Garden of the Faculty of Agriculture, Wiralodra University, Indramayu Regency from March to August 2018. This study used Factorial Pattern Randomized Block Design with two factors. The first factor is city waste compost and the second factor is cattle manure with 4 levels: 0%, 3.0%, 6.0%, and 9.0%, repeated twice. The results showed that there was an independent influence between city waste compost and cattle manure on plant height and number of leaves. The best combination to increase the feasibility weight of pakcoy consumption is by giving city waste compost 9.0% (S4) and cattle manure 6.0% (K3).
