Comparison of seminal characteristics of Aberdeen Angus, Holstein and Nelore bulls before and after cryopreservation

The aim of this study was to evaluate parameters indicative of sperm quality of fresh and post-thawed semen of Aberdeen Angus, Holstein and Nelore bulls. Thirty-nine bulls were used: Aberdeen Angus (n=13), Holstein (n=13) and Nelore (n=13). The ejaculate collects were performed twice a week using artificial vagina, totaling 792 semen collections, 307 for Aberdeen Angus, 225 for Holstein and 260 for Nelore bulls. After collection, fresh semen was evaluated and semen freezing was performed. After freezing, the batches were thawed and progressive motility was determined. The analysis of fresh semen showed that there was no difference (P = 0.053) between the Aberdeen Angus and Nelore breeds, while ejaculates from Holstein bulls showed a statistical difference (P = 0.024). As well, a difference (P<0.001) was identified in the sperm concentration of the three breeds. In the samples evaluated after thawing, a statistical difference was observed between Holstein and Nelore breeds (P<0.001), while the values of the Angus breed were similar to the other two breeds. The difference in motility of fresh and post-thawing semen showed that Nelore and Angus bulls showed greater variation in values between the analyzes (26.0±8.9% and 25.3±8.4%, respectively) showing a significant difference (P<0.001) in relation to Holstein bulls (20.6±9.3%) that obtained the smallest difference. The analysis of fresh and post-thawing semen did not show any significant difference (P=0.13) between breeds. In conclusion, the semen cryopreservation process causes a decrease in the physical parameters of the semen and these quality losses suffer interference according to the breeds.

This study aimed to find out the quality of fresh semen from Sumba Ongole (SO) bulls which was diluted with Bestvile Thawing Solution (BTS) diluent modified with soybean milk (SKD). The study used a completely randomized design consisting of four treatments and 5 replications, P0 (100 % BTS), P1 (95 % BTS + 5 % SM), P2 (90 % BTS + 10 % SM), and P3 (85 % BTS + 15 % SM). The parameters observed were the colour, smell, consistency, pH, volume, motility, viability, and abnormalities of spermatozoa. The results showed that the characteristics were in the normal, creamy white semen colour, semen volume 3,6 ml, medium consistency, pH 6,3, typical SO bulls smell, sperm motility 85 %, mass activity +++, sperm concentration 1276 million/ml, live sperm 90,05 % and sperm abnormalities 6,15 % and quality there was no significant difference between the four treatments. The spermatozoa motility and viability in each treatment was able to which achieved on day 1. The characteristics of the spermatozoa of SO bulls were in the normal category. Meanwhile, BTS diluent with the addition of SKD was unable to maintain sperm quality.

The objective of this study was to investigate the effect of using different extenders <italic>viz.</italic> Androhep, Safecell Plus and SUS during cryopreservation on quality of frozen-thawed boar semen. Semen volume, sperm motility, sperm concentration, percentage of morphologically abnormal spermatozoa, total number of spermatozoa per ejaculate and activity of the enzyme aspartate aminotransferase (AST) were assessed in fresh semen collected from 39 fertile AI boars. Semen from each boar was divided into three portions and diluted 1:1.5 in extender Androhep, Safecell Plus and SUS and keep at 17°C for 15-h holding time before cryopreservation. Then sperm was cryopreserved. Straws were thawed in a water bath at 38°C for 40s and post-thaw sperm motility with AST activity was assessed. Significant difference in post-thaw sperm motility was found between extender Androhep and Safecell Plus (P<0.05). AST activity did not differ significantly between tested extenders (P>0.05). In conclusion, the results of the study indicate that using Safecell Plus extender during holding period before cryopreservation significantly affected post-thaw sperm motility.

AbstractThis study aimed to define potential markers that could determine the suitability of ejaculate for cryopreservation. Fresh semen from eleven boars (4–7 ejaculates/boar), regardless of their sperm motility, was subjected to a cryopreservation procedure. The sperm quality before and after freezing was assessed based on the sperm membrane permeability and acrosome integrity. The results showed that it was possible to effectively cryopreserve ejaculates below the accepted standards of 70–80% of fresh motile sperm and still obtain a high cryosurvival rate. Moreover, a significant correlation was found between the percentage of viable sperm with apoptotic-like changes, viable sperm with reacted acrosomes, and the cryosurvival rate. The proposed markers for assessing the quality of fresh semen could be used to predict the success of cryopreservation procedures.

Considering the major role of human resource in achieving the company’s goal the company should provide an operational skill or knowledge directly from manager to employee’s levels. The company should give the employees a better understanding that the training is provided not only to execute the pre-determined program smoothly, but also to increase the quality of their achievement in their own field that in turn will influence their income, performance and future career planning. Comparing the statistical scores gained by the employee before and after the training proves the significant difference. After the training the score was higher (3.87%) than before the training (3.08%), which means that the training gave good impact. Other statistical computation using t – paired samples with α = 0.005 and degree of freedom (df = 29) also proved the difference where t counted (-38.445) is bigger than t table (-1.669). The training was basically aiming at formulating the expected ability from the employees to change their behavior. Such a change was formulated in terms of behavioral change and a short-term educational program by using systemic and organized procedures, so that operative employees learn technical and skill knowledge for specific purpose.

Background: Cervical herniated nucleus pulposus compresses the nerve roots with clinical pain manifestation and causes sleeping disturbances. The aim of this study was to determine the difference in the quality of sleep before and after surgery.Method: This is a retrospective cohort study. Patients were asked to complete the Pittsburgh Sleep Quality Index (PSQI) and all data was analyzed with the McNemar Test.Result: The outcome revealed that in 98 patients, 72 (73.4%) patients had increased their quality of sleep after surgery.Conclusion: There was a significant difference in the quality of sleep pre- and post-surgery.

Objectives: This study is designed to statistically analyze data of outpatients visiting for recent 3 years. The purpose is to identify tendencies of patients who visit the hospitals before and after COVID-19. Methods: This study retrospectively analyzed the medical records of 452,487 patients who visited to the Daejeon·Cheonan·Cheongju Korean Medicine Hospital of Daejeon University from January 1, 2018 to August 31, 2020. The data is classified according to year, month, gender, age, and visit type. The statistical analysis was performed using IBM SPSS 25.0. Results: The total number of patients decreased in 2020 compared to before 2020, and the number of patients in each hospital also decreased from the previous year. According to the year of each hospital, the difference by year was not statistically significant at Daejeon Hospital, but Cheonan and Cheongju Hospital showed statistical difference. The change in monthly according to the year by hospital has continued to decrease from January to March in 2020, unlike the previous year, which recovered from a decline in March. In the analysis of patients by age, there was no statistically significant difference in the number of patients in the above 60s according to the year, while those under 60s have a tendency to significantly decrease.. Conclusion: We expect that the results of this study will be used as reference materials in analyzing effects of COVID-19 at health care utilization.

The goal of psycho acoustic or subjective voice analysis, in a phoniater's everyday work, is to describe a subjective experience based on the physical parameters created in the process of phonation. The work was a clinical prospective study and the sample consisted of 80 people of both sexes, 40 people with benign and pseudo tumors of vocal cords and 40 people with malign tumors of vocal cords. All the patients were otorinolaringologically and phoniatrically examined. The subjective acoustic analysis was done with the patients pronouncing numbers from 1 to 10 in the comfortable zone. Afterwards, the quality of the voices was estimated in RBH scale. The subjective acoustic analysis found roughness in the voices of 87,50% patients in the first group and the most frequent value was Mod=3 ( intense roughness), 62,50% patients. Hoarseness was present in 90,00 % cases , with largest value Mod=2 (moderate hoarseness), 55,00% patients. In the second group, roughness existed in the voices of 70,00% patients, most often intense one (Mod=3), 30,00% patients. Hoarseness existed in 95,00% cases, 45,00% with moderate (Mod=2) and 35,00% with intense one. T test showed that there is a statistically significant difference between the strength of the roughness determined by the subjective acoustic analysis in the first and the second group, with p<0,01. The difference between the strength of the hoarseness in the first and the second group is also statistically significant, with p<0,01. All the growths on vocal cords irrespective of their nature change the characteristics of the voice, most of all its clearness. In cases of vocal cords tumors, by the subjective acoustic analysis, i.e. the perception of the psycho physiological characteristics of voice, a human ear can register pathological phenomena of the voice but cannot determine the character of the growth on the vocal cords.

<p>The objective of the study was to know the difference of X:Y spermatozoa ratio and sperm quality of Kacang and Ettawa-crossed breed goats. It was conducted at Laboratory of Animal Physiology and Reproduction, Faculty of Animal Science Universitas Gadjah Mada for three months, starting December 2009 to February 2010. Materials of the study were sperm collected from six two-year Ettawa-crossed breed goats weighing 31.5±2.2 kg and from six two-year Kacang goats weighing 18.0±2.1 kg. The sperm was collected using artificial vagina with frequency of twice a week for each goat and it was repeated eight times. The quality and quantity of the sperm was immediately evaluated by its volume, concentration, motility, viability and abnormality. To determine the ratio of X:Y spermatozoa, smear preparation was made and spermatozoa head was then measured using Scion Image software. The results of the measurement were used for identification of X or Y spermatozoa. Spermatozoa having bigger head than the average were identified as X and those having smaller head as Y. The result showed that the ratio of X:Y spermatozoa of Ettawa-crossed breed goats (50.6±1.8:49.4±1.8 %) was not significantly difference with that of Kacang goats<br />(49.7±1,7:50.3±1.7 %). Sperm volume of Ettawa-crossed breed goats (1.00±0.2 ml) was higher than that of Kacang goats (0.62±0.2 ml). Sperm concentration of Ettawa-crossed breed goats (2,865±431 million/ml) was not significantly difference compared to that of Kacang goats (2,840±383 million/ml). Spermatozoa motility of Ettawa-crossed breed goats (66.7±9.8 %) was higher (P&lt;0.05) than that of Kacang goats (50.0±7.1 %). Spermatozoa viability of Ettawa-crossed breed goats (80.0±7.1 %) was higher (P&lt;0.05) than that of<br />Kacang goats (67.5±9.4 %). Spermatozoa abnormality of Ettawa-crossed breed goats (8.2±3.3 %) was not significantly difference compared to that of Kacang goats (8.6±2.4 %).
