Abstract A study was conducted to determine if aflatoxin and fumonisin are concentrated in the poor-quality grade components of shelled corn. Four 1.0 kg test samples were each taken from 23 lots of shelled corn marketed in North Carolina. Inspectors from the Federal Grain Inspection Service divided each test sample into 3 grade components: (1) damaged kernels (DM), (2) broken corn and foreign material (BCFM), and )3) whole kernels (WH). The aflatoxin and fumonisin concentration was measured in each component and a mass balance equation was used to calculate the total concentration of each mycotoxin in each test sample. Averaged across all test samples, the aflatoxin concentrations in the DM, BCFM, and WH components were 1300.3, 455.2, and 37.3 ppb, respectively. Averaged across all test samples, the fumonisin concentrations in the DM, BCFM, and WH components were 148.3, 51.3, and 1.8 ppm, respectively. The DM and BCFM components combined accounted for only 5.0% of the test sample mass, but accounted for 59.8 and 77.5% of the total aflatoxin and fumonisin mass in the test sample, respectively. Both aflatoxin mass (ng) and aflatoxin concentration (ng/g) in the combined DM and BCFM components had high correlations with aflatoxin concentration in the lot. The highest correlation occurred when aflatoxin mass (ng) in the combined DM and BCFM components was related to aflatoxin concentration in the lot (0.964). Similar results were obtained for fumonisin. This study indicated that measuring either aflatoxin or fumonisin in the combined DM and BCFM grade components could be used as a screening method to predict either aflatoxin or fumonisin in a bulk lot of shelled corn.

Abstract A direct competitive enzyme-linked immunosorbent assay screening method for af latoxins at 20 ng/g in corn was studied by 15 collaborating laboratories. Test samples of corn were extracted by blending with methanol-water (8 + 2). The extracts were filtered and the filtrates were diluted with buffer to a final methanol concentration of <30%. Each diluted filtrate was applied to a test device containing a filter with immobilized polyclonal antibodies specific to aflatoxins B1, B2, and G1. Aflatoxin Bi-peroxidase conjugate was added, the test device was washed with water, and a mixture of hydrogen peroxide and tetramethylbenzidine was added. A test sample was judged to contain ≥20 ng af latoxins/g when, after exactly 1 min, no color was observed on the filter; if a blue or gray color developed, the test sample was judged to contain <20 ng aflatoxins/g. All laboratories correctly identified naturally contaminated corn test samples. Only one false positive was found for controls containing no aflatoxins. The correct responses for positive test samples spiked at levels of 10,20, and 30 ng aflatoxins/g (the ratio of B1:B2:G1 was 15:1:3) were 67,97, and 100%, respectively. This method was adopted first action by AOAC INTERNATIONAL as a change in method for 990.34 for screening for aflatoxins B1, B2, and G1 in corn at total aflatoxin concentrations of ≥20 ng/g.

Abstract A screening method for corn and corn products, based on a minicolumn, was modified slightly to assay 60 lots of corn at one elevator to determine whether they could be sold as animal feed. To be salable, the lots had to contain less than 20 ppb total aflatoxin. Aflatoxin levels in the lots were later determined by the official AOAC method for corn to check effectiveness of the screening. No lot had been designated for sale that contained 20 or more ppb total aflatoxin.

Abstract The minicolumn screening method for aflatoxins was collaboratively tested on naturally contaminated almonds. The nuts were extracted, and the extract was cleaned up and applied to a Velasco-type minicolumn. This permits the detection of total anatoxins (B1, B2, G1, G2) as a fluorescent band on the Florisil layer of the column. The results of 20 collaborators are presented. Samples containing 0, 2, 5, 10, and 25 ng aflatoxin/g were analyzed. Ninety-six per cent of the samples containing 5—25 ng total aflatoxins/g and 83% of the negative samples were correctly identified. The method has been adopted as official first action for detection of total aflatoxin levels of ≥5 ng/g.

There is a great need for the development of a rapid and reliable bioassay to evaluate the pulmonary toxicity of inhaled particles. A number of methods have been proposed, including lung clearance studies, bronchoalveolar lavage analysis, and in vitro cytotoxicity tests. These methods are often limited in scope inasmuch as they measure only one dimension of the pulmonary response to inhaled, instilled or incubated dusts. Accordingly, a comprehensive approach to lung toxicity studies has been developed.To validate the method, rats were exposed for 6 hours or 3 days to various concentrations of either aerosolized alpha quartz silica (Si) or carbonyl iron (CI) particles. Cells and fluids from groups of sham and dust-exposed animals were recovered by bronchoalveolar lavage (BAL). Alkaline phosphatase, LDH and protein values were measured in BAL fluids at several time points postexposure. Cells were counted and evaluated for viability, as well as differential and cytochemical analysis. In addition, pulmonary macrophages (PM) were cultured and studied for morphology, chemotaxis, and phagocytosis by scanning electron microscopy.

SummaryPatients undergoing orthopedic surgery are at high risk of developing deep vein thrombosis. One hundred and thirty-eight consecutive patients undergoing total hip replacement or hip fracture surgery were included in this study. They were surveilled with colour Doppler ultrasound (CDU) and bilateral ascending contrast phlebography. The prevalence of proximal and distal DVT in this study was 5.8% and 20.3% respectively.CDU has a satisfactory sensitivity in patients with symptomatic deep vein thrombosis, especially in the proximal region. These results could not be confirmed in the present study of asymptomatic patients. The sensitivity was 62.5% (95% confidence interval: C.I. 24-91%) and the specificity 99.6% (C.I. 98-100%) for proximal DVT; 53.6% (C.I. 34-73%) and 98% (C.I. 96-99%) respectively for distal thrombi. The overall sensitivity was 58.1% (C.I. 39-75%) and the specificity 98% (C.I. 96-99%). The positive predictive value was 83.3% (C.I. 36-99%) and 75% (C.I. 51-91%) for proximal and distal DVT respectively. The negative predictive value was 98.9% (C.I. 98-100%) and 94.9% (C.I. 92-98%) for proximal and distal DVT respectively. The results of this study showed that even with a highly specialised and experienced investigator the sensitivity of CDU was too low to make it suitable for screening purposes in a high risk surgical population.

SummaryLiposomes of pure phospholipids were used in a modified APTT test system and the role of phosphatidyl serine (PS) in determining the sensitivity of the test system to the presence of lupus anticoagulants was assessed. Six consecutive patients with lupus anticoagulants and seven haemophiliacs with anticoagulants directed at specific coagulation factors, were studied. Increasing the concentration of phospholipid in the test system markedly reduced the sensitivity to lupus anticoagulants but had marginal effect on the specific factor inhibitors. The same effect was achieved when the content of PS alone was increased in a vehicle liposome of constant composition.The results suggest that the lupus anticoagulants can best be detected by a screening method using an APTT test with a reagent of low PS content. The use of a reagent rich in PS will largely abolish the lupus anticoagulant’s effect on the APTT. An approach using the two different types of reagent may facilitate differentiation of lupus inhibitors from other types of anticoagulant.
