Soil microorganisms play crucial roles between plants and soil following afforestation. However, the relationship between the microbial community and carbon:nitrogen:phosphorus (C:N:P) stoichiometry in the plant–soil–microbe continuum remains unclear. In this study, we investigated this relationship by collecting plant and soil samples from Caragana korshinskii Kom. plantations with different years of afforestation (17-, 32-, and 42-year-old plantations), and from farmland. Illumina sequencing of the 16S rRNA and internal transcribed spacer (ITS) ribosomal RNA was used to examine the soil microbial community and the C, N, and P concentrations in plants, soil, and microbial biomass. Other soil characteristics were also measured. The results showed that the C and N concentrations in plants (leaves, herbs, and litter), soil, and microbial biomass increased as the vegetation restoration stage increased, but the P concentration in leaves and herbs slightly decreased. The C:P and N:P ratios in the plant–soil–microbe continuum substantially increased over time, particularly that of the microbial biomass. These results suggest that the unbalanced increase of C, N, and P following vegetation restoration may result in a P limitation in plant–soil systems. Moreover, bacterial and fungal alpha diversity significantly increased following afforestation. Afforestation had a greater impact on bacterial diversity (both alpha and beta diversity) than did fungal diversity. Among the dominant bacterial taxa, Proteobacteria increased significantly with afforestation time, whereas Actinobacteria decreased and Acidobacteria peaked in 32-year-old C. korshinskii plantations. However, there were no significant changes in the dominant fungal taxa. Collectively, we found that microbial diversity and dominant phyla were closely associated with the C:P and N:P ratios in the plant–soil–microbe continuum, particularly the N:P ratio. These results suggest that microbial diversity and composition may be limited by the imbalances of C, N, and especially P in afforested ecosystems, which provides evidence of linkages between microbial diversity and plant–soil systems in afforested ecosystems and could help in improving the predictions of sustainably restoring C. korshinskii plantations.

AbstractMicrobial respiration is critical for soil carbon balance and ecosystem functioning. Previous studies suggest that plant diversity influences soil microbial communities and their respiration. Yet, the linkages between tree diversity, microbial biomass, microbial diversity, and microbial functioning have rarely been explored. In this study, we measured two microbial functions (microbial physiological potential, and microbial respiration), together with microbial biomass, microbial taxonomic and functional profiles, and soil chemical properties in a tree diversity experiment in South China, to disentangle how tree diversity affects microbial respiration through the modifications of the microbial community. Our analyses show a significant positive effect of tree diversity on microbial biomass (+25% from monocultures to 24-species plots), bacterial diversity (+12%), and physiological potential (+12%). In addition, microbial biomass and physiological potential, but not microbial diversity, were identified as the key drivers of microbial respiration. Although soil chemical properties strongly modulated soil microbial community, tree diversity increased soil microbial respiration by increasing microbial biomass rather than changing microbial taxonomic or functional diversity. Overall, our findings suggest a prevalence of microbial biomass over diversity in controlling soil carbon dynamics.

There is little information on the effects of S management strategies on soil microorganisms under zero tillage systems o n the North American Prairies. Experiments were conducted to examine the effects of tillage and source and placement of S on soil microbial biomass (substrate induced respiration) and functional diversity (substrate utilization patterns) in a canola-wheat rotation under conventional and zero tillage systems at three sites in Gray Luvisolic and Black Chernozemic soils. Conventional tillage significantly reduced microbial biomass and diversity on an acidic and C-poor Luvisolic soil, but it had mostly no significant effects on the near-neutral, C-rich Luvisolic and Chernozemic soils, which underlines the importance of soil C in maintaining a healthy soil. Sulphur had no significant effects on soil microbial biomass, and its effects on microbial diversity were more frequent on the near-neutral Luvisol, which was more S-deficient, than on the acidic Luvisol or the Chernozem. Significant S effects on microbial diversity were observed both in the bulk soil (negative effects, compared with the control) and rhizosphere (positive effects) of the acidic Luvisol, but all significant effects (positive) were observed in root rhizospheres in the other soils. Sulphur by tillage interactions on acidic Luvisolic soil indicated that the negative effects of S in bulk soil occurred mostly under zero tillage, presumably because the fertilizer is concentrated in a smaller volume of soil than under conventional tillage. Sulphate S effects, either negative or positive, on microbial diversity were usually greater than elemental S effects. Therefore, S application can have direct, deleterious effects on soil microorganisms or indirect, beneficial effects through crop growth, the latter presumably due to increased root exudation in the rhizosphere of healthy crops. Abstract. Agriculture-driven declines in plant biodiversity reduce soil microbial biomass, alter microbial functions, and threaten the provisioning of soil ecosystem services. We examined whether increasing temporal plant biodiversity (by rotating crops) can partially reverse these trends and enhance microbial biomass and function. We quantified seasonal patterns in soil microbial biomass, respiration rates, extracellular enzyme activity, and catabolic potential three times over one growing season in a 12-year crop rotation study at the W.K. Kellogg Biological Station LTER. Rotation treatments varied from one to five crops in a three-year rotation cycle, but all soils were sampled under corn to isolate historical rotation effects from current crop effects. Inorganic N, the stoichiometry of microbial biomass and dissolved organic C and N varied seasonally, likely reflecting fluctuations in soil resources during the growing season. Soils from biodiverse cropping systems increased microbial biomass C by 28–112 % and N by 18–58 % compared to monoculture corn. Rotations increased potential C mineralization by as much as 64 %, and potential N mineralization by 62 %, and both were related to substantially higher hydrolase and lower oxidase enzyme activities. The catabolic potential of the microbial community, assessed with community-level physiological profiling, showed that microbial communities in monoculture corn preferentially used simple substrates like carboxylic acids, relative to more diverse cropping systems. By isolating plant biodiversity from differences in fertilization and tillage, our study illustrates that crop biodiversity has overarching effects on soil microbial biomass and function that last throughout the growing season. In simplified agricultural systems, relatively small increases in plant biodiversity have a large impact on microbial community size and function.

During a 3-year study, soil microbial biomass C and N, length of the fungal hyphae, soil respiration, and the percent mass loss of needle litter were recorded in coniferous forest soil humus layers following a prescribed burning (PB) treatment or a forest fire simulation (FF) treatment (five plots per treatment). Unburned humus from adjacent plots served as controls (PC and FC, respectively). Prescribed burning was more intensive than the forest fire, and this was reflected in all the measurements taken. The amounts of microbial biomass C and N, length of fungal hyphae, and soil respiration in the PB area did not recover to their controls levels, whereas unchanged microbial biomass N and recovery of the length of the fungal hyphae to control levels were observed in the FF area. The mean microbial C/N ratio was approximately 7 in all the areas, which reflected the C/N ratio of the soil microbial community. Deviation from this mean value, as observed during the first three samplings from the PB area (3, 18, and 35 days after fire treatment), suggested a change in the composition of the microbial community. Of the two treated areas, the decrease in soil respiration (laboratory measurements) was much more pronounced in the PB area. However, when the humus samples from both areas were adjusted to 60% water holding capacity, no differences in respiration capacity were observed. The drier humus, due to higher soil temperatures, of the PB area is a likely explanation for the low soil respiration. Lower soil respiration was not reflected in lower litter decomposition rates of the PB area, since there was a significantly higher needle litter mass loss during the first year in the PB area followed by a decline to the control level during the second year. Consistently higher mass losses were recorded in the FC area than in the FF area.

ABSTRACTThe Green Revolution of agriculture was in part driven by application of synthetic mineral fertilizers, largely supplanting organic manure as a source of the major nutrients nitrogen, phosphorous and potassium (NPK). Though enhancing crop production and global food security, fertilizers have contributed to soil acidification, eutrophication of water bodies, and greenhouse gas emissions. Organic agriculture, employing manures or composts, has been proposed as a way of mitigating these undesirable effects. Of particular interest is the effect of fertilizer regime on soil microbes, which are key to nutrient cycling, plant health and soil structure. Meta-analyses of experimental studies indicate that mineral fertilizer increases soil microbial biomass over unfertilized controls, and that organic fertilizers increase microbial biomass and activity over mineral fertilizers. However, the effect of fertilizers on soil microbial diversity remains poorly understood. Since biological diversity is an important determinant of ecosystem function and a fundamental metric in community ecology, the effects of fertilizer regimes on soil microbial diversity are of theoretical and applied interest. Here, we conduct a meta-analysis of 31 studies reporting microbial diversity metrics in mineral fertilized (NPK), organically fertilized (ORG) and unfertilized control (CON) soils. Of these studies, 26 reported taxonomic diversity derived from sequencing, gradient gel electrophoresis, RFLP, or dilution plate assay. Functional diversity, derived from Biolog Ecoplate™ measures of carbon substrate metabolism, was reported in 8 studies, with 3 studies reporting both diversity metrics. We found that functional diversity was on average 2.6 % greater in NPK compared with CON, 6.8 % greater in ORG vs CON and 3.6 % greater in ORG vs NPK. Prokaryote taxonomic diversity was not significantly different between NPK and CON, 4.2 % greater in ORG vs CON and 4.6 % greater in ORG vs. NPK. Fungal taxonomic diversity was not significantly different between NPK or ORG vs CON, but 5.4 % lower between ORG and NPK. There was very high residual heterogeneity in all meta-analyses of soil diversity, suggesting that a large amount of further research with detailed analysis of soil properties is required to fully understand the influence of fertilizer regimes on microbial diversity and ecosystem function.

<p>Soils are made up of organic and an organic material. The organic soil component contains all the living creatures in the soil and the dead ones in various stages of decomposition.  Biological activity in soil helps to recycle nutrients, decompose organic matter making nutrient available for plant uptake, stabilize humus, and form soil particles.<br />The extent of the diversity of microbial in soil is seen to be critical to the maintenance of soil health and quality, as a wide range of microbial is involved in important soil functions.  That ecologically managed soils have a greater quantity and diversity of soil microbial. The two main drivers of soil microbial community structure, i.e., plant type and soil type, are thought to exert their function in a complex manner. The fact that in some situations the soil and in others the plant type is the key factor determining soil microbial diversity is related to their complexity of the microbial interactions in soil, including interactions between microbial and soil and microbial and plants. <br />The basic premise of organic soil stewardship is that all plant nutrients are present in the soil by maintaining a biologically active soil environment. The diversity of microbial communities has on ecological function and resilience to disturbances in soil ecosystems. Relationships are often observed between the extent of microbial diversity in soil, soil and plant quality and ecosystem sustainability. Agricultural management can be directed toward maximizing the quality of the soil microbial community in terms of disease suppression, if it is possible to shift soil microbial communities.</p><p>Keywords: structure, microbial, implication, sustainable agriculture<br /><br /></p>
