Tilletia controversa J. G. Kühn is a causal organism of dwarf bunt in wheat. Understanding the interaction of wheat and T. controversa is of practical and scientific importance for disease control. In this study, the relative expression of TaLHY and TaPR-4 and TaPR-5 genes was higher in a resistant (Yinong 18) and moderately resistant (Pin 9928) cultivars rather than susceptible (Dongxuan 3) cultivar at 72 h post inoculation (hpi) with T. controversa. Similarly, the expression of defensin, TaPR-2 and TaPR-10 genes was observed higher in resistant and moderately resistant cultivars after exogenous application of phytohormones, including methyl jasmonate, salicylic acid, and abscisic acid. Laser confocal microscopy was used to track the fungal hyphae in the roots, leaves, and tapetum cells, which of susceptible cultivar were infected harshly by T. controversa than moderately resistant and resistant cultivars. There were no fungal hyphae in tapetum cells in susceptible cultivar after methyl jasmonate, salicylic acid and abscisic acid treatments. Moreover, after T. controversa infection, the pollen germination was of 80.06, 58.73, and 0.67% in resistant, moderately resistant and susceptible cultivars, respectively. The above results suggested that the use using of resistant cultivar is a good option against the dwarf bunt disease.

Phytophthora nicotianae is a devastating root and stem pathogen of tobacco (Nicotiana tabacum) in South Africa. Growers strive to control the resulting disease, known as black shank, with metalaxyl treatments and resistant cultivars. The aim of this study was to consider whether development of metalaxyl resistance in P. nicotianae has contributed to poor disease control and if recently developed cultivars with high levels of resistance require metalaxyl for effective control. One hundred and thirty-two isolates of P. nicotianae were screened for sensitivity to metalaxyl. P. nicotianae isolates from most tobacco farms were metalaxyl sensitive. Growth of most isolates was inhibited completely at 1.0 μg a.i./ml. However, isolates from the MKTV tobacco producing area showed EC50 values ranging from 1.02 μg a.i./ml to 3.57 μg a.i./ml. Twenty-one tobacco cultivars were planted and treated with and without metalaxyl in two different growing seasons to evaluate their resistance to P. nicotianae and the value of using metalaxyl. Hicks was the most susceptible cultivar. Vuma/3/46, LK30/40/60-1, and LK33/60 exhibited the greatest resistance to P. nicotianae. Use of metalaxyl in combination with moderately resistant cultivars such as NC60 × TL33 and LK10/80/60 effectively reduced black shank in the field. Resistant cultivars were healthy and no significant difference between metalaxyl treated and untreated plants was observed.

Sorghum genes responsive to purple witchweed parasitism were isolated, and their expression was analyzed. Using the suppression-subtractive hybridization strategy, 30 genes that were up-regulated in response to purple witchweed parasitism were isolated from the roots of a susceptible sorghum cultivar ‘Abu 70’. The changes in the expression of each gene were investigated in the roots and leaves of the sorghum cultivars ‘Wad Ahmed’ and ‘Tabat’ were parasitized by purple witchweed. Tabat is more susceptible and Wad Ahmed is less susceptible to purple witchweed than Abu 70. Further, the changes in the gene expression and host susceptibility to purple witchweed were studied in the roots of the three sorghum cultivars after treatment with salicylic acid (SA) or methyl jasmonate (MeJA). Purple witchweed parasitism induced jasmonic acid (JA)-responsive genes and suppressed SA-responsive genes in the roots of the cultivars Abu 70 and Tabat. In contrast, purple witchweed parasitism in the less-susceptible cultivar Wad Ahmed induced SA-responsive genes and induced JA-responsive genes to a small extent. SA, but not MeJA, decreased the susceptibility of all the sorghum cultivars to purple witchweed. Systemic expression of a few genes was observed in the leaves of the purple witchweed–parasitized sorghum cultivars.

The short shelf-life and loss of bioactive compounds of strawberry fruit are the most important problems during strawberry refrigerated storage. This study was carried out to evaluate the effect of the pre-harvest foliar application of salicylic acid (SA) (2 and 4 mM), abscisic acid (ABA) (0.25 and 0.50 mM), and methyl jasmonate (MeJA) (0.25 and 0.50 mM) three times, 10 d apart, at fruit development and ripening stages on storage ability and bioactive compounds of strawberry fruit (cv. Festival) stored at 4 °C for 12 d. Our results showed that fruit obtained from both concentrations of ABA and 0.25 mM MeJA was firmer and had higher total soluble solids (TSS) than fruit from non-treated plants. However, all previous applications had no significant effect on weight loss, pH, or color. Applications of 4 mM SA and 0.25 mM MeJA conserved fruit from ascorbic acid (AsA) loss compared to control at the end of the storage period. In addition, all pre-harvest applications remained higher in total phenolic compounds (TPC) and anthocyanin contents compared to controls at the last storage period. Hence, the pre-harvest application of SA, ABA, and MeJA could be used to conserve TPC and anthocyanin as well as the quality of strawberry fruits during refrigerated storage.
