Multi-factorial mitochondrial damage exhibits a “vicious circle” that leads to a progression of mitochondrial dysfunction and multi-organ adverse effects. Mitochondrial impairments (mitochondriopathies) are associated with severe pathologies including but not restricted to cancers, cardiovascular diseases, and neurodegeneration. However, the type and level of cascading pathologies are highly individual. Consequently, patient stratification, risk assessment, and mitigating measures are instrumental for cost-effective individualized protection. Therefore, the paradigm shift from reactive to predictive, preventive, and personalized medicine (3PM) is unavoidable in advanced healthcare. Flavonoids demonstrate evident antioxidant and scavenging activity are of great therapeutic utility against mitochondrial damage and cascading pathologies. In the context of 3PM, this review focuses on preclinical and clinical research data evaluating the efficacy of flavonoids as a potent protector against mitochondriopathies and associated pathologies.

One in every four deaths is due to cancer in Europe. In view of its increasing incidence, cancer became the leading cause of death and disease burden in Denmark, France, the Netherlands, and the UK. Without essential improvements in cancer prevention, an additional 775,000 cases of annual incidence have been prognosed until 2040. Between 1995 and 2018, the direct costs of cancer doubled from EUR 52 billion to EUR 103 billion in Europe, and per capita health spending on cancer increased by 86% from EUR 105 to EUR 195 in general, whereby Austria, Germany, Switzerland, Benelux, and France spend the most on cancer care compared to other European countries. In view of the consequent severe socio-economic burden on society, the paradigm change from a reactive to a predictive, preventive, and personalized medical approach in the overall cancer management is essential. Concepts of predictive, preventive, and personalized medicine (3PM) demonstrate a great potential to revise the above presented trends and to implement cost-effective healthcare that benefits the patient and society as a whole. At any stage, application of early and predictive diagnostics, targeted prevention, and personalization of medical services are basic pillars making 3PM particularly attractive for the patients as well as ethical and cost-effective healthcare. Optimal 3PM approach requires novel instruments such as well-designed liquid biopsy application. This review article highlights current achievements and details liquid biopsy approaches specifically in cancer management. 3PM-relevant expert recommendations are provided.

The number of data sources available for academic research on retirement economics and policy has increased rapidly in the past two decades. Data quality and comparability across studies have also improved considerably, with survey questionnaires progressively converging towards common ways of eliciting the same measurable concepts. Probability-based Internet panels have become a more accepted and recognized tool to obtain research data, allowing for fast, flexible, and cost-effective data collection compared to more traditional modes such as in-person and phone interviews. In an era of big data, academic research has also increasingly been able to access administrative records (e.g., Kostøl and Mogstad, 2014; Cesarini et al., 2016), private-sector financial records (e.g., Gelman et al., 2014), and administrative data married with surveys (Ameriks et al., 2020), to answer questions that could not be successfully tackled otherwise.

Universal area-under-the-curve (AUC) guided vancomycin therapeutic drug monitoring (TDM) is resource-intensive, cost-prohibitive, and presents a paradigm shift that leaves institutions with the quandary of defining the preferred and most practical method for TDM. We report a step-by-step quality improvement process using 4 plan-do-study-act (PDSA) cycles to provide a framework for development of a hybrid model of trough and AUC-based vancomycin monitoring. We found trough-based monitoring a pragmatic strategy as a first-tier approach when anticipated use is short-term. AUC-guided monitoring was most impactful and cost-effective when reserved for patients with high-risk for nephrotoxicity. We encourage others to consider quality improvement tools to locally adopt AUC-based monitoring.

Due to the increasing popularity of new research in medicine thisstudy was conducted to determine recent research trends of predictive, preventive and personalized medicine (PPM). We identified the terms relevant to PPM using own search engine based on neural network processing in PubMed database. We extracted initially about 15000 articles. Then we carried out the statistical analysis for identifying research trends. The article presents the results of solving the problem of evaluating research topics at the level of thematic clusters in a separate subject area. An approach based on the analysis of article titles has been implemented. Identification of terms, connections between them and thematic clustering were carried out using the free software VOSViewer, which allows to extract terms in the form of noun phrases, as well as to cluster them.

AbstractLow-coverage whole genome sequencing followed by imputation has been proposed as a cost-effective genotyping approach for disease and population genetics studies. However, its competitiveness against SNP arrays is undermined as current imputation methods are computationally expensive and unable to leverage large reference panels.Here, we describe a method, GLIMPSE, for phasing and imputation of low-coverage sequencing datasets from modern reference panels. We demonstrate its remarkable performance across different coverages and human populations. It achieves imputation of a full genome for less than $1, outperforming existing methods by orders of magnitude, with an increased accuracy of more than 20% at rare variants. We also show that 1x coverage enables effective association studies and is better suited than dense SNP arrays to access the impact of rare variations. Overall, this study demonstrates the promising potential of low-coverage imputation and suggests a paradigm shift in the design of future genomic studies.

Abstract Objectives Few studies reported the periodontal disease-related metabolic profile of end-stage renal disease (ESRD) patients. The present study aimed to compare the inflammatory and metabolic differences between patients with ESRD and healthy controls, and to identify potential useful biomarkers for predictive, preventive, and personalized medicine (PPPM) in GCP and serum of ESRD patients.Methods Patients with ESRD (ESRD group; n = 52) and healthy controls (HC group; n = 44) were recruited. Clinical periodontal parameters were recorded. The differential metabolites in the GCF and serum were identified by liquid chromatography/mass spectrometry. Inflammatory markers including Interleukin-1β (IL-1β), Interleukin-6 (IL-6), Interleukin-8 (IL-8) and C-reactive protein (CRP) were also assessed. Results In ESRD group, IL-8 and CRP were significantly higher in GCF, whereas IL-6 and CRP were significantly higher in serum, compared with HC group (all P < 0.05). In the case of GCF, taurine levels were positively correlated with IL-8 levels in both groups (all P < 0.05). In the case of serum, L-phenylalanine and p-hydroxyphenylacetic acid levels were positively correlated with CRP levels in both groups (all P < 0.05). Significant positive correlation was observed between pseudouridine and IL-6 levels only in ESRD group. Conclusions IL-8 and CRP were potential inflammatory makers. Metabolites of taurine in GCF as well as L-phenylalanine and p-hydroxyphenylacetic acid in serum were possible biomarkers that correlated with inflammatory cytokine. All these biomarkers may consider as a potential strategy for the prediction, diagnosis, prognosis, and management of personalized periodontal therapy in the population with ESRD.
