Background: Tear film fluid serves as a dynamic barrier that both lubricates the eye and protects against allergens and infectious agents. However, a detailed analysis of a bacteria-induced immune response on the tear film lipidome has not been undertaken. Methods: We undertook a high-resolution mass spectrometry lipidomics analysis of endogenous anti-inflammatory and structural tear film lipids in bovine pink eye. Results: Bovine pink eye resulted in dramatic elevations in tear fluid levels of the anti-inflammatory lipids resolvin E2, cyclic phosphatidic acid 16:0, and cyclic phosphatidic acid 18:0. In addition, there were elevated levels of the structural lipids (O-acyl)-ω-hydroxy-fatty acids, cholesterol sulfate, ethanolamine plasmalogens, and sphingomyelins. Lipid peroxidation also was augmented in pink eye as evidenced by the hydroperoxy derivatives of ethanolamine plasmalogens. Conclusions: Ocular infections with Moraxella bovis result in the induction of a number of endogenous anti-inflammatory lipids and augmentation of the levels of structural glycerophospholipids and sphingolipids. Increased levels of hydroperoxy glycerophospholipids also indicate that this bacterial infection results in lipid peroxidation.

Seven new xanthones, diaporthones A−G (1−7), together with 13 known analogues, including five mono- (8−14) and six dimeric xanthones (15−20), were obtained from the ascidian-derived fungus Diaporthe sp. SYSU-MS4722. Their planar structures were established by extensive spectroscopic analyses, including 1D and 2D NMR and high-resolution mass spectrometry (HR-ESIMS). The absolute configurations of 1−7 were clearly identified by X-ray crystallographic analysis and calculation of the ECD Spectra. Compounds 15−20 showed significant anti-inflammatory activity with IC50 values between 6.3 and 8.0 μM. In addition, dimeric xanthones (15−20) showed selective cytotoxicity against T98G cell lines with IC50 values ranging from 19.5 to 78.0 μM.

The aim of our study was to determine the relationship of processes of atherogenesis and indicators of endothelial dysfunction with the processes of activation of lipid peroxidation (LPO) and chronic inflammation in patients with CKD stages II-IV. Material and methods. Levels of proinflammatory and anti-inflammatory cytokines, indicators of lipid peroxidation, the thickness of the intima-media (IMT) of the carotid artery, ankle-brachial index (ABI) were measured in 90 patients with CKD stage II-IV (30 - CKD st. II, 31 patients with CKD st. III and 29 st. IV CKD patients) and 30 healthy subjects (control group). The correlation analysis was performed to identify the possible association between the obtained parameters of atherosclerotic vascular lesions and endothelial dysfunction and indices of lipid peroxidation and pro- and anti-inflammatory cytokines. Results. The average levels of interferon-y, interleukin (IL) -1p and IL-10 in patients with CKD II-IV Art. were significantly increased compared with the corresponding values in the control group. The average levels of malondialde- hyde (MDA) of serum and MDA of erythrocyte in CKD patients was significantly increased compared with the corresponding values in the control group, and the antioxidant system indices - total peroxidase activity of erythrocytes and serum levels of the sulfhydrylgroups was significantly reduced relative to the corresponding values in the group control. 39 (43%) patients with CKD st. II-IV were recorded carotid atherosclerotic changes. The IMT (from 0.91 to 1.29 mm) defined in 22 (56%) patients and atherosclerotic plaques (IMT > 1,3 mm) defined in 17 (44%) patients. Bilateral carotid atherosclerotic lesions was observed in 14 (36%) patients with CKD st. II-IV. Endothelial dysfunction was observed in 71 (79%) patients with CKD st. II-IV. There was a statistically significant association between indicators of chronic inflammation, lipid peroxidation and indicators of atherosclerotic vascular lesions and endothelial dysfunction. Conclusion. Pathological processes ofLPO activation, chronic inflammation are closely linked between themselves and progression of atherosclerotic vascular lesions in CKD.

Two new cembrane-type diterpenoids, lobophytins A (1) and B (3), and four new prostaglandins, (5E)-PGB2 (10), (5E)-13,14-dihydro-PGB2 (11), 13,14-dihydro-PGB2 (12) and 13,14-dihydro-PGB2-Me (13), together with ten known compounds were isolated from the soft coral Lobophytum sarcophytoides. The structures of these new secondary metabolites were identified by high resolution mass spectrometry (HR-ESIMS), nuclear magnetic resonance (NMR) and electron circular dichroism (ECD) analyses, as well as the modified Mosher’s method. Compounds 6, 7, 9, 10, 12, 13, 15 and 16 showed potential anti-inflammatory activity by inhibiting the production of nitric oxide (NO) in RAW264.7 cells that were activated by lipopolysaccharide, with IC50 values ranging from 7.1 to 32.1 μM and were better than the positive control indomethacin, IC50 = 39.8 μM.

A novel series of 1-(4-phenoxyphenyl)-3-[5-(substituted aryl)-1,3,4-oxa- diazol-2-yl]propan-1-one was synthesized by reaction of 3-(4-phenoxybenzoyl)propionic acid with several aryl acid hydrazides in phosphorus oxychloride. The structures of the compounds were supported by IR, 1H- and 13C-NMR, MS data and elemental analysis results. These compounds were tested for their anti-inflammatory, analgesic, ulcerogenic and lipid peroxidation actions. A few compounds were found to have very good anti-inflammatory activity in the carrageenan-induced rat paw edema test, while a fair number of the compounds showed significant analgesic activity in the acetic acid-induced writhing test. These new compounds showed very low ulcerogenic action with reduced malondialdehyde content (MDA), which is one of the by-products of lipid peroxidation.

The cardioprotective effects of resveratrol are well established in animal models of metabolic disease but are yet to be investigated in a combined model of hypertension and diabetes. This study investigated the ability of resveratrol’s antioxidant and anti-inflammatory effects to prevent cardiovascular complications in the spontaneously hypertensive streptozotocin-induced diabetic rat. Diabetes was induced in eight-week-old male spontaneously hypertensive rats via a single intravenous injection of streptozotocin. Following this, resveratrol was administered orally for an eight-week period until the animals were sixteen weeks of age. Upon completion of the treatment regime assessments of oxidative stress, lipid peroxidation, inflammation, and cardiovascular function were made. Resveratrol administration to hypertensive-diabetic animals did not impact upon blood glucose or haemodynamics but significantly reduced oxidative stress, lipid peroxidation, and inflammatory cytokines. Reductions in systemic levels of oxidative stress and inflammation conferred improvements in vascular reactivity and left ventricular pump function and electrophysiology. This study demonstrates that resveratrol administration to hypertensive diabetic animals can elicit cardioprotective properties via antioxidant and anti-inflammatory effects. The observed preservation of cardiovascular function was independent of changes in blood glucose concentration and haemodynamics, suggesting that oxidative stress and inflammation are key components within the pathological cascade associated with hypertension and diabetes.
