The analysis of DNA polymorphism of Nitraria sibirica Pall. was carried out at 13 natural populations of the Republic of Altai and Altai Territory using the ISSR technique. Seven effective ISSR primers have been identified to analyze DNA polymorphism in N. sibirica. 99 DNA fragments were yielded at DNA amplification with these primers, 66 of them were polymorphic. The genetic distance Nei (D) between the studied populations of N. sibirica averaged 0.32, at mean 0.09 - within populations. An identification ISSR marker has been revealed, which can be used to study the genetic variability of the genus Nitraria L. (Nitrariaceae) species.

Genetic Analysis ofInonotus ObliquusStrains by RAPDRAPD profiling of eightInonotus obliquusstrains isolated from sclerotia collected from different areas of China was conducted to determine the genetic variability within this important medicinal fungus and to better define relationships between the genotype and geographical origins of isolation. Twelve 10-mer primers generated a total of 167 stable and reproducible DNA fragments, of which 101 (60.5%) were polymorphic. DNA fingerprints revealed genetic diversity among the strains tested, but there was the little intraspecific difference between the fingerprints of individual strains. A phenogram constructed based on UPGMA analysis of genetic distances calculated from RAPD fragment data identified three distinct groupings: (1) BCX01 and BCX02, (2) JL01, JL02, JL03, JL04 and JL05, (3) HLJ01. Our data confirm that the genetic variability among different strains may be a useful ancillary tool for identifyingl. obliquussclerotia of different geographical origins.

Insufficient genetic variability is one of the major problems of plant breeding programmes, especially in sesame. Gamma radiation has been reported to be very effective in creating genetic variability in plants. Three varieties of Nigerian sesame were assessed for spectrum and frequency of mutation induced by Gamma radiations in M1 and M2 generations. The varieties (NCRIBEN-04E, NCRIBEN-01M and NCRIBEN-03L) were treated with four different doses of gamma rays (250, 350, 450 and 550 Gy). The treated and untreated seeds (control) were sown in planting bags (under field condition) to raise M1 plants. Four treatments: V1D5, V2D3, V3D2 and V3D4 (from M1 plants) were selected and bulked to obtain M2 populations. The results of M1 revealed four mutant fruit traits: multicarpellate capsule, multiple capsule per leaf axil, indehiscent capsule and terminal capsules. The highest frequencies of the traits in M1 generation were 2.50×10-2, 9.17×10-2, 1.67×10-2and3.33×10-2 respectively. The highest branching (7) was from NCRIBEN-01M, while the least (2) was from NCRIBEN-04E. The M2 plants were grouped into eight M2 lines. The dose range (250-550 Gy) was proved to be effective in inducing viable mutations in sesame.

Genetic variability at 7 blood-group and 10 biochemical genetic loci was examined in 3 South African horse breeds, the Nooitgedacht, Boerperd and Basuto Pony. Observed heterozygosity for these breeds was intermediate for domestic horses, with the highest heterozygosity in the Boerperd and the lowest in the Basuto Pony. The 3 breeds show greater genetic similarity to each other than to other domestic horse breeds. Compared to other breeds, the South African breeds show greater genetic similarity to breeds such as the Thoroughbred, Holstein, Trakehner and Hanovarian and also to North American breeds such as the Saddlebred, Standardbred and Morgan Horse.

In this work, RAPD molecular markers were used to access the genetic variability and to study the inter and intraespecifc relationship in a group of 37 species, including 56 individuals. A total of 15 RAPD primers were selected for DNA amplification. From a total of 221 bands analyzed, 209 (95%) were polymorphics. The level of interespecifc genetic similarity ranged from 37% between Catasetum complanatum and Catasetum laminatum to 83% between Catasetum triodon and Catasetum uncatum. The intraspecifc genetic similarity varied 88% for the individuals of Catasetum triodon to 93% between the individuals of Catasetum atratum and Catasetum macrocarpum. These results would contribute to understand the genetic relationship in Catasetum, to define the strategies to establish a germplasm core collection for the genus and to provide support for breeding programs.

Maytenus ilicifolia is a medicinal plant largely used in the South Brazilian folk medicine. The aim of this study was to quantify the intra and inter populational genetic variability in three populations of M. ilicifolia, focusing on the genetic conservation of this species, which has been threatened by anthropic action. RAPD (Random Amplified Polymorphic DNA) markers were used to analyze 30 plants of each of the three populations collected in the Alto Uruguai Gaúcho region. Fourteen selected primers generated a total of 158 bands, 71.5% of which were polymorphic. The comparison of Jaccard’s distances showed that the intra populational variation was higher than the inter populational variability, and cluster analysis allowed the separation of the three populations. Just 7.6% of the bands were specific of at least two populations. Data indicate that the analyzed M. ilicifolia populations represent a single genetic pool, and therefore any of the population thoroughly can represent the overall genetic variability of the species in the sampled region.

The genetic variability of 257 sheep from 10 breeds; North Country Cheviot (NCC), Cheviot (CHE), Dorset (DOR), Suffolk (SUF), Scottish Blackface (SBF), Texel (TEX), Romanov (ROM), Finnish Landrace (FIN), Icelandic (ICE) and Red Masai (MAS) was assessed using 10 microsatellite loci. The average number of alleles per locus was 4.3 in ROM, 5.0 in MAS, and spanned a narrow range (5.4 to 6.0) in the other breeds. Estimates of expected heterozygosity (HE) of the breeds varied within 0.05 point of each other (0.62 in FIN to 0.67 in CHE), except for ROM (0.53) which was lower (P < 0.05) than those of the other breeds, and in MAS (0.57), which was lower (P < 0.05) than those of NCC, CHE and SBF. Estimates of observed heterozygosity (HO) of the breeds were the lowest in MAS, ROM and SUF (0.50 to 0.53) and the highest in NCC and CHE (0.64 and 0.67). The HE was greater (P < 0.01) than HO only in SUF. The results suggest that there have not been drastic losses of genetic variability in the intensely selected breeds. The low genetic variability of ROM was probably due to a small number of animals imported to North America. The British breeds (NCC, CHE, SUF, DOR, SBF) were genetically close to each other, as were the North European breeds (ROM, ICE, FIN). MAS was remotely related to the British breeds, but it was surprisingly close to the North European breeds. TEX was more closely related to the British breeds than to the North European breeds. More than 90% of 1000 simulated individuals from each breed were assigned to the correct breed, indicating that this panel of markers is useful for the identification of breed membership of individual animals, and could be used to protect the integrity of registered breeds.
