The Antioxidant Resveratrol from Polygonum Cuspidatum Reverses Memory Impairment and Brain Oxidative Stress in SAMP8 Mice

Resveratrol (RVT) is a phytoalexin polyphenolic compound found in various plants, including grapes, berries and peanuts. Recently, studies have documented various health benefits of resveratrol including cardiovascular and cancer-chemopreventive properties. The aim of the present study was to demonstrate the effects of resveratrol on the learning and memory impairment. The senescence-accelerated mice (SAM) were introgastric gavage administrated resveratrol (25,100mg/(kg•bw)) for 60 days. The learning and memory behavior was assessed using open-field test while the parameters of oxidative stress assessed were malondialdehyde (MDA) and superoxide dismutases (SOD).The results showed that resveratrol significantly improved the learning and memory ability in open-field test. Further investigation showed that resveratrol restored SOD levels, but decreased MDA level in the mouce brain. These results indicated that the pharmacological action of RVT may offer a novel therapeutic strategy for the treatment of age-related conditions.

Objective: The aim of the present study is to investigate the effect of paradoxical sleep deprivation (SD) on learning and memory impairment and anxiety-like behavior in female Wistar albino rats. Methods: Eight-arm radial maze, open-field test, and light and dark test were used to assess the animals learning and memory and anxiety-like behavior. Results: SD associated with weaker learning and memory and increased anxiety- and depressive-like behavior in animals. Conclusion: Animals were exposed to SD showed learning and memory impairment and also exhibited increased anxiety- and depressive-like behavior when compared to control animals.

The present study deals with the evaluation of potential effects of Eclipta alba (EA) in memory impairment of mice. Memory impairment was induced by scopolamine (3 mg/kg, i.p) in animals. To assess learning and memory in mice Morris water maze test was employed. The acetylcholinestrase enzyme (AChE) activity in brain was measured to evaluate the central cholinergic activity. The levels of thiobarbituric acid-reactive species (TBARS) and reduced glutathione (GSH)in brain were estimated to assess the degree of oxidative stress. Scopolamine treatment produces significant impairment of learning and memory in mice, as reflected by a significant decrease in MWM performance. Scopolamine also produced a significant enhancement of brain AChE activity and brain oxidative stress (increase in TBARS and decrease in GSH) levels. EA (300 and 600 mg/kg,oral) significantly prevented scopolamine-induced learning and memory deficits along with decrease of scopolamine-induced rise in brain AChE activity and brain oxidative stress levels. It may be concluded that Eclipta alba has significant protective action against scopolamine induced memory deficits in mice that can be attributed to its anti AChE and anti oxidant actions. Abstract Aim: To examine the protective outcome of quercetin against streptozotocin induced dementia in adult zebrafish. Materials and method: In this study, adult zebrafish (weighing 470-530 mg) were subjected to the streptozotocin administration (300 mg/kg; i.p) followed by a quercetin (50 & 100mg/kg i.p) pre-treatment before 24 hr of streptozotocin administration followed by blood glucose level measurement, behavioral parameters (light-dark test, novel diving test, open field test and T-maze test), biochemical parameters (lipid peroxidase, reduced glutathione, nitrite and AChEs activity), molecular and histopathological analysis. Results: In light-dark test, streptozotocin treated zebrafish shown their preference in dark compartment as compared to normal group. In novel diving tank, streptozotocin zebrafish spent less time and decrease in total number of entries to the top zone when compared to the normal group. In the T-maze test, streptozotocin treated zebrafish has shown significantly more time spent in unfavorable zone and less time spent in favorable zone with increase in total latency as compared to normal group. However, in the open field test no significant effect was seen among any group. Moreover, pre-treatment with quercetin (50 mg/kg and 100 mg/kg) significantly ameliorated the memory deficits in all the behavioral, biochemical and molecular parameters as compared to animals treated with streptozotocin. Additionally, results of histopathology studies showed less disruption in neuronal cells in quercetin treated group when compared to STZ treated animals.Conclusion: Pre-treatment with quercetin ameliorates streptozotocin induced dementia in adult zebrafish by preventing oxidative stress and neuroinflammation.

Desynchronosis may be the cause of many diseases. Oxidative stress plays an important role in the pathogenesis of various diseases. The present study investigates the effect of constant light on biomarkers of oxidative stress and content of glial intermediate filaments protein in the brains of old rats. We found that desynchronosis led to development of oxidative stress in the hippocampus, cerebral cortex and cerebellum of old rats. Prolonged continuous lighting led to an increase in the content of TBA-reactive products in all studied regions of the brains of old rats. This indicates an activation of lipid peroxidation and oxidative stress. Significant changes in the content of TBA-reactive products were found in the departments responsible for the functions of the higher nervous activity, particularly in the hippocampus and cerebral cortex. The level of restored glutathione in all three regions of the brain decreased in the group of rats kept under constant illumination in comparison with the control group. The results of the indicators of locomotor and orienting-investigative activity of the animals in the "open field" test revealled changes in the indices for desynchronosis. This showed a reduction in locomotor activity, inhibition of exploratory activity and development of emotional stress. In the brains of old rats kept under constant illumination a significant increase in the content of glial fibrillary protein (GFAP) was shown. There was a significant increase in the intensity of the polypeptide zone 49 kDa in the filamentous and soluble fraction of the cerebellum and hippocampus. This fact indicates that desynchronosis activates fibrillogenesis in glial cells. At the same time, there is degradation of polypeptides GFAP with Mr in the field of 46 kDa. Melatonin is a universal adaptogen that regulates the function of many body systems. The amount of melatonin which is synthesized depends on the illumination mode. Violation of the global mode reduces the amount of melatonin and leads to the development of desynchronosis, which may be the cause of many diseases. The administration of melatonin helped reverse the changes – raising the level of restored glutathione and preventing the growth of the content of peroxidation products and indices of "open field" test, and also decreasing the degradation of GFAP and the amount of protein. The results indicate the protective effect of melatonin, showing reductions in glial reactivity and in the level of oxidative stress in the brains of old rats subject to desynchronosis.

Background and Aims: The therapeutic effects of the olibanum, the resin of Boswellia serrata (B. serrata) from the Burseraceae family in inflammatory disease have been reported. There are more than 200 active ingredients in this resin, including Boswellic acid. It is proposed that aqueous extract of B. serrata can improve memory impairment induced by cerebral inflammation result in the administration of lipopolysaccharide (LPS). Materials and Methods: In this study, after the treatment of rats with LPS, brain toxicity induction was performed, and finally, the behavioral tests were evaluated. Following cerebral inflammation induction and treatment, behavioral performance biochemistry tests and molecular methods were assessed in all groups. Results: LPS administration increased the duration and distance to find the platform in the Morris water maze test compared to the control group in 5 days (p<0.05 to p<0.001). Furthermore, LPS reduced the peripheral, central, and total locomotion compared to the control group (p< 0.001) in the open field test. Pretreatment with both doses of aqueous extract of B. serrata enhanced performances of the rats in Morris water maze (p<0.05 to p<0.01) and open field test (p<0.01 to p<0.001). LPS also increased hippocampus Interleukin-6, malondialdehyde levels (p<0.001). Conclusion: Aqueous extract of B. serrata can be a useful drug in memory impairment caused by LPS-induced inflammation.

Introduction: The effectiveness of antioxidants on learning and memory improvement has been shown, previously. Due to the high level of antioxidants, available in Origanum vulgare, the present experiment aimed to examine the effect of aqueous extract of O. vulgare on passive avoidance learning (PAL) in male Wistar rats. Methods: This study was performed on 30 male Wistar rats weighing 250 to 290 g. The rats were randomly assigned into five groups (n=6), as follows: the control, sham (saline), and three groups treated with different doses of O. vulgare extract (150, 250, and 350 mg/kg). The saline or extract was administered via daily oral gavage for 14 days. The groups were then subjected to the passive avoidance task, and their behaviors were recorded. The rats’ locomotor activity was also measured using the open field test. Results: The number of trials to acquisition was significantly lower in the "O. vulgare (350 mg/ kg)" group than the control group. The step-through latency and the time spent in the dark compartment in the retention test, was significantly higher and lower in the "O. vulgare (250 and 350 mg/kg)" groups than the control group, respectively. No significant differences were found in the distances traveled among the experimental groups in the open field test. Conclusion: Aqueous extract of O. vulgare can enhance learning and memory. The high levels of antioxidants in O. vulgare extract may be responsible for its effectiveness in learning and memory.

Elenine is the aglycone of elenoside, a cytotoxic arylnaphthalene lignan (NSC 644013-W/1) derived fromJusticia hyssopifolia. (Family: Acanthaceae). Elenoside is a β-D-glucoside, with a similar chemical structure to etoposide, exhibiting central depressant activity. In the present study, elenine was given to mice and rats at doses of 10, 20, and 40 mg/kg. Acute toxicity (24 h) and general behaviour in mice was studied as well as its effects on muscular relaxant activity, locomotor activity (Varimex test), and the open-field test and were compared with 10 mg/kg of chlorpromazine. Elenine produced a reduction in the permanence time in muscular relaxant activity (traction test). Spontaneous activity was lower in the Varimex test. The ambulation and rearing were lower compared with the control group, and an increase in boluses was observed in the open-field test. Thus, it can be concluded that elenine has central sedative effects at lower doses than those used with elenoside and has a possible application in conditions of anxiety.
