Parameters Optimization of Two-Phase United Anaerobic Fermentation Hydrogen-Methane Production

The two-phase united anaerobic fermentation has been paid more and more attention in recent years. But how to increase the biogas production yield and the degradation rate of organic matter needs more research. The batch anaerobic fermentation studies demonstrated the feasibility of H2 and CH4 generation utilizing kitchen wastewater as substrate, and the vaccination rate, dilution rate, substrate concentration were optimized. The conclusion showed that dilution rate is the most important factor, following by kitchen wastewater concentration and inoculum volume. The most excellent program of the organic load, the inoculation volume and the dilution volume are 150 +300 ml, 135 ml, 225 ml, respectively. Under these conditions the hydrogen and methane production rate were the highest of 4.15 ml/ml,2.55 ml/ml (calculated as the wastewater volume), respectively. The acid-forming pathways of kitchen wastewater was typical butyric- propionic acid mixed fermentation type. The appropriate dilution could decrease the concentration of the total acids and increase the biogas production yield.

In order to improve the rate of degradation of cellulose in corn straw, the study has an important significance that compost corn straw with inoculating high-efficient microbe agents. The experiment inoculated a cellulose-degrading strain F2 which was screened from compost into compost pretreatment, the VS of corn straw reduced from 93.14% to 71.69% after 15 days, the content of cellulose reduced from 34.12g·kg-1 to 25.66g·kg-1, the rate of degradation was 24.79% which was 10.60% higher than those without the strain. An anaerobic fermentation experiment was carried out with the two groups of composted corn straw and mixed pig feces with a certain ratio, and investigations of biogas production, pH, content of volatile fatty acids(VFA) and rate of methane production were conducted. The results were that the corn straw composted with the cellulose-degrading strain peaked 4 days earlier, the maximal daily biogas production was 1470mL, the cumulative biogas production reached 23641mL which was 16.87% higher and operated stably earlier. The study showed that the cellulose-degrading strain had a strong capacity to degrade cellulose in corn straw, and then improved the performance of anaerobic digestion.

The changing parameters, as the biogas production rate, the methane production rate, the cumulative biogas amount, the cumulative methane amount, the biogas composition, pH etc. in high temperature anaerobic fermentation of chicken manure and stalks were analyzed by experiments with different mass ratios of chicken manure or livestock manure and stalks with a high C/N ratio. The methane production mechanism of high temperature anaerobic digestion of chicken manure and stalks was discussed in detail. It showed that not only the biogas production rates but also the methane production rates of R1–R7 demonstrated the trend of initial increase and then decrease after 50 d of high temperature anaerobic digestion. Besides, the gas production of R1 with pure chicken manure stopped on the 30th d of the reaction. The gas production of other groups R2–R7 also stopped on the corresponding 34th, 36th, 36th, 37th, 37th, and 37th day, respectively. At the end of the reaction, the cumulative biogas amounts and the cumulative methane amounts of R1–R7 were 411.58 and 269.54, 459.91 and 314.41, 425.32 and 294.11, 401.85 and 272.54, 382.63 and 257.07, 363.04 and 218.16, and 257.15 and 160.10 N ml/(g VS). The biogas slurry pH of R1–R7 all demonstrated a trend of initial decrease and then increase, e. g., pH of R2 reached the minimum of 5.94 on the 5th day. pH values of other groups were between 6.01 and 6.39. After the addition of 4 g of sodium bicarbonate on the 7th day, biogas slurry pH of R1–R7 all increased. pH was maintained between 7.16 and 7.44 until the end of the reaction.

The present study investigates the thermophilic biohydrogen and methane production from olive pulp, which is the semi-solid residue coming from the two-phase processing of olives. It focussed on: a) production of methane from the raw olive pulp; b) anaerobic bio-production of hydrogen from the olive pulp; c) subsequent anaerobic treatment of the hydrogen-effluent with the simultaneous production of methane; and d) development of a mathematical model able to describe the anaerobic digestion of the olive pulp and the effluent of hydrogen producing process. Both continuous and batch experiments were performed. The hydrogen potential of the olive pulp amounted to 1.6 mmole H2 per g TS. The methane potential of the raw olive pulp and hydrogen-effluent was as high as 19 mmole CH4 per g TS suggesting that: a) olive pulp is a suitable substrate for methane production; and b) biohydrogen production can be very efficiently coupled with a subsequent step for methane production.

The traditional pig manure wastewater treatment in Taiwan has been low in methane production efficiency due to unstable influent concentration, wastewater volume, and quality. Two-stage anaerobic systems, in contrast, have the advantage of buffering the organic loading rate in the first stage (hydrolysis-acidogenesis phase), allowing a more constant feeding rate to the second stage (methanogenesis phase). Response surface methodology was applied to optimize the operational period (0.5–2.0 d) and initial operational pH (4–10) for hydrolysis and acidogenesis of the swine manure (total solid 5.3%) at 35 °C in batch operation mode. A methanogenesis verification experiment with the optimal condition of operational period 1.5 d and pH 6.5 using batch operation resulted in peak volatile acid production 7 g COD/L, methane production rate (MPR) 0.3 L-CH4/L-d, and methane yield (MY) 92 mL-CH4/g-CODre (chemical oxygen demand removed). Moreover, a two-stage system including a hydrolysis-acidogenesis reactor with the optimal operating condition and a methanogenesis reactor provided an average MPR 163 mL/L-d and MY 38 mL/g volatile solids, which values are 60% higher than those of a single-stage system; both systems have similar dominant methane-producing species of Firmicutes and Bacteroidetes with each having around 30%–40%. The advantages of a two-stage anaerobic fermentation system in treating swine manure for biogas production are obvious.

Produksi biogas pada skala rumah tangga umumnya menggunakan teknologi fermentasi anaerobik di dalam satu biodigester (satu tahap), yang mengakomodasi dua tahap utama prinsip pembentukan biogas, yakni tahap asetogenesis dan tahap metanogenesis. Permasalahan yang muncul dari penggunaan digester biogas satu tahap adalah ketidakseimbangan proses fermentasi (peningkatan laju beban organik, waktu retensi senyawa organik yang lebih cepat, dan produktivitas biogas yang menjadi tidak maksimal). Untuk mengatasi hal tersebut, dilakukan penelitian yang bertujuan melakukan produksi biogas melalui sistem fermentasi anaerobik dua-tahap (two-stage anaerobic digestion), yang didukung dengan pengaturan pH pada proses metanogenik. Pada penelitian ini telah dilakukan proses aklimatisasi (aktivasi) bakteri yang menunjang proses asetogenik dan metanogenik pada skala laboratorium (19 L), dan selanjutnya menjadi inokulum untuk proses fermentasi skala pilot dengan kapasitas biodigester asetogenik 125 L dan metanogenik 500 L. Hasil proses aklimatisasi bakteri asetogenik pada media kotoran sapi menunjukkan adanya kestabilan pH yang dibutuhkan untuk reaksi asetogenik, yaitu pada kisaran pH 5-6, sedangkan kontrol menunjukkan perubahan pH yang masih ada di rentang pH netral yaitu 6-7. Kotoran sapi yang telah melalui proses asetogenik selama 2 minggu (pH awal 5,5), menjadi bahan baku pembuatan biogas pada digester metanogenik. Hasil dari proses metanogenik menunjukkan terjadinya peningkatan volume biogas dan komposisi gas metana (CH4) di dalam biogas. Komposisi CH4 tertinggi diperoleh pada hari ke-20 yakni 74,82% dengan volume produksi biogas tertinggi ada pada hari ke-22, dengan laju 8,87 L/hari. Potensi energi tertinggi yang diperoleh mencapai 217,66 kJ/hari.Generally, biogas production on the household scale is using one-stage anaerobic fermentation technology, which accommodates two main processes of biogas production, namely acetogenesis and methanogenesis. An obstacle of using one-stage biogas digester is the imbalance of the fermentation process that indicated by the increase of organic load rate and shorter retention time that lead to un-optimal biogas productivity. This research undertook the application of two-stage anaerobic digestion, supported by adjusting the initial pH for both acetogenic and methanogenic processes. Firstly, the research initiated by acclimatization (activation) process of acetogenic and methanogenic bacteria via fermentation in laboratory scale (19 L) digesters, separately. The results of acetogenic bacteria acclimatization process on cow dung media showed the pH stability needed for the reaction acetogenic, in the range of 5-6, while the control showed the pH changes still in the neutral pH range (6-7). The substrate from lab-scale acetogenic and methanogenic digester, then used as a starter for pilot-scale digester (125 L and 500 L, respectively). The mixture of water and cow dung were adjusted at initial pH 5.5 on acetogenic digester for 2 weeks. Those material were used for biogas production in the methanogenic digester. The result of the methanogenic process showed an increasing volume of biogas and the composition of methane (CH4) in the biogas. The highest CH4 composition was obtained on the 20th day, which reached 74.82%, and the highest volume of biogas production was at day 22, with the rate of 8.87 L/day. The highest energy potential obtained was 217.66 kJ/day.

<p class="abstrak2">Cellulose from abundant newspaper waste could be transformed into methane through anaerobic fermentation. This research was carried out to compare the gas production including methane between samples containing feces and waste paper mixture as inoculum and substrate, respectively and added with and without BS4 enzyme. The enzyme was produced in Indonesian Research Institute of Animal Produce (IRIAP) by growing Eupenicillium javanicum BS4 in coconut meals. There were three treatments,  i.e., 30% manure (M30); 15 %  manure + 15 % paper waste  (MP 30); MP30 + 3 mL BS4 enzyme equal to 0.42 U/g dry matter (MPE30) The percentage of waste papers addition in feces was calculated on dry matter (DM) basis and every treatment had five replications. Total gas and methane productions were measured weekly, while dry matter losses were determined during 5 week fermentation. Interactions between treatments and incubation time were analyzed using completely randomized design each week. Kind of substrates influenced both total gas and methane productions during incubation time. Both waste papers and enzyme addition enhanced gas production. The highest total gas and methane productions for five weeks incubation were highly significantly observed (P&lt;0.01) in MP30 and MPE30 compared to M30. Addition of enzyme significantly increased total gas and methane productions in the first week. The highest methane and total gas yield/g dry matter were obtained by BS4 enzyme addition. It was concluded that BS4 enzyme is good in accelerating and increasing the transformation efficiency of waste paper and manure mixture for biogas production.</p><p><strong>Key Words</strong>: Methane, Fibrenolytic-Enzyme, Waste Papers, Cattle Manures</p>

POME or palm oil mill effluent is currently still a waste problem that has not been utilized optimally. POME waste has the potential for renewable energy in the form of biogas, but some research results have shown that production is not optimal, so the addition of cow dung needs to be done to increase biogas production because methanogen bacteria found in cow dung help to maximize the anaerobic fermentation process and methane production. This research was conducted to determine the potential for biogas production from a mixture of POME and cow dung for 25 days by conducting a study of the biogas production process. The results of this study indicate that the biogas pressure increases with the addition of the loading rate, which is 101.102 N/m2/day to 101.107 N/m2/day with a daily biogas production of 0, 24247 liters/day with a total accumulation of biogas production for 25 days of 6.1 liters.

This paper performs a batch experiment for pre-acidification treatment and methane production from chicken manure by the two-stage anaerobic fermentation process. Results shows that the acetate was the main component in volatile fatty acids produced at the end of pre-acidification stage, accounting for 68% of the total amount. The daily biogas production experienced three peak period in methane production stage, and the methane content reached 60% in the second period and then slowly reduced to 44.5% in the third period. The cumulative methane production was fitted by modified Gompertz equation, and the kinetic parameters of the methane production potential, the maximum methane production rate and lag phase time were 345.2 ml, 0.948 ml/h and 343.5 h, respectively. The methane yield of 183 ml-CH4/g-VSremoved during the methane production stage and VS removal efficiency of 52.7% for the whole fermentation process were achieved.
