Acute heat stress (AHS) modifies the structure of myofibrils affecting functional properties of meat, mainly the water holding capacity. This experiment aimed to identify changes in proteolysis and migration between the myofibrillar and sarcoplasmatic fractions due to pre-slaughter AHS. Myofibrillar fragmentation index (MFI), SDS-PAGE, western blot of vinculin (WB) and shear force (SF) were determined. Six hundred broilers (Gallus gallus) were slaughtered in three different days (ST). In each ST, groups of ten animals were placed in transport crates and submitted to AHS (35ºC, 75 - 85% RH) for 2 hours. Simultaneously, the non-stressed broilers (NS) were kept in thermoneutral environment (22ºC, 83 ± 6.6% RH) within the crates in the same density. After slaughter, the breast muscles were kept refrigerated until the withdrawal of all samples (0, 1, 2, 6 and 24 hours after slaughter). Sampling within AHS and NS birds was collected according to lightness value (normal L* < 49, and high > 51), except for determination of MFI and SF. The lightness was used later to perform SDS-PAGE and WB analyses. MFI kinetics showed that the fragmentation rate was superior in animals NS, indicating that AHS can harm proteolysis and rate of myofibrillar fragmentation. However, the extent of fragmentation did not change, as well as SF values. SDS-PAGE for Troponin fragments indicated a differentiated pattern between AHS and NS. The WB did not show alterations in vinculin fragmentation. Modifications in sarcoplasmatic fraction are observed in meat with high L*values, independent of environmental condition.

Abstract The values obtained by the isotope method for the direct determination of oxygen in rubber agree very nicely with the values obtained for the gain in weight on oxidation. The values obtained by the Schütze-Unterzaucher method, reported herein, are all low. This study indicates that the Schütze-Unterzaucher may give low oxygen values for compounds which form non-vaporizable carbon residues, such as rubber.

Aspergillus fumigatus, Mucor rouxii, and Sporormia minima have been isolated from the rumen contents of sheep grazing permanent pasture at Nappan, Nova Scotia. To determine the ability of these fungi to survive and grow at the low oxygen partial pressure present in the rumen, a method of determination of oxygen, nitrogen, and carbon dioxide has been developed. The lowest partial pressures of oxygen [Formula: see text] and nitrogen that could be detected were 0.0005 cm Hg and the precision of the determination was ±0.001 cm Hg. Carbon dioxide was determined with slightly less precision than achieved for oxygen and nitrogen. Using this method, respiration was detected in cultures of all the fungi named at [Formula: see text] Hg and growth was observed at [Formula: see text] Hg in the case of M. rouxii. It is concluded that all these fungi are capable of survival in the ovine rumen.

Sahidin A, Zahidah, Herawati H, Wardiatno Y, Setyobudiandi I, Partasasmita R. 2018. Macrozoobenthos as bioindicator ofecological status in Tanjung Pasir Coastal, Tangerang District, Banten Province, Indonesia. Biodiversitas 19: 1123-1129. Theexistence of macrozoobenthos organism allow for bioindicator of ecological status and detected water pollution in coastal environmentwith observing the response, diversity, and abundance. The one of biological method for water quality analysis is AZTI Marine BioticIndex (AMBI). This research aims to determine water quality status in Tanjung Pasir water coastal Tangerang, Banten. The research wasconducted in April - August 2014 in Tanjung Pasir Coastal, Tanggerang, Banten with purposive sampling method and be forwarded withwater pollution analysis with AZTI Marine Biotic Index (AMBI) model. The research showed 4.552 individual of macrozoobenthosfound divided into 52 species with the average of diversity amount 634 ind./m2 with dominated by bivalve species Nuculana bicuspidate22.89%, it is a bivalve an extreme species that can live in waters with very low oxygen concentrations and hight pollution. The result ofAMBI (4.01 to 5.02) indicates water pollution in Tanjung Pasir Coastal classified in medium-heavy pollution and concentred in coastalzone. Polychaeta is a key bioindicator in the determination of water pollution and ecological status in Tanjung Pasir coastal.

The work presents the results of investigations of the effect of moderate heat stress on the acidobasal status in high-yield dairy cows in early lactation. Improving performance in high-yield dairy cows increases their inclination toward metabolic disorders. The most likely to be affected is the acid-base balance, in particular when cows are exposed to heat stress. Investigations so far have shown that the taking of urine samples and their analysis is the best and fastest way to diagnose disorders in the acid-base balance. Investigations were carried out on 7 clinically healthy cows of the Holstein- Friesian breed in the phase of early lactation, 30 to 40 days (on days 30, 33 and 40 ) following calving during the summer period, during the month of July, when there were significant variations in daily and nightly temperatures. The cows were in the second and fourth lactation, the annual milk yield was 8000 L milk per cow. The average daily milk production in the early phase of lactation ranged from 35 to 40 L. The cows were fed mixed rations (TMR) twice daily. Lucerne hay in limited quantities was given to the cows prior to the mixed feed ration. The ration for this animal category was optimized on the grounds of the daily milk production. The balance of cations and anions in the feed ration stood at 95 mEq/kg DM. The results clearly demonstrate the cows' response to moderate heat stress through the defense parameters in urine (urine pH, NABE (net-acid-base-excretion), acids, bases, ammonium ion (NH4). The determination of the kidney NABE yields more correct data than the urine pH on acidotic conditions. The results of examinations of the urine pH do not show any digressions from physiological values and are approximately the same in all cows during the investigated periods. Normal NABE values are from 100-200 mmol/L. Burdening with acid products results in a NABE range from 0-100mmol/L, and metabolic acidosis results in NAB<0 mmol/L. NABE in the cows in this experiment revealed that the organism is burdened by acid metabolic products and it stood at 58.28?27.96 mmol/L on day 30, 69.28?29.89 mmol/L on day 33, and 60.60?26.88mmol/L on day 40. Consequently, it seems that NABE values are decreased in cases of acidosis due to stepped up H+ excretion and stepped up bicarbonate reabsorption.
