Background: Hospital wastewaters contain blends of inorganic, natural constituents and contaminants that carry significant health risk when released directly into the environment. The aim of this study is to investigate the correlation between faecal indicator bacteria in diarrheagenic stools and wastewaters generated in University of Medical Sciences Teaching Hospital complex, Akure, Nigeria.Methodology: Quantification of faecal indicator bacteria was carried out on diarrheagenic faecal samples collected from 55 hospitalized patients and 68 wastewater samples from the medical laboratory science and laundry units of the hospital over of period of 12 weeks. Standard membrane filtration technique was performed using membrane intestinal enterococcus (m-ENT), membrane faecal coliform (m-FC), membrane lauryl sulphate (MLSA), eosin methylene blue (EMB) and Salmonella-Shigella (SS) agar plates, which were incubated at 37ºC for 24 hours (MLSA, EMB and SSA), 44ºC for 24 hours (m-FC); and 37ºC for 48 hours (m-ENT). Bacterial colonies on agar plates were counted and expressed as colony forming units (CFU) per 100ml of diarrheagenic stool and wastewater. Pearson’scorrelation analysis was used to determine the relationship between the level of faecal indicator bacteria in diarrheagenic stools and wastewaters at p<0.05 level of significance (and 95% confidence interval).Results: The faecal coliform counts (log 10 CFU/100ml) ranged from 1.18 to 1.54 in diarrheagenic stools, 1.32 to1.64 in laboratory wastewater and 1.08 to 2.19 in laundry wastewater. Escherichia coli count (log 10 CFU/100ml) ranged from 1.08 to 1.40 in diarrheagenic stools, 1.20 to 1.86 in laboratory wastewater and 0.30 to 1.81 in laundry wastewater. Intestinal enterococci count (log 10 CFU/100ml) ranged from 0 to 0.30 in diarrheagenic stools, 0.78 to 0.90 in laboratory wastewaters and 0.48 to 1.11 in laundry wastewaters. Pearson’s correlation co-efficient showed that all the faecal indicator bacteria count in diarrheagenic faecal samples exhibited positive correlation with those in laboratory wastewaters, but not with those from laundry wastewaters.Conclusion: The findings suggest that diarrheagenic stools should be properly disinfected after the performance of laboratory tests to prevent transmission of potential pathogens, and wastewater generated from hospitals should be treated prior to discharge into the environment, to prevent possible infections in the community. The objective of this study was to evaluate the usefulness of monitoring streams in Hawaii for FRNA coliphages as a reliable indicator of sewage contamination. This study was undertaken as a result of our previous findings that monitoring streams in Hawaii for traditional faecal indicator bacteria (faecal coliform, Escherichia coli, enterococci) was not useful in determining when streams are contaminated with sewage, because environmental (soil) sources rather than sewage accounted for the high concentrations of faecal bacteria in streams. Two perennial streams, sewage and soil samples were monitored for traditional faecal indicator bacteria (faecal coliform, E. coli, enterococci) and FRNA coliphages. The results showed that sewage treatment processes and disinfection drastically reduced the concentrations of traditional faecal indicator bacteria but FRNA coliphages were still present in significant concentrations in the treated sewage effluents. These results indicate that monitoring sewage effluents and environmental waters for only traditional faecal indicator bacteria may not be adequately protective of human health effects. Ambient concentrations of traditional faecal indicator bacteria in soil and streams of Hawaii were consistently high but consistently low for FRNA coliphages, indicating that monitoring streams of Hawaii for FRNA coliphages can be used to determine when streams are contaminated with sewage.

Abstract Background Hepatitis E is an enteric and zoonotic disease caused by hepatitis E virus (HEV) that is mainly transmitted via the faecal-oral route through contaminated food or the environment. The virus is an emerging infectious agent causing acute human infection worldwide. A high seroprevalence of the disease was reported in pregnant women in Addis Ababa, Ethiopia, raising significant public health concern. The presence of HEV specific antibodies were also reported in dromedary camels in the country; however, the infectious virus and/or the viral genome have not been demonstrated to date in animal samples. Methods To address this gap, a total of 95 faecal samples collected from both apparently healthy pigs of uncharacterised types (50 samples) in Burayu and Addis Ababa areas and camels (Camelus dromedarius, 45 samples) in west Hararghe were screened for the presence of HEV genome using universal primers in a fully nested reverse transcription polymerase chain reaction (nRT-PCR). The protocol is capable of detecting HEV in faecal samples from both pigs and camels. Results The nRT-PCR detected HEV genes in six (12%) pig faecal samples and one camel sample (2.2%). Therefore, the results indicate that HEV is circulating in both pigs and camels in Ethiopia and these animals and their products could serve as a potential source of infection for humans. Conclusion The detection of HEV in both animals could raise another concern regarding its public health importance as both animals’ meat and camel milk are consumed in the country. Further studies to determine the prevalence and distribution of the virus in different animals and their products, water bodies, food chain, and vegetables are warranted, along with viral gene sequencing for detailed genetic characterisation of the isolates circulating in the country. This information is critically important to design and institute appropriate control and/or preventive measures.

AbstractBackground Microbial water quality serves to indicate health risks associated with the consumption of contaminated water. Nevertheless, little is known about the microbiological characteristics of water in Lake Bunyonyi. This study was therefore undertaken to examine the spatial and temporal variations of faecal indicator bacteria (FIB) in relation to physicochemical parameters in Lake Bunyonyi. Result The FIB concentration was consistently measured during sampling months and correlated with each other showing the presumed human faecal pollution in the lake. The highest concentration values for E. coli (64.7 ± 47.3 CFU/100 mL) and enterococci (24.6 ± 32.4 CFU/100 mL were obtained in the station close to the Mugyera trading centre. On a temporal basis, the maximum values were recorded during the rainy season in October 2019 (70.7 ± 56.5 CFU/100 mL for E. coli and 38.44 ± 31.8 CFU/100 mL for enterococci. FIB did not differ significantly among the study stations (p > 0.05) but showed significant temporal variations among the months (p < 0.05) with concentrations being significantly high in wet season than dry season (U = 794, p < 0.0001 for E. coli; U = 993.5, p = 0.008 for enterococci). Spearman’s rank correlation revealed that FIB concentrations were significantly positively correlated with turbidity and DO concentration levels (p < 0.05). Approximately 97.2% of the water samples had E. coli and enterococci concentrations levels below USEPA threshold for recreational waters. Likewise, 98.1 and 90.7% of samples recorded E. coli and enterococci counts exceeding the UNBS, APHA, WHO and EU threshold values for drinking water. Conclusion The FIB counts show that the Lake Bunyonyi water is bacteriologically unsuitable for drinking unless it is treated since the FIB pose health risks to consumers. Besides, the water can be used for recreational purposes.

A study was conducted between May and August, 2018 to determine the prevalence of intestinal helminths of pigs in Chikun and Jema’a Local Government Areas, Kaduna State. 203 faecal samples were collected, processed and examined under light microscopy. Intestinal helminths recorded were: Ascaris suum (57.1%), Oesophagostomum dentatum (37.0%), Hyostrongylus rubidus (14.8%), Metastrongylus elongates (8.9%), Strongyloides (4.5%), Stepanurus dentatum (2.5%), Trichuris suis(1.5%), Globocephalus urobuslatus (2.0%) and Physocephalus sexalatus (1.5%). The overall prevalence of helminth infection in both LGAs was 71.9%. Pigs in Jema’a LGA had significantly higher prevalence (87.9%) than pigs from Chikun LGA (56.7%). Female pigs had relatively higher (74.4%) infection than male pigs (68.6%). This pattern was also reflected in Chikun LGA where female pigs had a prevalence of 63.9% compared to 46.5% in males, however, the reverse was the case in Jema’a LGA where males had 90.7% prevalence and females 85.7%. In Chikun LGA, the highest prevalence of 58.6% was recorded among pigs aged 25-36 months, followed by pigs aged 5-12 months with prevalence of 57.1%, and the least prevalence of 55.0% was recorded in pigs aged 13-24 months. The prevalence of intestinal helminthosis in pigs tends to decrease with age in Jema’a LGA. The presence of these parasites in pigs examined indicates a potential public health problem in Chikun and Jema’a Local Government Areas of Kaduna state. Enlightenment of pig farmers on the need for periodic veterinary care and restriction of stray pigs through legislation formulation and enforcement are recommended as control measures
