Summary This study was made to evaluate, for use with dry milk, several media used in selective quantitative analyses for staphylococci in food products. The principal problems involved in culturing staphylococci from dry milk are (a) growth of Bacillus organisms and (b) obtaining good recovery of coagulase-positive staphylococci while eliminating growth of micrococci and coagulase-negative staphylococci, or at least providing a means of differentiating colonies of these organisms on the plates. Tellurite-polymyxin-egg yolk (TPEY) agar produced highest counts of coagulase-positive staphylococci for two-thirds of 40 strains tested in comparisons with Staphylococcus 110 (S110) and tellurite-glycine (TG) media. Highest counts of coagulase-negative staphylococci and of micrococci were found on S110 medium for 9 to 12 cultures. TG agar gave the poorest recovery in both experiments. TG and Vogel-Johnson (VJ) media were found too inhibitory to be used for staphylococcus counts on dry milk. Mannitol salt (MS) agar reacted very similarly to S110 agar. Colonies of Bacillus were easily discernible on TPEY medium but often were mistaken for staphylococci on the media which depend upon salt concentration for selectivity.

Three media for isolation of Bacillus cereus from foods were compared: mannitol-egg yolk-polymyxin (MYP) agar, polymyxin pyruvate-egg yolk-mannitol-bromothymol blue agar (PEMBA) and non-selective blood agar. Twenty-six of 45 samples of different reconstituted and incubated dry food products and 18 of 29 samples of milk and cream (incubated overnight) contained B. cereus. None of the media performed significantly better than the others as regards quantitative recovery or selectivity.

Six selective media for the detection and enumeration of coagulase-positive staphylococci werl ctudied evaluated for their efficiency in the recovery and easy recognition of five strains of Staphylococcus aureus. They were Mannitol Salt agar (MS', Tellurite Polymyxine Egg Yolk agar (TPEY),Egg Yolk Azide Agar (EYAA), Egg yolk Tellurite Glycine Pyruvate Agar (ETGPA). Milk Salt Agar (MSA), in combination with Salt Egg Yolk Agar (SEYA), and Vogel-Johnson agar (VJ), after enrichement in Trypticase Soy Broth 10V» NaCl.The recovery of the used strains was evaluated in pure cultures and after inoculation in various foods. None of the tested media was proved to be the ideal one. The media TPEY, ETGRA. and EYAA were considered as the nost appropriate for the detection of S. aureus in foods. The selective ability of eash medium is greatly depended on the strain and the food involved The simultaneoue use of two selective media is suggested if that is possible.

Methicillin-resistant Staphylococcus aureus (MRSA) strains have veterinary and public health importance as they are responsible for a wide range of difficult to treat infections and food poisoning. Two hundred samples (50 samples each of minced meat, beef luncheon, Karish cheese, and human samples (pus swab from open wounds)) were cultured, and MRSA strains were identified using disk diffusion tests and mecA gene-based PCR. A total of 35% (70/200) of the examined samples were confirmed as coagulase-positive S. aureus in minced meat (46%), beef luncheon (44%), Karish cheese (44%), and human samples (22%). The MRSA strains showed resistance to amoxicillin (91.4%), penicillin (97.1%), cefoxitin (85.7%), cephradine (82.9%), tetracycline (57.2%), and erythromycin (52.8%). More than half of the tested S. aureus isolates harbored the mecA gene. The sequence analysis of the mecA gene from the minced meat, Karish cheese, and human samples revealed high genetic similarities between the S. aureus isolates from these sources. In conclusion, our findings indicate a risk for the transmission of the mecA gene of S. aureus across the food chain between humans and animal food products. Further studies should focus on finding additional epidemiological aspects of the MRSA strains in food chain.

AbstractMicroRNAs (miRNAs) are short, non-coding RNAs, 21–23 nucleotides in length which are known to regulate biological processes that greatly impact immune system activity. The aim of the study was to compare the miRNA expression in non-infected (H) mammary gland parenchyma samples with that of glands infected with coagulase-positive staphylococci (CoPS) or coagulase-negative staphylococci (CoNS) using next-generation sequencing. The miRNA profile of the parenchyma was found to change during mastitis, with its profile depending on the type of pathogen. Comparing the CoPS and H groups, 256 known and 260 potentially new miRNAs were identified, including 32 that were differentially expressed (p ≤ 0.05), of which 27 were upregulated and 5 downregulated. Comparing the CoNS and H groups, 242 known and 171 new unique miRNAs were identified: 10 were upregulated (p ≤ 0.05), and 2 downregulated (p ≤ 0.05). In addition, comparing CoPS with H and CoNS with H, 5 Kyoto Encyclopedia of Genes and Genomes pathways were identified; in both comparisons, differentially-expressed miRNAs were associated with the bacterial invasion of epithelial cells and focal adhesion pathways. Four gene ontology terms were identified in each comparison, with 2 being common to both immune system processes and signal transduction. Our results indicate that miRNAs, especially miR-99 and miR-182, play an essential role in the epigenetic regulation of a range of cellular processes, including immunological systems bacterial growth in dendritic cells and disease pathogenesis (miR-99), DNA repair and tumor progression (miR-182).

Although investigative reports have contributed to the social movement against white-collar crime, few studies assess the extent to which the media socially construct corporate violence as a “crime.” We examine this issue through a content analysis of newspaper coverage of the fire-related deaths of 25 workers at the Imperial Food Products chicken-processing plant, which resulted in the company's owner pleading guilty to manslaughter. The analysis revealed that newspaper reports largely attributed the deaths to the lax enforcement of safety regulations but did not initially construct the deaths as a crime or subsequently publicize the criminal convictions.

Abstract The aim of this study was to evaluate the prevalence and antimicrobial resistance profile of Gram-positive cocci and Gram-negative bacilli isolated from the surgical environment. All samples were collected during the intraoperative period of clean/clean-contaminated (G1) and contaminated (G2) surgery. A total of 150 samples were collected from the surgical wound in the beginning (n = 30) and end (n = 30) of the procedure, surgeon’s hands before (n = 30) and after (n = 30) antisepsis and the surgical environment (n = 30). Forty-three isolates with morphological and biochemical characteristics of Staphylococcus spp. and 13 of Gram-negative bacilli were obtained. Coagulase-negative staphylococci (85.71% [18/21]), coagulase-positive staphylococci (9.52% [2/21]) and Pseudomonas spp. (47.52% [1/21]) in G1, and coagulase-negative staphylococci (40% [14/35]), coagulase-positive staphylococci (20% [7/35]), Proteus spp. (17.14% [6/35]), E. coli (8.57% [3/35]), Pseudomonas spp. (2.86% [1/35]) and Salmonella spp. (2.86 [1/35]) in G2 were more frequently isolated, and a high incidence of multidrug resistance was observed in coagulase-negative staphylococci (87.5% [28/32]), coagulase-positive staphylococci (100% [11/11]) and Gram-negative bacilli (76.92% [10/13]). Methicillin-resistant Staphylococcus spp. accounted for 83.72% (36/43) of the Staphylococcus strains. Gram-negative bacilli cefotaxime-resistance constituted 81.82% (9/11) and imipenem resistance constituted 53.85% (7/13). The high rate of resistance of commensal bacteria found in our study is worrying. Coagulase-negative staphylococci are community pathogens related to nosocomial infections in human and veterinary hospitals, their presence in healthy patients and in veterinary professionals represent an important source of infection in the one health context. Continuous surveillance and application of antimicrobial stewardship programs are essential in the fight against this threat.

The aim of this paper is to determine the awareness measures of consumers from Eastern Poland and Western Ukraine towards regional food products, including consumer knowledge on regional products available in the media and their availability on the food market. The effort was made to compare consumers’ opinions on the reasons for purchasing regional food and ways of distinguishing it from conventional products, as well as on the availability of regional products. Consumer awareness—that is, making informed choices based on the knowledge we have—is a measure of attitudes and cognition, and sometimes can be directed towards the brand, which is the product’s regional designation. Therefore, it is necessary to comment that attitudes towards regionality can generate a behavioral intent. A diagnostic survey with an author’s questionnaire was used in the study, which helped to survey 1128 respondents from Eastern Poland—that is, from the Podlaskie, Lublin, and Subcarpathia regions—and 1072 from Western Ukraine, including the Volyn, Lviv, and Transcarpathia regions. Discriminant function analysis was used in statistical analysis. Both residents of Eastern Poland and Western Ukraine obtained information on regional food products from their friends or family and from television (TV), internet, and regional fairs. Consumers from both countries pointed at too many possibilities of purchasing regional products; at the same time, they paid attention to a limited number of points of sale. TV and Internet have a great promotional potential to educate young consumers focused on the purchase of regional food products.

Abstract Eleven collaborators studied the reproducibility of the method, proposed in 1965, for the isolation of coagulase-positive staphylococci. Samples studied were milk, beef and turkey samples, and fresh, headless shrimp. Based upon statistical analysis of collaborative results, the laboratory means did not vary significantly. The method is recommended for adoption as official, first action.

Abstract A collaborative study was conducted to compare proposed dry-film plating methods, using aerobic count plates and coliform count plates, to standard agar plating methods for quantifying aerobic bacteria and coliforms in dairy products. In this study, 5 food products (chocolate milk, pasteurized cheese, nonfat dry milk, evaporated milk, and vanilla ice cream), selected as representative dairy products, were analyzed by 11 collaborating laboratories. The results indicate that the dry-film plating methods are equivalent to or better than the agar plating methods. The aerobic count and coliform count dry-film plating methods have been adopted official first action.
