Significance of Psychrotrophic Strains of Serratia liquefaciens in Milk1

Enterobacter cloacae, Citrobacter freundii, Serratia liquefaciens and Escherichia coli were the predominant Enterobacteriaceae species isolated from raw milk samples collected from refrigerated bulk tank trucks at the entry of a milk processing plant. About half of the 181 Enterobacteriaceae isolated were psychrotrophs and these included 80% of E. cloacae, 80% of Klebsiella ozaenae and 62% of S. liquefaciens. S. liquefaciens grew in raw and in pasteurized milk at refrigeration temperatures, causing significant pH reduction but only slight lipolytic changes. The significant proteolytic activity of S. liquefaciens in refrigerated milk samples is assumed to have been the cause of the unclean flavor detected.

No Estado de São Paulo, Brasil, os pássaros como os canários-da-terra têm sido uma das espécies mais frequentemente resgatadas do tráfico illegal e enviadas aos centros de vida selvagem. Em situações de estresse estas aves podem ser acometidas por infecções causadas por bactérias oportunistas. Este fato é de grande importância quando é planejada da reintrodução das aves na natureza. O presente trabalho foi delineado para avaliar o estado de saúde de canários-da-terra resgatados do tráfico ilegal. Foram colhidas soabes da traqueia e da cloaca de 100 aves resgatadas durante os anos de 2012 e 2013. Os resultados obtidos revelaram alta frequência de bactérias gram-negativas nas fezes e no orofaringe dos animais, com maior frequência para os membros da família Enterobacteriaceae (97,5%). Os gêneros mais frequentes foram Escherichia coli (46,55) e Klebsiella pneumoniae (10,4%). Outros microorganismos incluindo Enterobacter cloacae, Serratia liquefaciens, Serratia spp, Klebsiella oxytoca e Citrobacter freundii também foram isolados em menor frequencia de aves assintomáticas. A presença de estirpes de Escherichia coli enteropagênicas (EPEC) e as produtoras da toxina de Shiga confirmam o risco de zoonose e a importância para saúde pública deste tipo de ave.

SummaryThe microorganisms present in raw and pasteurized milk and the sources of contamination in the milk after it had arrived at the processing plant in Addis Ababa were studied. The lowest count registered for raw milk samples was 4 × 107cfu/ml while the highest was 1 × 109cfu/ml. Pasteurized milk had mesophilic aerobic counts of 7 × 105cfu/ml as it left the pasteurizing unit, but the population increased 2- to 4-fold as a result of subsequent contamination. Of the total counts in raw milk, psychrophilic, thermoduric and thermophilic organisms made up 98·l, 1·4 and 0·5% respectively. In pasteurized milk, the amounts were 53·0, 39·5 and 7·5% respectively. Samples of milk pasteurized in the laboratory contained only 74·5% thermoduric and 25·5% thermophilic organisms. The isolates mostly belonged to the generaBacillus, Streptococcus, Lactobacillus, Arthrobacter, Alcaligenes, AeromonasandPseudomonas. Cocci were more predominant than rod-shaped bacteria. Of the rod-shaped bacteria, 73% were Gram-negative. The utensils holding the raw and pasteurized milk and the plastic sheets used for bagging the pasteurized milk contributed unusually high numbers of bacteria which were either thermoduric or thermophilic. More isolates were obtained from the pasteurized than the raw milk. The keeping quality of the pasteurized milk was found to be much lower than that of the laboratory-pasteurized milk.

Psychrotolerant endospore-forming bacteria Bacillus and Paenibacillus spp. are important spoilage organisms in fluid milk. A recently developed rpoB subtyping method was applied to characterize the diversity and phylogenetic relationships among Bacillus and related sporeformers associated with milk processing systems. Milk samples representing the processing continuum from raw milk to pasteurized products were collected from two fluid milk processing plants, held at 6°C uptothe code date that had been established by each processing plant (i.e., either 18 or 21 days), and plated for bacterial enumeration throughout storage. Bacterial colonies selected to represent the visible diversity in colony morphology on enumeration plates were examined further. Among 385 bacterial isolates characterized, 35% were Bacillus spp., and 65% were Paenibacillus spp. A total of 92 rpoB allelic types were identified among these isolates, indicating considerable diversity among endospore-forming spoilage organisms present in fluid milk systems. Of the 92 allelic types identified, 19 were isolated from samples collected from both processing plants. The same rpoB allelic types were frequently identified in paired raw milk and packaged product samples, indicating that Bacillus and Paenibacillus spp. can enter dairy processing systems through raw milk. Certain subtypes were found exclusively in pasteurized samples, including those that were temporally independent, suggesting the possibility of in-plant sources for these spoilage organisms, including through the persistence of selected subtypes in processing plants. Development of effective control strategies for the diverse array of psychrotolerant endospore-forming organisms that currently limit the shelf lives of high-temperature short-time fluid milk products will require comprehensive, integrated efforts along the entire milk processing continuum.

Foram estudados 51 casos de infecção urinária, em cães, considerando-se diversos fatores, tais como: agente etiológico, localização da infecção, fatores predisponentes, sexo, idade e raça. O diagnóstico da infecção do trato urinário (ITU) foi baseado no exame bacteriológico, sendo considerado positivo quando a amostra de urina, colhida com auxílio de cateter, apresentava acima de 105 bactérias/ml. Dos animais examinados, quatro cães apresentaram infecção mista, totalizando 55 microorganismos isolados. Escherichia coli foi a mais freqüentemente isolada (35,3%), seguida de Staphylococcus sp (23,5%), Proteus mirabilis (15,7%), Streptococcus sp (13,7%), Klebsiella sp (9,8%), Pseudomonas aeruginosa (3,9%), Enterobacter cloacae (2.0%), Citrobacter freundii (2.0%) e Providencia rettgeri (2,0%). Quanto à sensibilidade dos germes isolados frente a diversos agentes antimicrobianos, a norfloxacina e a gentamicina mostraram-se eficazes no tratamento de microorganismos Gram-negativos, enquanto a cefalotina e a nitrofurantoina foram mais eficazes contra bactérias Gram-positivas. Os animais que apresentaram maior frequência de ITU pertenciam às raças Cocker Spaniel e Pastor Alemão, envolvendo mais machos do que fêmeas com predominância de pielonefrites. Embora as infecções urinárias tivessem sido observadas em todas as idades, houve um predomínio nos cães de média idade. Observou-se ainda que a urolitíase foi um fator pré-disponente ou adjacente de ITU, envolvendo germes como Staphylococcus sp. e Proteus mirabilis naqueles casos com pH urinário alcalino.

The growth of 7 strains belonging to the order of Enterobacterales, represented by the species of Citrobacter Freundii, Enterobacter cloacae, Escherichia coli, Klebsiella oxytoca, Obesumbacterium proteus, Rahnella aquatilis, Raoultella terrigena, Serratia marcescens and Shimwellia pseudoproteus, was monitored on selected cultivation media. Three types of agars - Endo, MacConkey and Chromocult Coliform agar together with two incubation temperatures of 28 and 37 °C were tested under aerobic conditions. The aim of the study was to detect such essential enterobacteria harmful to beer that cannot be proven at 37 °C, which is the temperature usually used in operational laboratories in breweries. Our results showed that most of the tested strains of enterobacteria were able to grow at 28 °C on all selected types of agar. The exception was just the representatives detection of which is problematic at 37 °C. Nevertheless, a little or no growth was always observed on just one of the tested media.

ABSTRACT For milk hygiene and safety, the milking phase is a critical moment because it is a probable pathway for the introduction of unwanted microorganisms in the dairy chain. In particular, Listeria monocytogenes and Escherichia coli are known as possible microbial contaminants of raw sheep's milk, although extensive knowledge regarding their contamination dynamics on sheep farms is still lacking. This study aimed to examine the occurrence and concentration of these microorganisms in milk samples collected from farm bulk tanks in the region of Lazio (Central Italy) and to investigate the related risk factors. Over a period of 1 year, we collected 372 milk samples from 87 sheep farms and administered a questionnaire to acquire information regarding relevant farm management variables. L. monocytogenes was not found in any of the samples, which indicates a low occurrence of this pathogen in sheep's bulk tank milk. In contrast, E. coli was found in almost two-thirds of milk samples (61%) but at levels below 102 CFU/mL in most of them (approximately 75%). Statistical analysis indicated that, during the warmest seasons, E. coli presence is more probable and counts are significantly higher. Unexpectedly, milk collected by hand milking had a lower level of contamination. Although further studies are necessary to clarify some aspects, the reported data add to the knowledge about the occurrence of L. monocytogenes and E. coli in raw sheep's milk and will be useful for future risk assessments. Summary An 8-month survey of the bacteriological quality of bulk tank produced fluid milk supplies for four major markets in Ohio, was made utilizing the Standard Plate Count (SPC), the preliminary incubation count (PI), the thermoduric (pasteurized milk) count, and the coliform count. In terms of maximum standards of 200,000 and 100,000 organisms/ml, the SPC would have eliminated 13% and 20% of the milk samples, respectively. A SPC of 50,000/ml, a PI count of 200,000/ml, a thermoduric count of 500/ml, and a coliform count of 100/ml would have eliminated 37%, 34%, 40%, and 40%, respectively, of the samples but not all of the samples eliminated by one test were eliminated by another test. All of the tests employed showed a seasonal trend especially in the high count categories but the trend was less noticeable in the results of the preliminary incubation count. A combination of two of the methods was superior to any single bacteriological method employed in detecting unsatisfactory milk. Of the tests used, the combination of the thermoduric count (500/ml) and the coliform count (100/ml) was the most effective in the detection of unsatisfactory milk samples.
