This study was performed on an industrial dairy farm near Tehran province, Iran, on 666 lactating Holstein cows from September 2019 to June 2020 in order to eliminate the harmful effects of heat stress on the reproductive performance of the cows. The hypothesis of the recent study was that by increasing the serum concentration of progesterone (P4) in the luteal phase before insemination in the form of Presynch-Ovsynch+CIDR (POC) and G6G protocols, the pregnancy rate of cows would be increased compared to the Presynch-Ovsynch (PO) protocol. The aim of the present study was to evaluate the effects of Presynch-Ovsynch (PGF2α-14d- PGF2α-12d-Ovsynch (OVS: GnRH(GnRH1)- 7d-PGF2α(PG)-2d-GnRH)-18h-Fixed-time artificial insemination (FTAI), n=212), Presynch-Ovsynch+CIDR (similar to the PO protocol, plus the use of a CIDR for 7 days, from the start of the OVS+FTAI protocol to the time of PG injection, n=230), and G6G (PGF2α- 2d-GnRH-6d-OVS+FTAI, n=224) protocols on the reproductive performance and pregnancy rate of lactating Holstein cows. The average body condition score (BCS) for the cows was about 2.5 at the time of the onset of the OVS+FTAI program. The average daily milk production for cows at the time of the start of the OVS+FTAI program was 38.3 kg/day. On average, the studied cows were inseminated 86 days postpartum. Implementation of the G6G protocol in comparison to the PO and POC protocols increased the serum concentration of P4 at GnRH1 (P=0.04). The cows that received the G6G protocol had a greater number of corpuses luteum (CL) on their ovaries at PG in comparison to the cows in the PO and POC protocols (P=0.03 and P=0.05, respectively). For all treatment protocols, the pregnancy rate of cows with >2 CLs on their ovaries at PG was significantly higher than for cows with ≤2 CLs on their ovaries at this time. The overall pregnancy rate for all cows studied was 42%. Separately, pregnancy rates for cows in PO, POC, and G6G protocols were calculated at 36.7%, 41.7%, and 47.3% respectively. The highest and the lowest pregnancy rates were obtained in the G6G and PO protocols, respectively (P=0.03). It can be concluded that the G6G protocol increased the pregnancy rate of cows in comparison to the PO but not to the POC protocol. This result can be attributed to the increase in serum concentration of P4 at GnRH1 and the greater number of CLs at PG following implementation of the G6G compared to the PO protocol. Although CIDR administration in the POC protocol led to an increase in the pregnancy rate of cows compared to the PO protocol, possibly by increasing the synchrony rate and the quality of the ovulatory follicle, this increase was not statistically significant (P=0.09).

An important proportion of embryonic deaths in cattle occur during the critical period of embryo development and maternal recognition of pregnancy. High serum progesterone concentrations during this period are associated to higher embryo survival, and the use of eCG could enhance luteal activity. Two experiments were carried out to evaluate the effect of administering eCG at the time of removal of a progesterone-releasing device and 14 days after fixed-time artificial insemination (FTAI) in beef cattle. Multiparous suckled cows between 60 and 90 days postpartum (n = 297; Exp. 1), and two-year old prepuberal heifers (n = 689; Exp. 2) with a body condition score (BCS) between 3.5 and 5 (1 to 8 scale) were used. At the beginning of the experiments, none of females used had a corpus luteum detected by ultrasonography. Females received an intravaginal device containing 0.5 g progesterone (DIB 0.5, Syntex, Buenos Aires, Argentina) and 2 mg oestradiol benzoate i.m. (Benzoato de Oestradiol, Syntex). Eight days later, 500 μg of cloprostenol (Ciclase DL, Syntex) and 0.5 mg oestradiol cypionate (Cipiosyn, Syntex) were given i.m. and devices were removed; FTAI was performed 52 to 56 h after DIB removal. At the time of device removal, the animals were randomly allocated to 4 groups, to receive 400 IU of eCG i.m. (Novormon, Syntex) or placebo at device removal and/or 14 days after FTAI, in a 2 × 2 factorial design. Control group did not receive eCG. In Exp. 1, nose plates were placed on calves on the day of DIB insertion and left for 10 days in order to prevent suckling. Pregnancy diagnosis was performed by ultrasonography at 30 and 60 days after FTAI. Data were analysed by logistic regression. Pregnancy rate at 30 days after FTAI was affected by treatment (P < 0.05; Table 1). Late embryonic/fetal loss rates (non-pregnant females at 60 days after FTAI over those pregnant at 30 days) were 1.6% (3/182) for cows in Exp. 1 and 4.8% (14/292) for heifers in Exp. 2; no differences were detected among groups. In conclusion, administration of 400 IU of eCG at device removal increases pregnancy rate, both in anestrus cows and heifers. However, the administration of a second dose of eCG 14 days after FTAI suggests a possible effect on embryo survival within the first 30 days of pregnancy in cows but not in heifers. Further studies are required to confirm these results. This study evaluated the effectiveness of Ovsynch/FTAI protocol (day 0 fertirelin inj. 100μg I/M, day 7 cloprostenol inj. 500μg I/M, day 9 fertirelin inj. 100μg I/M, and day 10 fixed time artificial insemination 18 hours after second injection of fertirelin) in anestrous buffaloes of Chitwan, Nepal (n=14). Protocol was evaluated in terms of estrus characteristics, ovulation and pregnancy rate. Responses were analyzed by grouping the buffaloes based on type of anestrus, length of anestrus, body condition score (BCS), milking status, parity, age, management condition, follicle count on day of second GnRH treatment and site of semen deposition. Intensity of estrus characteristics on day of AI was mild. Major estrus signs observed were uterine tonicity, alertness, mucus discharge on rectal manipulation of internal genitalia, swelling of vulva and teat engorgement. Mucus discharge on rectal manipulation of internal genitalia and teat engorgement were clearly noticed in 50 % buffaloes. There was higher tendency (P=0.08) of estrus expression by buffalos with anestrous period of 10 months or lower compared to those with anestrous period longer than 10 months. Overall ovulation rate was 85.7%. Pregnancy rate to FTAI was 28.6% (22.2% and 40% in true anestrous and silent estrus buffaloes). 64.3% (66.7% and 60% of true anestrous and silent estrus) buffaloes were pregnant when checked at 102 days of FTAI including subsequent breeding. Although Ovsynch/FTAI seems to be alternative breeding technique in silent estrus buffaloes, further study with increased sample size is recommended. This protocol can potentially be used for resumption of ovarian cyclicity in true anestrus buffaloes.

Two experiments were designed to evaluate the effect of different treatments for the synchronization of ovulation on pregnancy rates following fixed-time AI (FTAI) of lactating dairy cows. In Experiment 1, 394 Holstein cows that were 61.7 � 13.6 days postpartum (range 35 to 94 days), with a milk yield of 30.7 � 6.8 liters per day (range 12.0 to 52.4 liters) and a body condition score (BCS) between 2.5 to 3.5 out of 5, were used. Cows were blocked by days postpartum and milk yield and randomly assigned to one of four treatment groups. Cows in P4+EB treatment groups received an intravaginal DIB device (1 g P4; Syntex, Argentina) and 2 mg of estradiol benzoate (EB; Syntex) i.m. on Day 0. On Day 8, DIB devices were removed, and cows received PGF (150 �g D+cloprostenol: Ciclase; Syntex), and were subdivided to receive 400 IU eCG (Novormon 5000, Syntex) i.m. or no further treatment at that time. On Day 9, all cows received 1 mg of EB and were FTAI 60 h after DIB removal. Cows in P4-Synch groups received a DIB device and 50 �g of GnRH (Lecirelina, Gonasyn; Syntex) i.m. on Day 0. On Day 7, DIB devices were removed; cows received PGF and were divided to receive 400 IU eCG i.m. or no further treatment. On Day 9, all cows received a second GnRH treatment and were FTAI 60 h after DIB removal. Blood samples were taken on Days -10 and 0 to determine plasma P4 concentrations; 93% of the cows had >1 ng/mL P4 in at least one sample. In Experiment 2, 200 lactating cows from the same farm were treated with the P4+EB+eCG and P4-Synch without eCG treatments with either a DIB or a CIDR-B (1.9 g P4; Pfizer Animal Health, Groton, CT, USA) in a 2 � 2 factorial design. Cows were examined by rectal palpation 50 days after FTAI to determine pregnancy status, and data were analyzed by chi-square Mantel-Haenszel test. There was an EB/GnRH by eCG interaction (P < 0.05) which was attributed to a higher pregnancy rate in the P4+EB+eCG group (44/98; 44.9%) than in the P4+EB without eCG group (30/100; 30.0%) and P4-Synch+eCG group (30/98; 30.6%); the P4-Synch without eCG group had an intermediate pregnancy rate that did not differ from the other treatment groups (37/98; 38.8%). In Experiment 2, no significant differences (P = 0.40) in pregnancy rates were detected between cows treated with DIB (51/100; 51.0%) or CIDR-B (42/100; 42.0%), and there was no difference (P = 0.18) between P4+EB+eCG (52/100; 52.0%) and P4-Synch without eCG (41/100; 41.0%) treated groups. Results suggest that the addition of eCG will improve pregnancy rates following FTAI in lactating dairy cows treated with EB but not in those treated with GnRH at the time of insertion and after removal of a P4 releasing device. Although treatment with P4+EB+eCG resulted in numerically higher pregnancy rates, results were not different from those obtained in the GnRH-based treatment, without the addition of eCG.

The aim of this study was to establish a comparison between heterospermic and monospermic semen on the pregnancy rate of fixed-time AI Bos indicus cows. The experiment was conducted in the state of Mato Grosso, Brazil, during December 2010 and February 2011. Multiparous (n = 173) and primiparous (n = 204) Nelore cows, 45 ± 15 days postpartum, were evaluated on body condition score (BCS; score 1 to 9), and those that presented BCS between 5 and 8 were selected. Two protocols of estrous cycle synchronization were performed (PeCG and Pshang). Synchronization of the estrous cycle was achieved with the aid of the following protocol: on Day 0, all animals were treated with a progesterone vaginal implant with 1.9 g of progesterone combined with an IM injection of 2 mg of estradiol benzoate. On Day 7, an IM injection of 12.5 mg of dinoprost was administered. On Day 9, implants were withdrawn and the animals received an IM injection of 0.5 mg of estradiol cypionate. On Day 9, part of the animals (n = 222) received an IM injection of 300 IU mg of eCG (PeCG) and the other part (n = 155) had their calves removed for 48 hours (Pshang). All animals were inseminated 46 to 52 h after the end of the protocol. Cows were allocated randomly in 4 groups according to the donor male sample in the semen straw and groups were designated as MA (monospermic straw from bull A; n = 110), MB (monospermic straw from bull B; n = 102), MC (monospermic straw from bull C; n = 108), and HT (heterospermic sample from bulls A, B, and C; n = 57). Commercially available semen straws from Aberdeen Angus donors were used (ABS Pecplan, Brazil). Pregnancy diagnosis was performed 35 days after AI using real-time ultrasonography. Results are summarized in Table 1. There was no statistical difference between MA, MB, MC, and HT in any group according to the chi-square test (P > 0.05). However, there was a remarkable difference in pregnancy rates among the 3 bulls tested considering both protocols, probably due to the variation in the ovulation moment. Three experiments were conducted to compare pregnancy rates in cows and heifers treated with intravaginal progesterone (P4)-releasing devices impregnated with either 0.5 or 1.0 g of P4 and inseminated at a fixed time (FTAI). In Experiment 1, 102 postpartum beef cows (Hereford and Angus crossbred with Tuli, 60 to 90 d postpartum) with a body condition score (BCS) of 2.5 to 3.0 (1 to 5 scale) were used. On Day 0, all cows received 2 mg of estradiol benzoate (EB; Syntex, Argentina) i.m. and were randomly assigned to receive an intravaginal progesterone-releasing device with 0.5 g of P4 (DIB 0.5 g; Syntex) or 1.0 g of P4 (DIB 1.0 g; Syntex). On Day 7, DIB devices were removed and all cows received 150 µg D(+)cloprostenol (Ciclase; Syntex) i.m. On Day 8, all cows received 1 mg EB and were FTAI 52 to 56 h after DIB removal. In Experiment 2, 93 Holstein cows that were 90 to 110 days in milk (DIM), producing on average 5000 kg of milk per lactation and with a BCS of 2.5 to 3.0 received the same treatments as those in Experiment 1. In Experiment 3, 100 Holstein heifers, 20 to 22 months old, with a BCS of 2.5 to 3.0, also received the same treatments as those in Experiment 1. Pregnancy rates in all experiments were determined by ultrasonography 35 days after FTAI and compared by chi-square test. No differences in pregnancy rates were detected between lactating beef cows, dairy cows, or dairy heifers treated with DIB devices impregnated with 0.5 or 1.0 g of P4 (Table 1). It was concluded that vaginal P4 releasing devices impregnated with 0.5 g or 1.0 g of P4 results in comparable pregnancy rates in FTAI protocols in beef and dairy cattle. Table 1. Two experiments were designed to compare the effects of eCG treatment and temporary weaning on ovulation and pregnancy rates in postpartum cows. In Experiment 1, 39 lactating multiparous crossbred Bos indicus cows, 60 to 80 d postpartum with a body condition score (BCS) between 2.0 to 2.5 out of 5 were randomly allocated to one of four treatment groups in a 2 � 2 factorial design. On Day 0, all cows received a progesterone (P4) intravaginal device (DIB; Syntex, Argentina) and 2 mg estradiol benzoate (EB) i.m. (Syntex). On Day 8, DIB devices were removed and all cows received 150 �g D(+)cloprostenol i.m. (Ciclase; Syntex) and were randomly divided to receive 400 IU eCG i.m. (Novormon 5000; Syntex) at the same time or no treatment. In addition, half of the cows in each group had their calves weaned temporarily (TW) for 56 h at the time of DIB removal; the other half remained with their calves. All cows received 1 mg EB i.m. on Day 9 and were examined every 8 h by ultrasonography from the time of DIB removal until ovulation. Data were analyzed by ANOVA and chi-square test. The diameter of the dominant follicle on Day 8 (eCG: 7.8 � 0.5 mm vs. no eCG: 8.2 � 0.4 mm, and TW: 7.6 � 0.4 mm vs. no TW 8.4 � 0.5 mm), the proportion of cows that ovulated (eCG: 12/20, 60% vs. no eCG: 9/19, 47%; and TW: 13/20, 65% vs. no TW: 8/19, 42%, and the interval to ovulation (eCG: 72.0 � 1.4 h vs. no eCG: 75.6 � 2.0 h, and TW: 73.8 � 1.6 h vs. no TW: 73.0 � 1.8 h) did not differ among groups (P > 0.05). Although there was no effect of eCG treatment or eCG by TW interaction (P > 0.3) on the size of the preovulatory follicle (eCG: 11.1 � 0.4 mm vs. no eCG: 10.1 � 0.6 mm), it was smaller in cows TW (9.9 � 04 mm), compared to those not TW (11.8 � 0.3 mm; P < 0.05). Nevertheless, the growth rate of the ovulatory follicle was greater (P < 0.02) in cows treated with eCG (1.1 � 0.1 mm/day) than in those not treated with eCG (0.6 � 0.1 mm/day). In Experiment 2 769 lactating multiparous crossbred Bos indicus cows received the same treatments as those in Experiment 1 and were fixed-time inseminated (FTAI) 52 to 56 h after DIB removal. Cows were examined by ultrasonography on Day 0 (22.5% with corpora lutea (CL), 30.0% with follicles >8 mm, and 47.5% with follicles <8 mm) and 42 d after FTAI to determine pregnancy. Data were analyzed by logistic regression. Pregnancy rates were not affected (P > 0.7) by TW (TW: 141/397, 37.2% vs. no TW: 141/390, 36.1%) or the TW by eCG interaction (P > 0.7). However, pregnancy rates were higher (P < 0.05) in cows treated with eCG (154/377, 40.8%) than in those not treated with eCG (128/392, 32.6%). The use of eCG but not TW improved pregnancy rates following FTAI in postpartum Bos indicus cows. Results also suggest that the eCG-related increase in pregnancy rates may be due to the final growth rate of the ovulatory follicle.

Equine AI with frozen semen has become more frequent in reproduction centers, but osmotic shock during thawing is still a major problem as it impairs gamete viability and fertility. To overcome this, multi ultrasonographic (US) exams must be performed to ensure AI at the closest time to ovulation possible. The objective was to evaluate, in vivo, the effects of dilution of the cryoprotectant dimethylphormamide on the success of a fixed timed (20 h post-ovulation) AI protocol, in an attempt to reduce osmotic shock and the intense labour usually required during equine AI with frozen semen. Thirty-five Mangalarga Marchador mares were used in this trial. When dominant follicles reached at least 35 mm in diameter, ovulation was induced with 6 mg of equine gonadotrophin extract (EGE) intravenously. Artificial insemination was performed 20 h after ovulation according to 1 of the following protocols: no dilution (n = 14): three 0.5-mL straws were thawed and the semen deposited by a flexible rod at the uterine tip in the ipsilateral horn where the pre-ovulatory follicle was detected; or dilution (n = 21): three 0.5-mL straws were thawed as described and centrifuged after addition of 1.5 mL of BotuSêmen® extender (1:1 dilution). Pregnancy rates were evaluated between treatments, age and presence of intrauterine liquid (IUL) classes and period between ovulation and AI. Data were analysed as a binomial distribution with the GENMOD procedure (SAS, Cary, NC, USA). Pregnancy rate was higher (P < 0.05) for mares inseminated with diluted semen (42.9%) compared to those inseminated with no semen dilution (4.8%). Age and IUL class did not influence pregnancy rates. It can be concluded that reduction of osmotic stress through the dilution of cryopreserved equine semen increased pregnancy rates after AI. Semen dilution, as tested in this trial, yielded good pregnancy rates in a fixed-time AI protocol in mares, simplified mare management greatly and allowed the use of a fixed-time protocol successfully. Abstract Clitoral stimulation after artificial insemination (AI) in beef cattle is a common practice utilized by AI technicians; however, the effect of clitoral stimulation during fixed-time AI (TAI) is still unknown. The objective of the present experiment was to evaluate the effect of clitoral stimulation on temperament responses and pregnancy rates to TAI in Bos indicus beef cows. A total of 1,186 multiparous Nellore cows across three different locations were assigned to an estradiol/progesterone-based estrus synchronization + TAI protocol (day −11 to 0). Cows were randomly assigned to receive either 3 s of clitoral stimulation (n = 602) or no clitoral stimulation (n = 584) immediately after TAI (day 0). Cow body condition score (BCS) was recorded on day −11. Estrus expression was evaluated based on estrus detection visual aid patch activation on day 0 (estrus, ≥50% activated; no estrus, &lt;50% activated). Temperament was assessed by individual chute score based on a 5-point scale before TAI, and individual exit velocity was measured after clitoral stimulation. Pregnancy diagnosis was performed 30 d after TAI via transrectal ultrasonography. Pregnancy rate to TAI was positively affected by BCS (P &lt; 0.01) and estrus expression (P = 0.03). Pregnancy rates of cows receiving clitoral stimulation did not differ (P = 0.39) from cows non-stimulated (47.5 ± 4.6% vs. 44.3 ± 4.6%, respectively). No interaction of clitoral stimulation and estrus expression was observed (P = 0.26). Chute score was positively correlated to exit velocity (P &lt; 0.01; r = 0.29); however, clitoral stimulation did not affect exit velocity (P = 0.86). In summary, pregnancy rates to TAI are influenced by various factors and multiple strategies have the potential to increase the fertility of beef cows submitted to TAI; however, clitoral stimulation of Bos indicus beef cows did not improve TAI pregnancy rate.

ABSTRACT: This study evaluated the reproductive performance of Nellore heifers without previous vaccination undergoing fixed-time artificial insemination (FTAI) and subsequent natural breeding under different vaccination protocols against infectious bovine herpesvirus 1 (BHV-1), bovine viral diarrhea virus (BVDV), and bovine leptospirosis (BL). A total of 763 animals were included; the group assignment was as follows: T1, control without vaccination; T2, vaccination against BL; T3, vaccination against BHV-1and BVDV; T4,vaccination against combination of BL + BHV-1and BVDV. Vaccination was started 30 days before FTAI (D-40), and a booster dose was administered at D-10. Serological tests were used to verify the occurrence rate of natural infection by BL, BHV-1, and BVDV. Initial pregnancy was diagnosed at D30, D60, and D120 after FTAI. Final pregnancy rates were evaluated by group, weight, body-condition score, occurrence of estrus, inseminator, bull, and occurrence of abortions or embryonic death. In all groups, results indicated seropositivity of 100% for leptospirosis, 77.0-94.0% for BHV-1, and 64.0-87.0% for BVDV. The vaccination protocol was not a significant influencing factor of pregnancy rate and embryonic or fetal mortality; whereas, animal weight at the beginning of the breeding season and presence of estrus at FTAI were significant influencing factors of pregnancy rate. The vaccination protocol had no interference with the reproductive performance of the experimental groups.

Studies have shown that gonadotropin-releasing hormone-based protocols that reduce the period of progestin insertion and prolong the period from progestin removal to gonadotropin-releasing hormone and fixed-time AI (FTAI; named 5-day Co-Synch) results in similar or higher pregnancy rates than the conventional 7-day Co-Synch protocol in beef cows and heifers (Bridges et al. 2008 Theriogenology 69, 843–851). Similar findings have been reported following the use of an oestradiol-based protocol that also provides for a longer period of proestrus (named J-Synch; de la Mata and Bó 2012 Taurus 55, 17–23). An experiment was designed to compare the J-Synch protocol for synchronization of ovulation that allows for a prolonged proestrus with a conventional 7-day oestradiol-based protocol for FTAI in heifers. Cycling 18-month old Angus and Hereford heifers (n = 208) with a body condition score of 6 to 7 (scale of 1 to 9) were randomly allocated to 1 of 2 treatment groups. Heifers in the 7-day EB group (n = 105) received a progesterone (P4) device (DIB 0.5 g of P4, Syntex SA, Buenos Aires, Argentina) and 2 mg of oestradiol benzoate (EB, Syntex SA) on Day 0 and 500 μg of cloprostenol (PGF; Ciclase DL, Syntex SA) and 0.5 mg oestradiol cypionate (Cipiosyn, Syntex SA) on the day of DIB removal (Day 7). Heifers were also tail painted at the time of DIB removal and observed for signs of oestrus (i.e. tail paint rubbed off). Those with the tail paint rubbed off by 36 h after DIB removal were inseminated 12 h later, whereas those not showing oestrus by 36 h were FTAI at 54 h. Heifers in the J-Synch group (n = 103) received DIB and 2 mg of EB on Day 0 and PGF on the day of DIB removal (Day 6). Heifers in this group were also tail painted at DIB removal, and those with their tail paint rubbed off by 48 h were inseminated 12 h later; those not showing oestrus by 60 h received 100 μg of gonadorelin acetate (gonadotropin-releasing hormone, Gonasyn gdr, Syntex SA) and were FTAI at 72 h after DIB removal. Pregnancy was diagnosed by ultrasonography at 55 days after FTAI (Honda 101V, 5.0-MHz transducer). Data were analysed by logistic regression. Oestrus detection rate and pregnancy rate to FTAI did not differ (P > 0.1) between groups (38.8%, 40/103 and 60.3%, 38/ 63 for heifers in the J-Synch group v. 28.5%, 30/105 and 45.3%, 34/75 for those in the 7-day EB group). However, pregnancy rates to observed oestrus tended (P < 0.09) to be higher and the overall pregnancy rate was significantly higher (P < 0.01) in heifers in the J-Synch group (80.0%, 32/40 and 67.9%, 70 /103) compared with those in 7-day EB group (50%, 15/30 and 46.6%, 49/105). Furthermore, heifers within the J-Synch group that had their tail paint rubbed off by 48 h after DIB removal and were AI 12 h later (i.e. 60 h) had higher (P < 0.05) pregnancy rate than those in the same group that were FTAI. In conclusion, reducing the time of progestin device insertion and lengthening the proestrus period, as in the J-Synch protocol, results in higher pregnancy rates than with the conventional oestradiol-based protocol. Furthermore, the combination of oestrus detection and FTAI would appear to improve the pregnancy outcome even more.
