Background: Molecular markers based approaches are essential to select fertile bulls for frozen semen production at an early age. The present investigation was undertaken to perform the molecular characterization and identify single nucleotide polymorphisms (SNP) in Y-chromosome specific DDX3Y gene in Murrah buffalo bulls. Methods: The genomic DNA isolated from the blood samples of 70 Murrah buffalo bulls, covering bulls with normal seminal traits and poor production performance (poor semen quality, freezability, libido), were subjected to PCR amplification. The sequences of DDX3Y gene were analyzed for single nucleotide polymorphism using the seqman module of DNASTAR LASERGENE software. The single nucleotide variations in the sequences with reference to the Bos taurus sequence were determined using Clustal W. The phylogenetic tree and genetic distance were constructed using the MegAlign module. Result: The analysis of sequences revealed that the exons and their adjacent intronic regions of the DDX3Y gene are monomorphic in nature without any variations indicating that the sequences are highly conserved in the studied population of Murrah buffalo bulls. However, a considerable number of single nucleotide variations were observed in the sequences of Murrah buffalo compared with Bos taurus sequences. Furthermore, the phylogenetic tree analysis revealed less divergence and close genetic association between the sequences of Murrah buffalo and other species in the bovinae family than the caprinea species. Further studies on DDX3Y gene in a more extensive and diverse population of Murrah buffalo bulls distributed in different regions could aid to discover substantial SNPs.

The success of artificial insemination must be supported by a balance of nutrients for livestock. AI combined with sexing produces offspring according to wishes of the breeder. This study aims to determine the increase in concentration on the success of Artificial Insemination using frozen semen sexing. This research was conducted at the people's beef cattle farm, Sumber Pucung District, Malang Regency. The limousine crosser used was 30 cows. Cattle aged 1.8 – 5 years with BCS 3-5 (score 1-9) Treatment T0 : The usual feed given by farmers (Control). T1: Control+HQFS. Parameters NRR-1, NRR-2, Conception Rate (CR), Pregnancy Rate (PR). The additional feeding of concentrate did not significantly affect the reproductive parameters. The results showed that at T0 the results of NRR-1 and NRR-2 were 80% and 73.33%, respectively. while in T1 it was 73.3% and 66.67%. The results showed that the CR in both treatments was 26% and the results showed that the PR for T0 was 26% and for T1 was 33%. Insufficient feed consumption for all treatments based on NRC requirement data, T0 and T1 dry matter consumption respectively 6.6 and 7.3 Kg/head/day, Protein consumption 0.36 and 0.3 Kg/head/day and TDN consumption of 3.08 and 3.20. kg/head/day.

Ongole cattle in smallholder farms on Java, Indonesia, have been crossed with Limousin, which is not controlled repeatedly. The character of estrus affects the success of Artificial Insemination. This study aimed to determine the relationship between estrus characters before double dose artificial insemination using frozen semen sexing. The research was conducted in Senggreng Village, Sumber Pucung District, Malang Regency. The material was 45 head of Limousin crossbred cow inseminated using sexing semen produced by the Singosari National Artificial Insemination Center (SNAIC). The sexed semen processed with the Percoll Density Gradient Centrifugation (PDGC) method. The average body value of cows between 3-5 (scale 1-9) with an average age of 1.8-7 years, normal reproduction, clear signs of estrus. The reproductive organs were examined by rectal palpation to confirm that the cows are not pregnant. Parameters of estrus character were vulvar color, cervical mucus, vaginal temperature, pH, vulvar swelling, and heat detector value. The result showed that the estrus characters, including vulva color, vulva swelling, cervical mucus, vaginal temperature, cervical mucus pH, and heat detector score in the first and second artificial insemination have a low relationship. The regression models produced can be used but are less precise to estimate the relationship between estrus characters because the coefficient of determination shows a low influence.

Background and Aim: The Javan leopard (Panthera pardus melas Cuvier, 1809) is a subspecies of Panthera pardus spp., spread across the African and Asian regions. Information on reproductive aspects is crucial for wild animals, including the Javan leopard. In this study, we aimed to develop electroejaculator (EE) techniques and evaluate cryopreservation success in Javan leopard semen. Materials and Methods: The semen of four adult Javan leopards was collected once a week using EE. Placement of the EE probe in the rectum was performed after ultrasound imaging (ultrasonography) to determine the prostate body location. The semen obtained was then evaluated macroscopically and microscopically. Three Javan leopards were used for cryopreservation. The ejaculate was divided into two parts [i.e., one part diluted with AndroMed® (Minitüb, Tiefenbach, Germany) and the other part with Steridyl® (Minitüb, Tiefenbach, Germany)] at a 1:1 ratio immediately after collection and evaluation. The semen was then packed in a 0.25 mL MiniStraw® (Minitüb, Tiefenbach, Germany) then equilibrated at 4°C for 2 h. After equilibration, the straw was then frozen in liquid nitrogen vapor. Frozen semen was then stored in containers until further evaluation. Results: The results showed that ejaculation response occurred at all levels of stimulation, while erections did not always occur. The fastest ejaculation and erection occurred at the fourth voltage. The macroscopic evaluation showed that the semen volume was 0.80±0.26 mL, cloudy white, pH 7.44±0.14, and with watery semen consistency. The microscopic evaluation showed that the sperm motility was 66.98±0.39%, with sperm viability of 75.6±1.79%. Sperm concentration was 62.17±46.95×106 mL–1 with a total concentration of 42.14±23.51×106 cells. Normal sperm morphology is only 40.72±6.26%. Conclusion: This study concluded that the development of a semen collection technique using an EE preceded by imaging of the EE probe location using ultrasound was effective for the ejaculation of Javan leopards. The characteristics of the semen of the Javan leopard showed moderate semen volume, sperm motility, and viability. Javan leopard showed low sperm concentration and normal sperm morphology.

A chromosomal polymorphism was detected on karyological screening of Kangayam breeding sires prior to subjecting them for frozen semen collection. One bull possessed the chromosomal complement 2n = 60, consisting of 58 acrocentric autosomes, one large sub-metacentric X-chromosome, and one small acrocentric Y-chromosome with a small visible p-arm, which was further confirmed using CBG- and GTG-banding. This polymorphism was attributed to a heterochromatin variation of the acrocentric Y-chromosome routine in the Bos indicus Linnaeus, 1758 cattle.

Abstract This article addressed the development and the growth of offspring, the external characteristics and the productivity at different breeding periods of woolly-meaty sheep of Uzbekistan. It was observed that wool productivity was higher in sheep rich in wool-meat than ordinary sheep; sheared wool was 1.26 kg (49.6%), after washing it was 0.97 kg (67.8%), wool length was 2.13 (19.2%) higher. Live weights of lambs from ewes in the experimental groups were determined at birth, at 30 days of age, and 3 months of age. From September 1 to September 20 of the year, the live weight, body size and wool productivity of early inseminated ewes were higher than those of inseminated ewes from late October to November 10. The live weight of lambs obtained from artificially inseminated sheep with frozen semen of typical rams in world gene pool considered semi-fine wool in the meat-wool direction was higher than sheep naturally inseminated with pedigree rams. Accordingly, it was heavier by 00.4 kg (9.3%) at the first day of birth, followed by 0.5 kg (6.8%) at 10 days, and 0.4 kg (1.3%) at 3 months of age.

This research was conducted at the Teaching Farm of the Faculty of Veterinary Medicine, Airlangga University, to know the effect of age, temperature, and humidity on the quality of semen of bulls which are turned into frozen semen. This research uses fresh semen samples of bulls of various ages grouped into 3 groups, young, middle age, and old groups and measures temperature and humidity during the storage process. Fresh semen is collected using an artificial vagina and then examined macroscopic and microscopic tests which is then checked for post thawing motility. Data analysis uses Analysis of Variants (ANOVA) followed by Duncan. The results showed that the percentage of spermatozoa motility of fresh semen and post thawing differed and age affected the quality of semen produced. There was no effect of temperature and humidity when processing of collecting semen. Bull’s semen at Teaching Farm Faculty of Veterinary Medicine Airlangga University has good quality and is suitable for Artificial Insemination.

Same-sex female couples who wish to become pregnant can choose donor insemination or in-vitro fertilization (IVF)—a technique intended for infertile women. In general, women in same-sex female partnerships are no more likely to be infertile than those in opposite sex partnerships. This article investigates data available from the Government Regulator of UK fertility clinics—the Human Fertilization and Embryology Authority, which is the only data available worldwide on same-sex female couples and their fertility choices. IVF is increasing both in absolute numbers and relative proportions year on year in the UK, compared to licensed donor insemination for same-sex female couples. As IVF has greater human and financial costs than donor insemination, policies should not encourage it as the first choice for fertile women requiring sperm. Commercial transactions are taking place where fertile lesbians receive cut price, and arguably unnecessary, IVF intervention in exchange for selling their eggs to be used for other infertile customers. If women are not told about the efficacy of fresh vs. frozen semen, and the risks of egg ‘sharing’ or intra-couple donation, exploitation becomes possible.
