Cytogenetic status of blood lymphocytes in schoolchildren living middle and high latitudes

Intensive industrial development of the northern regions of the Russian Federation is primarily associated with the extraction and processing of natural resources, which leads to environmental pollution and makes the tasks of sanitary and hygienic monitoring urgent. Rapid methods are convenient for the rapid diagnosis of the genetic well-being of the environment, one of which is the micronucleus test in combination with the cytokinetic block cytochalasin B. mitosis. At the moment, there are scanty data on the inter-latitudinal comparison of the results of cytogenetic studies on cells of various human tissues, which greatly complicates the comparison of the results obtained in the Arctic zone of the Russian Federation with data from more southern regions. The purpose of this study is to identify and compare division abnormalities in cells of peripheral blood lymphocytes in two groups of older schoolchildren living in middle and high latitudes. The study was carried out in the city of Serpukhov, Moscow region (54 ° 54 ‘56 “N, 37 ° 24’ 40” E) and the city of Apatity, Murmansk region (67 ° 34’03 “N, 33 ° 23’36 ″ E). A total of 40 children aged 16-18 were examined - 20 children each from the city of Serpukhov and from the city of Apatity. The micronucleus test was carried out in accordance with the international protocol. Cytogenetic analysis of schoolchildren’s peripheral blood lymphocytes using the micronucleus test showed that the spontaneous number of cells with micronuclei is comparable in middle and high latitudes, and approaches the upper limit of the mean population norm. Therefore, when comparing the data of the micronucleus test on the cells of peripheral blood lymphocytes of the studied groups, it is permissible not to take into account the latitude of residence.

Frequency of micronuclei (MN) in peripheral blood lymphocytes is a measure of genotoxicity and spontaneous chromosomal instability with many modifying factors. The aim of this work was to examine the relation between results of micronucleus test and set of socio-demographic parameters in healthy population in south-east Serbia using structure equation approach. Cytokinesis–block (CB) technique was used for analysis of MN frequency in peripheral blood lymphocytes on 133 healthy volunteers of various ages. Socio-demographic data was collected through a questionnaire. The mean frequency of MN gradually rose with age from 0.56±0.71 in new-borns to 5.48 ±3.65 in the 61-80 years age group (AG), with a decrease in the 81-92 years AG. MN frequency was positively correlated with age, altitude of birth place, altitude of place of residence, nuclear buds and nucleoplasmatic bridges, and negatively with education level and smoking habits. Linear structural model revealed age to be related to all of the examined variables, and indicates probable existence of another factor, independent of age, influencing all of these except nuclear buds. It can be concluded that the frequency of micronuclei is influenced by age and factor/s resembling socioeconomic status or lifestyle and this influence is independent of age.

Engineered nanoparticles (ENPs) are used in a wide range of applications because of their unique properties. Cerium dioxide nanoparticles (CeO2 NPs) are one of the important ENPs, and they can cause negative health effects, such as genotoxicity, in humans and other living organisms. The aim of this work was to analyze the genotoxic effects of short-term (3–24 h) CeO2 NPs exposure to cultured human blood lymphocytes. Three genotoxicity systems “cytokinesis-block micronucleus test, comet assay, and gamma H2AX test” were used to show the genotoxic potential of CeO2 NPs (particle size <25 nm, concentrations: 6, 12, and 18 µg/mL). Hydrogen peroxide was selected as the positive-control genotoxic agent. Our results indicate that CeO2 NPs have genotoxic potential on human peripheral blood lymphocytes cells even at 3–24 h exposure under in vitro conditions.

The article focuses on the analysis of the methodology and content of the Strategy for Spatial Development of the Russian Federation until 2025. Particular attention is paid to the task set in the Strategy to improve the population settlement system and to reflection of regional issues in it. The orientation of the Strategy on large cities is in conflict with the objectives of the national project «Demography». Pulling the population into large cities makes it difficult to achieve the target indicator «Increase in the total fertility rate to 1.7 per woman by 2024». For the Strategy of spatial development, whose task was proclaimed to improve the system of population settlement, the problem of its reproduction and placement is far from an idle question. Nevertheless, in the Strategy this issue does not go beyond the framework of duty generalizations, which proclaim a differentiated approach to the directions and measures of the State support for the socio-economic development of territories, taking into account the demographic situation, characteristics of the settlement system, level and dynamics of the economic development and specifics of the environmental conditions. And the thesis formulated in the Strategy: «Stabilization of the population in most constituent entities of the Russian Federation», in fact, has no foundation. In 2005-2018 two thirds of the regions (67.1%) were losing their population. Analysis of the place and role in the Strategy of the region taken as an example (Komi Republic) shows that, in comparison with most other subjects of the Federation, quite a lot of attention and place is given to this republic. It is described as a part of the Northern macroregion, the economic specialization of the republic is indicated there, its capital — the city of Syktyvkar is represented among the promising economic centers, a number of municipal entities of the republic are identified as mineral resource centers, and the city of Vorkuta is presented as a part of the priority geostrategic territories of the Arctic zone. And this is quite understandable. The spatial development of the country is primarily oriented to the North and East.
