Variable preservation of tephra from the 1991 eruption of Volcán Hudson in small lakes: implications for reconstructing past eruption parameters

<p>Volcanic ash (tephra) deposits are used to reconstruct past eruption parameters. The ways in which tephra deposits are modified between deposition and their long-term preservation in the stratigraphic archive are poorly understood. In particular, we don’t know if tephra layers preserved in lake sediments from small lakes accurately reflect the initial tephra fallout. We address this by re-surveying tephra deposits from the 1991 eruption of Volcán Hudson, Chile. We measured tephra thickness, mass-loading and grain-size distribution of tephra from multiple cores in six small (<0.2 km<sup>2</sup>) lakes at locations 76-110 km from the volcano and in areas of contrasting land cover and climate. We also measured tephra preservation in terrestrial sites within each lake catchment. These data were compared with measurements taken shortly (days to weeks) after the eruption to determine how the tephra deposits have changed in the 29 years since the eruption. Preservation is variable within and between lakes, and also varies with the vegetation cover at terrestrial sites adjacent to the lakes. Tephra thicknesses are broadly comparable to the original fallout, but the degree of similarity varied notably and is sensitive to preservation environment. These findings have implications for reconstructing eruption parameters from tephra deposits in small lakes, and where the fallout area crosses large environmental gradients and contrasting vegetation regimes.</p>

Patients with primary open-angle glaucoma (POAG) and ophthalmic hypertension have an increased likelihood of developing occlusions (thrombosis) of the central retinal vein. Different groups of antihypertensive drugs differ in their mechanism of action and may affect concomitant ocular pathology, in particular, retinal edema, which occurs, for example, in occlusion of the central retinal vein. Used in most patients with glaucoma, prostaglandin analogs can contribute to the long-term preservation of macular edema due to the effect on the permeability of the vascular wall. Preparations of other pharmacological groups, reducing the production of aqueous humor, on the contrary, may contribute to its regression. Therefore, the question of choosing a drug for antihypertensive therapy in patients with primary open-angle glaucoma and concomitant macular edema is relevant and is for further study.

This chapter outlines the advantages of digital epigraphy in the context of the original monuments. It analyzes the perception of epigraphic publication of monuments, taking into account new technologies. 3DVR models can be created using architectural drawings and measurements (CAD and 3D modeling), 3D scanning, and Structure for Motion (SfM). These systems present different advantages and challenges, which are discussed. Current options for publication include VSim, 3D GIS, and Unity 3D platforms. The issues of peer review of publications and long-term preservation of data are addressed. The chapter concludes with a consideration of the issue of potentially misleading impressions given by 3DVR representations.

An optimized cryopreservation protocol for embryonic axes (EAs) of chestnut (Castanea sativa Mill.) has been developed based on the encapsulation–vitrification procedure. EAs of mature seeds were aseptically dissected and encapsulated in alginate beads with or without 0.3% (w/v) activated charcoal (AC). Embedded EAs were dehydrated with Plant Vitrification Solution 2 for different treatment times up to 120 min, followed by direct immersion in liquid nitrogen. Cryopreserved embryonic axes encapsulated with AC showed higher survival (70%) compared to those encapsulated without AC (50%). Sixty-four percent of embryonic axes, from synthetic seeds with AC, subsequently developed as whole plants. Plantlet regrowth was faster in AC-encapsulated EAs and showed enhanced postcryopreservation shoot and root regrowth over 2 cm after five weeks from rewarming. Results indicate that encapsulation–vitrification with activated charcoal added to the beads is an effective method for the long-term preservation of Castaneasativa embryonic axes.

(1) Background: giant vesicles (GVs) are widely employed as models for studying physicochemical properties of bio-membranes and artificial cell construction due to their similarities to natural cell membranes. Considering the critical roles of GVs, various methods have been developed to prepare them. Notably, the water-in-oil (w/o) inverted emulsion-transfer method is reported to be the most promising, owning to the relatively higher productivity and better encapsulation efficiency of biomolecules. Previously, we successfully established an improved approach to acquire detailed information of 1-Palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC)-derived GVs with imaging flow cytometry (IFC); (2) Methods: we prepared GVs with different lipid compositions, including phosphatidylcholines (PCs), phosphatidylethanolamines (PEs), and PC/PE mixtures by w/o inverted emulsion methods. We comprehensively compared the yield, purity, size, and encapsulation efficiency of the resulting vesicles; (3) Results: the relatively higher productivities of GVs could be obtained from POPC, 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), 1,2-dilauroyl-sn-glycero-3-phosphoethanolamine (DLPE), DOPC: DLPE (7:3), and POPC: DLPE (6:4) pools. Furthermore, we also demonstrate that these GVs are stable during long term preservation in 4 °C. (4) Conclusions: our results will be useful for the analytical study of GVs and GV-based applications.
