Anredera cordifolia [Ten] Steenis has been proven to have pharmacological activity that is thought to be influenced by the presence of phenolic compounds and flavonoids. However, both compounds are found in small concentrations through conventional extraction (maceration, decoction, soxhlet, and reflux). This study aims to prove that the ultrasonic assisted extraction method can obtain higher concentrations of phenolic and flavonoid compounds. Dried binahong leaves were extracted using ultrasonic assisted extraction with 70% ethanol. Phenolic levels were determined using Folin-Ciocalteu, while flavonoid levels were determined using the colorimetric aluminum method. The results showed binahong leaves ethanolic extract had phenolic levels of 10.16 % GAE and flavonoid levels of 10.58 % QE. These results are much higher than the results obtained using conventional extraction so that ultrasonic assisted extraction can be recommended to maximize the extraction of phenolic and flavonoid levels from the leaves of Binahong (Anredera cordifolia [Ten] Steenis).

Recently, it has been demonstrated that a rare flavonoid, 5,7-dihydroxy-6,8,4′-trimethoxyflavonol (1), isolated from the leaves of Rubus rosifolius showed significant antiproliferative activity against different cancer cell lines and had a diuretic effect. This study aimed to determine the best extraction method evaluating dynamic maceration extraction (DME), ultrasonic assisted extraction (UAE), and microwave assisted extraction (MAE). In addition, level optimization for different factors was focused using Design of Experiments. The flavonoid content was estimated by high performance liquid chromatography (HPLC). The results indicated that UAE provided the highest content with the shortest extraction time. For optimization, response surface methodology (RSM) was used, applying a Box-Behnken Design (BBD) with three factors at three levels. It was possible to observe that the flavonoid content significantly increased with the increase of temperature (max. 70°C). The time had little influence on the extraction and, for this reason, was set at 25 min. With regard to the drug/solvent ratio, it was observed that this had an impact on yield with a ratio of 3.5%. A model was built and a determination coefficient of 0.98 was obtained. After validation, an optimized method was developed, able to extract 1 with a lower time, lower solvent consumption, and affording higher contents.

<p class="1Body">The water-soluble polysaccharides content of soybean curd residue fermented by <em>Flammulina velutipes</em> was about 7 times higher than unfermented one and more than 4 times higher than <em>Flammulina velutipes</em> fruiting body. The optimized ultrasonic-assisted extraction method (80 °C, 20 min, 80 W and solid-liquid ratio of 1 g/30 mL) showed a water-soluble polysaccharides extraction yield of 7.88% which was higher than that of hot-water extraction method (6.11%). Moreover, the crude polysaccharides were purified and the main components showed strong activities on DPPH radical scavenging (IC50 0.21 mg/mL), ABTS radical scavenging (IC50 0.42 mg/mL), hydroxyl radical scavenging (IC50 0.28 mg/mL) and SOD-like activity (IC50 0.88 mg/mL). The results indicated that the fermentation method could be used as a new approach for bio-active polysaccharides production and a new method for reusing the food industrial waste. The ultrasonic-assisted extraction method could be considered as a more efficient method in polysaccharides extraction process. Furthermore, the fermented soybean curd residue could be expected as a new source of antioxidant with the further research value.</p>
