Genetic Divergence among Populations of Invasive Alien Species Lantana camara L. from Selected Areas of Bangladesh

Lantana camara L., a well-known invasive alien species causing invasion and posing threat to native plant species community in different regions of Bangladesh. The present study aimed to investigate the genetic diversity of L. camara populations in different regions of Bangladesh. Eight RAPD markers were used in order to probe into its genetic variability. Total number of bands (202), polymorphic loci (104), per-centage of polymorphism (97.20%), average Shanon’s information index (0.3051±0.115), Nei’s gene diversity (0.4733±0.144) was found and in different populations and multiple divergent genetic clustering along with presence of unique alleles (4) for RAPD revealed high genetic diversity among the populations of L. camara in different regions of Bangladesh.

A large scale of EST sequences of Polyporales was screened in this investigation in order to identify EST-SSR markers for various applications. The distribution of EST sequences and SSRs in five families of Polyporales was analyzed, respectively. Mononucleotide was the most abundant type, followed by trinucleotide. Among five families, Ganodermataceae occupied the most SSR markers, followed by Coriolaceae. Functional prediction of SSR marker-containing EST sequences inGanoderma lucidumobtained three main groups, namely, cellular component, biological process, and molecular function. Thirty EST-SSR primers were designed to evaluate the genetic diversity of 13 naturalPolyporus umbellatusaccessions. Twenty one EST-SSRs were polymorphic with average PIC value of 0.33 and transferability rate of 71%. These 13P.umbellatusaccessions showed relatively high genetic diversity. The expected heterozygosity, Nei’s gene diversity, and Shannon information index were 0.41, 0.39, and 0.57, respectively. Both UPGMA dendrogram and principal coordinate analysis (PCA) showed the same cluster result that divided the 13 accessions into three or four groups.

AFLP, RAPD and SSR molecular markers were used to study the genetic diversity and genetic structure of the Híbrido de Timor germplasm. The principal coordinate analysis, UPGMA cluster analysis based on genetic dissimilarity of Jaccard, Bayesian model-based cluster analysis, percentage of polymorphic loci, Shannon's information index and Nei gene diversity were employed to assess the genetic diversity. The analyses demonstrated a high genetic diversity among Híbrido de Timor accessions. UPGMA and Bayesian cluster analyses grouped the accessions into three clusters. The genetic structure of Híbrido de Timor is reported. The management of Híbrido de Timor germplasm variability and its potential use in breeding programs are discussed.

Stephanie Kwangsiensis H. S. Lo and Salvia prionitis Hance were two rare and valuable Chinese herbs. Genetic diversity of the two plants were studied by using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) markers after they were transplantation and tissue culture. Results indicated that Nei’s gene diversity (H), Shannon’s information index (I), the percentage of polymorphic loci (PPL) of source / transplantation / tissue culture materials of S. Kwangsiensis H. S. Lo were H=0.282 / 0.195/ 0.056, I=0.415 / 0.290 / 0.087, PPL=75.6% / 56.1% /19.5% based on RAPDs and H=0.295 / 0.292 / 0.111, I=0.445 / 0.435 / 0.166, PPL=85.3% / 85.3% / 32.8% based ISSRs. And the same parameters of source / transplantation / tissue culture materials of S. prionitis Hance were H=0.193 / 0.192 / 0.231, I=0.291 / 0.289 /0.347, PPL=57.8% / 57.2% / 68.6% based on RAPDs and H=0.217 / 0.217 / 0.155, I=0.327 / 0.326 /0.235, and PPL=64.2% / 64.2% / 49.1% based on ISSRs instead. For the transplanted materials, genetic diversity was slightly declining with the disappearance of adversity. And for the tissue culture materials, more genets sampled for tissue culture, less reduction of genetic diversity; and fewer genets sampled, more reduction of genetic diversity. In addition, high genetic diversity appearing in tissue culture materials of S. prionitis Hance partially generate from somatic mutations. And these mutations maybe related to RAPD-PCR regions and just had been checked out by RAPDs.

Calophyllum brasiliense populations are under severe depletion and criteria to improve production and quality of propagative material are therefore necessary. Genetic measures have the potential to reduce consanguinity and maximize allelic representation within target populations. Here, we explored genetic values for this species in a small relic of natural forest in Rio de Janeiro State (Brazil). The objective was to evaluate the potential of some genetic measures for seed orchard establishment. As genomic information of native trees is still scarce, we opted to use a dominant marker: RAPD. DNA from 17 phenotypically superior trees was obtained through the CTAB method and submitted for amplification by PCR. Electrophoresis and electronic documentation was then conducted. We calculated the percentage of polymorphic bands (PPB), gene diversity (Ht), Shannon’s information index (i), genetic distance (UPGMA) and parsimony analysis. Six primers were evaluated generating 34 loci. We found high genetic diversity PPB=70.6% with Ht=0.28 and i=0.41. Genetic relationships were reported in dendrograms (maximum parsimony and distance). Simulated sampling within and among clusters suggests that inter cluster sampling is more effective to capture the genetic diversity.

Calamus brandisii Becc. is one of the endemic slender rattans found in the Western Ghats of India. The genetic diversity of two main populations available in Kerala was investigated using 20 RAPD and 9 ISSR markers. Two parameters viz., gene diversity and genetic diversity within and among populations were analyzed. ISSR analysis showed quite high genetic diversity in Pandimotta compared to Bonacaud population whereas in RAPD markers both these populations were moderately diverse. The percentage of total genetic differentiation (Gst) among two populations is relatively higher than the mean Gst value indicating high genetic diversity within the populations. The genetic distance between these two populations was 0.1739 with ISSR markers and 0.1971 with RAPD markers. Because of its high genetic diversity, Pandimotta population can be treated as an important population of gene diversity with potentially useful genes. This may be included in the high priority reservoir for genetic conservation also.

Abstract Morinda (Rubiaceae) is considerably recognized for its multiple uses viz. food, medicine, dyes, firewood, tools, oil, bio-sorbent etc. The molecular characterization of such an important plant would be very useful for its multifarious enhanced utilization. In the present study, 31 Morinda genotypes belonging to two different species Morinda citrifolia and Morinda tomentosa collected from different regions of India were investigated using Inter Simple Sequence Repeat (ISSR) markers. Fifteen ISSR primers generated 176 bands with an average of 11.7 bands per primer, of which (90.34%) were polymorphic. The percentage of polymorphic bands, mean Nei’s gene diversity, mean Shannon’s information index in Morinda tomentosa and Morinda citrifolia was [(69.89%, 30.68%); (0.21 ± 0.19, 0.12 ± 0.20); (0.32 ± 0.27 0.17 ± 0.28)] respectively, revealing higher polymorphism and genetic diversity in Morinda tomentosa compared to Morinda citrifolia. Structure, and UPGMA cluster analysis placed the genotypes into well-defined separate clusters belonging to two species Morinda tomentosa and Morinda citrifolia revealing the utility of ISSR markers in species differentiation. Distinct ecotypes within a particular species could also be inferred emphasizing the collection and conservation of Morinda genotypes from different regions, in order to capture the overall diversity of respective species. Further higher diversity of M. tomentosa must be advanced for its utilization in nutraceutical, nutritional and other nonfood purposes.

Abstract Metal accumulation in soil and plant tissues has caused severe ecological damage in forest ecosystems in the Sudbury region. The main objective of the present study was to determine the levels of genetic diversity in jack and red pine populations growing in metal contaminated and uncontaminated areas. Newly introduced populations were compared to 40 to 60 old populations. For jack pine, the percentage of polymorphic loci (P %) ranged from 14.6% to 45.8% with a mean of 31.6%. Nei’s gene diversity (h) varied from 0.046 to 0.169 with an average of 0.100, and Shannon’s index (I) ranged from 0.070 to 0.250 with an average of 0.153. The level of genetic variation was much lower in the red pine populations. For this species, the level of polymorphic loci varied from 4.55% to 27.27%. The mean for Nei’s gene diversity and Shannon’s information index, were 0.034 and 0.053, respectively. The highest genetic diversity values were observed in new plantations being developed by the Sudbury reforestation program. Overall, the genetic distance among the Pinus banksiana populations revealed that all the populations analyzed were genetically close to each other. There was no association between metal accumulation and genetic diversity for both species.

Genetic information is essential for conservation and future aquaculture development of the endangered catfish Rita rita (Hamilton, 1822). Two hundred catfish, R. rita, 50 from four rivers, the Old Brahmaputra, Jamuna, Meghna and Kangsa were collected and analysed to evaluate the genetic diversity and population structure using five microsatellite primers (Cba06-KUL, Cba08-KUL, Cba09-KUL, Phy03-KUL and Phy07-KUL). Four of the five amplified loci were found polymorphic (P95) in all the populations and 46 alleles were recorded with 9 to 14 alleles per locus. Differences were observed in the total number of alleles ranging from 41 to 44, effective number of alleles from 29.96 to 37.46, observed heterozygosity from 0.57 to 0.76, Shannon’s information index from 2.09 to 2.30 and polymorphic information content from 0.84 to 0.88 among the four populations. Results exposed the highest levels of genetic diversity in the Meghna population while the lowest in the Kangsa population of R. rita. All the populations were significantly deviated (P < 0.001) from the Hardy-Weinberg equilibrium for all the loci. Nei’s genetic distance between populations ranged 0.007 to 0.017 with low overall genetic difference FST = 0.011 and high gene flow Nm = 24.333, indicating that R. rita populations were not subdivided. This study revealed a high level of gene diversity with deficiency in genetic heterogeneity in all the populations of R. rita, emphasising natural management, conservation and rehabilitation measures of this species.

Sandalwood (Santalum album L.) is highly valued aromatic tropical tree. It is known for its high quality heartwood and oil. In this study 39 genic and genomic SSR markers were used to analyze the genetic diversity and population structure of 177 S. album accessions from 14 populations of three states in India. High genetic diversity was observed in terms of number of alleles 127 expected heterozygosity (He) ranged from 0.63-0.87 and the average PIC was 0.85. The selected population had relatively high genetic diversity with Shannons information index (I) >1.0. 0.02 mean coefficient of genetic differentiation (FST) and 10.55 gene flow were observed. AMOVA revealed that 92% of the variation observed within individuals. Based on cluster and Structure result individuals were not clustered as per their geographical origin. Furthermore the clusters were clearly distinguished by principal component analysis analysis and the result revealed that PC1 reflected the moderate contribution in genetic variation (6%) followed by PC2 (5.5%). From this study, high genetic diversity and genetic differentiation was found in S. album populations. The genetic diversity information of S. album populations can be used for selection of superior genotypes and germplasm conservation to promote the tree improvement of S. album populations.

The deliberate or accidental introduction of invasive alien species (IAS) causes negative ecological and economic impacts altering ecosystem processes, imperiling native species and causing damage to human endeavors. A monthly monitoring program was performed in Lake Trasimeno (Central Italy) from July 2018 to July 2019 in order to provide an upgrade of the population ecology of Procambarus clarkii and to assess the genetic diversity by analyzing the relationships among mitochondrial DNA diversity. Our results confirmed that P. clarkii is well acclimatized in the lake, revealing a stable population structure favored by the resources and conditions typical of this ecosystem, which seem to be optimal for the maintenance of the species. Four distinct mitochondrial haplotypes were detected, but one of them was clearly overrepresented (76%), suggesting that a single predominant introduction event may have occurred in this area, likely followed by secondary events. The identification of the typical genetic variants provides a better understanding of the evolutionary scenarios of P. clarkii in this biotope and it can be helpful in management plans concerning the expanding populations of this invasive alien species.
