ABSTRACT The interspecific oil palm hybrid BRS Manicoré (E. guineensis x E. oleifera) has superior agronomic characteristics. However, the germination rate is low (30%) and the process is slow when the seeds are sown in a conventional form. The purpose of this study was to optimize the in vitro germination of zygotic embryos of this hybrid comparing seed lots. The viability of zygotic embryos was evaluated by the tetrazolium test (0.075%) for 4 h. The embryos were cultured on MS and Y3 culture media, with and without the addition of NaH2PO4, as well as on MS, MS1/2 and N6 medium. In MS medium containing NaH2PO4, the germination rate was increased from 40 to 70% in comparison with the medium without sodium phosphate. The comparison between the culture media MS, MS 1/2, N6 and Y3 showed that 75% of zygotic embryos cultured in the Y3 medium formed whole plants (with roots and shoots defined), a higher percentage than embryos cultured on MS, MS 1/2 and N6 media (46, 35 and 17% respectively). In the same Y3 culture medium, the embryos were larger (36% ≥ 2 cm and 30% ≥ 5 cm) than in the other media. Results obtained by the tetrazolium test were similar to those of germination, showing the effect of the genotype of each seed lot. For the germination and development of plantlets it is essential to add NaH2PO4 to a culture medium containing no phosphate or with a low phosphate concentration.

Asymbiotic germination is considered an efficient and viable technique that can increase germination rates. The effect of type and concentration of disinfestants, and the exposure time to disinfestants may differ according to the plant species. Therefore, species-specific standardization of disinfestation agent and procedure is necessary to achieve optimal germination rates. The objective of this study was to determine a disinfestation methodology to increase in vitro germination rates and the early development of seedlings of Dendrobium nobile and Dendrobium phalaenopsis, using different times for seed disinfestation and different culture media. Seeds were disinfected by soaking in a 0.8% sodium hypochlorite solution for 5 or 15 min under aseptic conditions, after which seed suspensions were either washed with water or left unwashed. Next, they were seeded in culture flasks containing four different culture media (MS, ½MS, K, and VW). The flasks were then transferred to a growth room under controlled photoperiod and temperature, where they remained under an irradiance of 20 μmol m-2 s-1. Germination rates of the species were evaluated 45 days after placement in the culture flasks. A higher germination rate was observed when the seeds were triple washed, regardless of the culture medium or soaking time. Seed soaking disinfestation for 5 min is also recommended. MS and ½MS media were the most effective culture media in promoting in vitro germination of the species under study.

ABSTRACT The aim of this study was to evaluate the in vitro germination of zygotic embryos and seeds of Caesalpinia ferrea Martius and the morphogenetic responses of the explants to different concentrations of growth regulators. Seeds and zygotic embryos were inoculated in MS culture medium and kept in a growth room at a temperature of 25 ± 2 ºC for 16 hours of photoperiod for 30 days. The seeds had a higher in vitro germination rate than the explants from zygotic embryos. However, zygotic embryos in MS medium supplemented with 0.9 mg L-1 BAP had the highest percentage of regeneration (50%), number of shoots (3.25), buds (2.85) and leaves (3.15), multiplication rate (27.75), and length of shoots (1.96 cm). The in vitro culture of zygotic embryos and seeds made possible the multiplication of a higher number of healthy seedlings. Thus, it can be used as an alternative technique for the propagation of this species.

ABSTRACT: The objective of the present study was to evaluate the effects of different culture media on the in vitro germination and initial development of Cattleya nobilior, and to acclimatize this species using soybean straw as a substrate component, aiming the production of plants at large scale to reintroduce it in natural environments. Anatomical characteristics associated with development during acclimatization are also depicted. The influence of Murashige and Skoog, Knudson, and Vacin and Went culture media on the in vitro germination and protocorm development were assessed. Acclimatization was accomplished using different proportions of soybean straw (SS) and Bioplant (BP) as substrates. Anatomical studies were conducted on the leaves of plants during acclimatization. KC was the most suitable culture medium for both germination and initial development of C. nobilior. Regarding acclimatization, it is recommended that a substrate composed of 60% SR and 40% BP be initially used. Upon transfer to shade-house conditions, plants should be grown in a mix of 40% SR: and 60% BP. The leaf anatomical characteristics observed are typical of epiphytic orchids adapted to water-poor environments, which indicates that SR did not negatively affect the development of the species. The in vitro propagation of C. nobilior as described herein is efficient for its multiplication for commercial and conservation purposes and SS can be used as an alternative component of the substrate for its acclimatization. Calophyllum brasiliense is a tree species with limited natural reproduction. In vitro germination may be an alternative for obtaining high-quality seedlings. Seeds were maintained in water before surface disinfestation and compared with control seeds (i.e. not immersed), without differences between treatments. HgCl2 used during surface-disinfestation reduced contamination rates of cultures. Fungal contamination was reduced with fungicide added to culture medium (23 to 6.4%), although bacterial contamination increased (24 to 36%). In another experiment, seeds were immersed in plant preservative mixture (PPM™) prior to surface disinfestation. By combining immersion for 48 h and 2 mL L-1 in culture medium, contamination was only 6%. Seeds immersion in GA3 prior to surface disinfestation reduced root formation as concentration increased. Germination rate and GSI were reduced, respectively, from 72% and 0.129 (24 h) to 60% and 0.092 (48 h) according to exposure time to GA3. After 90 days in multiplication medium containing benzylaminopurine, average number of shoots per nodal segment was 3.4. In conclusion, in vitro germination of C. brasiliense seeds is feasible in sucrose-free WPM medium and reaches a high contamination-free rate (up to 93.3%).

The plant Tropaeolum majus, popularly known as nasturtium, presents food, ornamental and medicinal importance. Studies on floral and carpometric characteristics generate basic knowledge for areas of science such as botany, genetics and plant breeding. This work has the objectives of analyzing the effect of combinations of doses of poultry litter on floral structures and the in vitro germination of T. majus pollen grains. From the experimental matrix Plan Puebla III were defined 9 combinations of poultry litter doses incorporated and covered. The design of the experiments was entirely random, varying the replicates number. The number of stamens, petals, sepals and carpels was verified per floral bud in each treatment. The in vitro germination rate of pollen grains was estimated in 5 culture media and 2 germination times, from flower buds randomly collected in the experiment. The overall mean of stamens, sepals, petals and carpels per floral bud was 8.23; 5; 5 and 3, respectively. The combinations of poultry litter doses influenced only the number of stamens. The in vitro germination rate of T. majus pollen grains varied in the culture media. There was no association between the combinations of poultry litter doses and the in vitro germination rate of pollen grains.

The goal of this study was to expedite galax seed germination in vitro. Galax seeds were collected from Yancey County, N.C., at an elevation of about 1100 m. Aseptic cultures were established using the tiny rust-colored seeds. In vitro seed germination was achieved under different pH conditions (4.2, 5.0, and 5.8). Seeds cultured in the medium with pH 4.2 tended to germinate early with a better rate than those cultured with a higher pH of 5.0 or 5.8 at the very beginning. Gradually, seeds from media with pH 5.0 and 5.8 caught up in germination. Eventually, seeds from all pH treatments produced a very similar germination rate. Attempts to use the matted and scaly rhizomes and very tender new growth as explant materials to establish aseptic cultures were not successful, due to severe contamination. However, our observations suggested that the very tender new growth could be a good source of explants once the optimum sterilization time is established.

ABSTRACT: Couroupita guianensis Aubl. is an Amazonian forest species with important medicinal and ornamental value. This study evaluated the effect of different culture media and light spectra on the in vitro germination and development of the zygotic embryos of C. guianensis. The culture media, MS and WPM, were evaluated without the addition of plant growth regulators and were associated with four LED light spectra: white (CW), 70% red + 30% blue (R2B), 100% red (R), and 100% blue (B). One hundred percent of the seeds successfully underwent in vitro germination, and the culture media did not interfere with embryo development. In addition to this, the different light spectra induced in vitro morphogenesis and R2B treatment significantly promoted the production of secondary roots. This effect may aid in the rooting and acclimatization of seedlings of this species.

Slipper orchids belonging to the genus Phragmipedium (subfam. Cypripedioideae) are seriously threatened and therefore listed in Appendix I of CITES. Less research has been conducted in this genus than in others belonging to the same subfamily. In this work, we evaluated development of capsules (seed pods) from Phragmipedium humboldtii, P. longifolium and P. peaarcei from the time of pollination until opening. Moreover, seed viability was tested with the tetrazolium method in each of the capsules that were subsequently used to evaluate the effect of light and two culture media (Knudson C vs. Murashige and Skoog half concentrated) on in vitro asymbiotic germination and seedling growth. 100% of the pollinated flowers developed capsules, which differentiated in terms of length and diameter among species. While the length of the capsules remained constant during development, their diameter increased during the first 6-8 weeks and then stopped. Time required for maturity and opening of the capsules also varied among species (31 weeks in P. humboldtii, 16 weeks in P. longifolium and 9.5 weeks in P. percei). Seed viability differed among species as well, averaging 34.3% in P. humboldtii, 44.7% in P. longifolium and 82.3% in P. pearcei. Furthermore, seed viability of each capsule was used to adjust the germination rate measured in each case. While very few P. humboldtii viable seeds germinated under the conditions tested (2.9%), better results were observed in the other two species (close to 40% germination). No significant effect of light/darkness regime or of culture medium was observed on germination. However, better growth of the germinated embryos was observed with the Knudson C medium and darkness conditions. Further subculture of the gro- wing plantlets under light conditions induced development of roots and allowed successful acclimatization of seedlings in the greenhouse.

Fruits and almond from the dendezeiro, oil palmbelonging to the Elaeis genus,are widely used for the production of cookingoils or for the pharmaceutical and cosmetic industries.In the last decade, this oil palm also emerged as a promising source for commercialbiofuel production. This study evaluated the effect of different culture media, MS (MURASHIGUE AND SKOOG) and Y3 (EEUWENS)and carbohydrates duringin vitro germination of zygotic embryos, the effect of growth regulators GA3, NAA and BA Ponin vitro seedling development, and the survival rate of acclimatized seedlingsof Manicoré hybrid (Elaeis oleifera x E. guineensis). Zygotic embryos were inoculated on MS and modified Y3 media, supplemented with different sucrose concentrations (30, 45, and 60 gL-1) or sorbitol (36 gL-1), and the germination rate was evaluated after 30 days. Subsequently, seedlings were transferred to modified Y3 culture medium supplemented with differentGA3 concentrations (3.5 and 7 mgL-1) or without it, combined or not with 1 mgL-1 of NAA, 5 mgL-1 of BAP.The highest germinationpercentage of germinated embryos (92%) was observed in MS medium supplemented with 36 gL-1 sorbitol. Culture media supplemented with growth regulatorsGA3, NAA and BAP promoted greater shoot lengththan control media. Rooted seedlings showed high survival percentage (85%) during acclimatization.
