On-farm evaluation of yellow mosaic virus (YMV) resistant varieties of black gram for southern zone of Tamil Nadu

The yellow mosaic disease of blackgram caused by Mungbean yellow mosaic virus has emerged as a serious threat to pulses production especially in the South Eastern Asia.  Seed borne nature of MYMV in blackgram seeds was determined using the seeds harvested from a MYMV resistant (either VBN-6 or VBN-8) and susceptible blackgram (CO-5) varieties grown in three different agroclimatic zones of Tamil Nadu in India for three consecutive cropping seasons namely, Rabi 2018 (October- December), Summer 2019 (March-May) and Kharif 2019 (June- August) at three different time intervals viz., 20, 40 and 60 days after sowing (DAS). Seed borne nature of MYMV was observed only in the susceptible variety CO-5 and was absent in the   resistant varieties. Transmission of MYMV from infected plant to seeds was observed in all the three parts of the seeds viz., seed coat, cotyledon and embryo. Seeds from infected plants also showed abnormalities like shrinking, discolouration, ill filling inside pods and misshapen appearance.

In glasshouse experiments, Zucchini yellow mosaic virus (ZYMV) was transmitted from infected to healthy zucchini (Cucurbita pepo) plants by direct contact when leaves were rubbed against each other, crushed, or trampled, and, to a lesser extent, on ZYMV-contaminated blades. When sap from zucchini plants infected with three ZYMV isolates was kept at room temperature for up to 6 h, it infected healthy plants readily. Also, when sap from ZYMV-infected leaves was applied to seven surfaces (cotton, plastic, leather, metal, rubber vehicle tire, rubber-soled footwear, and human skin) and left for up to 48 h before the ZYMV-contaminated surface was rubbed onto healthy zucchini plants, ZYMV remained infective for 48 h on tire, 24 h on plastic and leather, and up to 6 h on cotton, metal, and footwear. On human skin, ZYMV remained infective for 5 min only. The effectiveness of 13 disinfectants at inactivating ZYMV was evaluated by adding them to sap from ZYMV-infected leaves which was then rubbed on to healthy zucchini plants. None of the plants became infected when nonfat dried milk (20%, wt/vol) or bleach (sodium hypochlorite at 42 g/liter, diluted 1:4) were used. When ZYMV-infected pumpkin leaves were trampled by footwear and then used to trample healthy plants, all plants became infected; however, when contaminated footwear was dipped in a footbath containing bleach (sodium hypochlorite at 42 g/liter, diluted 1:4) before trampling, none became infected. This study demonstrates that ZYMV can be transmitted by contact and highlights the need for on-farm hygiene practices (decontaminating tools, machinery, clothing, and so on) to be included in integrated disease management strategies for ZYMV in cucurbit crops.

The Blackgram culture VBG04-008 is a cross derivative of blackgram Vamban 3 x Vigna mungo var. silvestris 8 is released as TNAU blackgram VBN (Bg) 7 maturing in 65-70 days with an average height of 17 cm and suited for cultivation under both under rainfed and irrigated conditions. It has a yield potential of 981 Kg per hectare. This culture is resistant to Yellow Mosaic Virus, Powdery mildew and Leaf Curl Virus and less damage of pod borer. It possesses desirable characters like high protein content (21.05%), crude fibre (5.90g/100g) and iron (3.76 mg/100g). Grains are medium sized with black in colour. One hundred and twenty eight (128) on-farm demonstrations on Improved Production Technology (IPT) for black gram were carried out in eighty (80) hectares of farmer’s fields in Sankarankovil, Vasudevanallur and Kuruvikulam blocks of Tirunelveli district of Tamil Nadu from 2010-11 to 2012-13 under Tamil Nadu – Irrigated Agriculture Modernization and Water Bodies Restoration and Management (TN-IAMWARM) project. Two methods viz., Improved Production Technologies (IPT) and Conventional Method (CM) were compared. The results revealed that the adoption of Improved Production Technologies IPT) favorably influenced yield attributes and yield of black gram. The maximum seed yield (1,053 kg ha-1) obtained from IPT which was higher than conventional method (720 kg ha-1). The best net income ( 25,650) and benefit:cost (2.60) were also associated with IPT than conventional method of black gram cultivation. The additional income of 11,762 ha-1 was obtained from IPT over Conventional Method of black gram cultivation.

A total of nine black gram genotypes were tested for yellow mosaic virus (YMV) resistance along with other nine biometrical traits. The genotypes VBN 4, KKB-14-015, KKB-14-45 and KKB-14-022 exhibited complete resistance to YMV in field screening. From the molecular characterization, out of 42 SSR markers studied, 15 SSR markers expressed polymorphism. The PIC values ranged from 0.37 (for SSR marker CEDG 024) to 0.79 (CEDG 154) with an average of 0.63. From the cluster analysis, YMV resistant genotypes KKB-14-045 and KKB-14-015 were located on clusters 6 and 7 and the susceptible but agronomic desirable genotypes IC 343943 and IC 436656 were located on cluster 3 which showed the diverse nature of these genotypes. Hence combinations viz., IC 343943 x KKB-14-015, IC 343943 x KKB-14-045, IC 436656 x KKB-14-015 and IC 436656 x KKB-14-045 would be ideal to produce desirable recombinants for YMV resistance breeding in black gram.
