Brassica napus L. is a major oilseed crop all over the world. The aim of this study was to investigate the genetic diversity of B. napus germplasm by using simple sequence repeats (SSR) markers. In the current study, ten SSR markers were used for studying genetic diversity of ten Brassica cultivars. Out of 110 total bands, 68 bands were polymorphic with 52.11% average polymorphism. Mean value of Nei’s genetic diversity and Polymorphism Information Content was 1.7, and 0.2630, respectively. These mean values show that there are moderate allelic differences between Brassica cultivars. The Nei’s genetic distance among various cultivars was 0.3281 and 0.125 which showed that germplasm of Brassica cultivars are different from each other, which is probably due to anthropogenic interventions and environmental factors. Thus, genetically different lines identified in this study could be employed in breeding programmes to develop higher-quality canola inbred varieties in future.

The genetic diversity of 94 accessions of winter oilseed rape (Brassica napus L.), representing past and contemporary material utilized in the Czech breeding programmes, was determined using microsatellites (SSRs), ISSRs and AFLPs. All three kinds of markers differed in the range of observed polymorphism and differentiated clearly each accession. Altogether 89 SSR, 1003 AFLP and 53 ISSR markers were evaluated. Their average rates of polymorphic bands were 100%, 53.9% and 90.6%, respectively, indicating high genetic diversity among the accessions. The greatest genetic distance was found by ISSRs (62.3%) whereas significantly lower distances of 49.4% in SSRs and 35.5% in AFLPs were observed. The genetic similarity matrix clearly distinguished all accessions. A set of the most distinct varieties was established. The analysis of the genetic pattern of the accessions indicated two groups comprising most of the modern Czech breeding materials, revealing a distinct shift in breeding. Surprisingly, molecular analyses did not support  breeders’ views about a narrow genetic base of the Czech breeding materials. The choice of appropriate technology for different aspects of germplasm evaluation is also discussed.

The Sorghum genus is extremely diverse both morphologically and geographically, however, relatively few of the 25 recognized species have been evaluated genetically. The apparent lack of basic knowledge pertaining to the levels of genetic diversity both within and between the 17 Australian wild species is a major obstacle to both their effective conservation and potential use in breeding programmes. Twelve Sorghum bicolor-derived simple sequence repeat (SSR) markers were evaluated for cross-species amplification in all 25 Sorghum species. The SSR markers were highly polymorphic, with diversity indices ranging from 0.59 to 0.99 with mean of 0.91. Five markers combined were able to differentiate 24 of the 25 Sorghum species, with intra-species polymorphism apparent. Sorghum bicolor-derived SSRs have proven to be an efficient source of markers for genetic diversity studies of the relatively poorly characterized Australian indigenous Sorghum species.

Early maturity is a major objective of oilseed rape (Brassica napus L.) breeding programs in western Canada. Maturity of crops is influenced by time of initiation and flowering. The presence of a vernalization requirement affects plant development by delaying floral initiation until the cold requirement of the plant has been satisfied. Five spring oilseed rape cultivars were screened for their response to vernalization. Vernalization treatments consisted of exposure of germinated seeds to 0–42 d at 4 °C. Plants were assessed under a 20-h photoperiod. In general, there was a cumulative response to vernalization, with a decrease in days to each developmental stage as exposure to 4 °C was increased. Vernalization treatment of 6 d at 4 °C was sufficient to decrease both the days to first flower and the final leaf number. The characterization of vernalization response is of interest because variation in flowering time in response to year-to-year variations in the environment could result. Genetic diversity is the source of novel allelic combinations that can be efficiently utilized in any crop improvement program. To facilitate future crop improvement programs in rice, a study was designed to identify the underlying genetic variations in the Sali rice germplasms of Assam using SSR markers. The 129 SSR markers that were used in the study amplified a total of 765 fragments with an average of 5.93 alleles per locus. The Shannon's Information Index was found to be in the range from 0.533 to 1.786. The Polymorphism Information Content (PIC) fell into the range from 0.304 to 0.691 with a mean value of 0.55. The overall FST value was found to be 0.519 that indicated the presence of genetic differentiation amongst the genotypes used in the study. The Sali population was divided into two clusters. The information obtained from the present study will facilitate the genetic improvement of Sali rice cultivars.

Abstract Background The genetic base of soybean cultivars in India has been reported to be extremely narrow, due to repeated use of few selected and elite genotypes as parents in the breeding programmes. This ultimately led to the reduction of genetic variability among existing soybean cultivars and stagnation in crop yield. Thus in order to enhance production and productivity of soybean, broadening of genetic base and exploring untapped valuable genetic diversity has become quite indispensable. This could be successfully accomplished through molecular characterization of soybean genotypes using various DNA based markers. Hence, an attempt was made to study the molecular divergence and relatedness among 29 genotypes of soybean using SSR markers. Methods and results A total of 35 SSR primers were deployed to study the genetic divergence among 29 genotypes of soybean. Among them, 14 primer pairs were found to be polymorphic producing a total of 34 polymorphic alleles; and the allele number for each locus ranged from two to four with an average of 2.43 alleles per primer pair. Polymorphic information content (PIC) values of SSRs ranged from 0.064 to 0.689 with an average of 0.331. The dendrogram constructed based on dissimilarity indices clustered the 29 genotypes into two major groups and four sub-groups. Similarly, principal coordinate analysis grouped the genotypes into four major groups that exactly corresponded to the clustering of genotypes among four sub-groups of dendrogram. Besides, the study has reported eight unique and two rare alleles that could be potentially utilized for genetic purity analysis and cultivar identification in soybean. Conclusion In the present investigation, two major clusters were reported and grouping of large number of genotypes in each cluster indicated high degree of genetic resemblance and narrow genetic base among the genotypes used in the study. With respect to the primers used in the study, the values of PIC and other related parameters revealed that the selected SSR markers are moderately informative and could be potentially utilized for diversity analysis of soybean. The clustering pattern of dendrogram constructed based on SSR loci profile displayed good agreement with the cultivar’s pedigree information. High level of genetic similarity observed among the genotypes from the present study necessitates the inclusion of wild relatives, land races and traditional cultivars in future soybean breeding programmes to widen the crop gene pool. Thus, hybridization among diverse gene pool could result in more heterotic combinations ultimately enhancing genetic gain, crop yield and resistance to various stress factors.

The effects of four sulphur levels: S0, S1, S2 and S3, including 0, 12, 24 and 36 kg S ha-1, respectively, along with 115 kg N ha-1 were studied on yield-related traits of oilseed rape (Brassica napus L.). The significant variance of treatments was determined for plant height, yield component characters, seed yield and oil content. The sulphur application significantly increased most of the traits compared to the S0 level. The S3 (36 kg S ha-1) treatment led to the highest mean value of plant height (132 cm) which was classified with S2 (24 kg S ha-1) in the same statistical group. Sulphur had an increasing effect on pods per plant, and it ranged from 92 to 196 for S0 and S3 applications, respectively. S0 and S1 with 92 and 121 pods per plant were grouped in the same statistical group. In addition, S2, and S3 with 165 and 196 pods per plant showed no significant statistical difference. The sulphur application significantly increased seed yield compared to control (S0 level), and it ranged from 2744 to 3215 kg ha-1 in S0 and S3, respectively. The average oil contents of 45.69, 46.96, 47.46 and 49.53 % were detected for 0, 12, 24 and 36 kg S ha-1, respectively.
